Discovery of penicillic acid from mangrove endophytic fungi: anti-Vibrio screening and cell envelope disruption in V. parahaemolyticus.

Mangrove endophytic fungi are recognized as prolific sources of structurally diverse and bioactive natural products. In this study, 19 cultivable endophytic fungal strains were isolated and taxonomically characterized from mangrove plant tissues collected at the Shankou Mangrove Nature Reserve (Guangxi, China). One isolate, Aspergillus sclerotiorum BM19, exhibited broad-spectrum inhibitory activity against multiple aquatic pathogenic Vibrio strains. Bioactivity-guided fractionation of its ethyl acetate crude extract led to the isolation of penicillic acid (1), whose structure was fully elucida

Neuropeptide Y drives vitiligo progression via NPYR-mediated NF-κB activation: a novel therapeutic target.

Neuropeptide Y (NPY) is a neurotransmitter involved in stress response and immune modulation and its dysregulation may contribute to the pathogenesis of vitiligo by influencing melanocyte function and autoimmune responses. This study aimed to explore the role and molecular mechanism of NPY in the progression of vitiligo. In vitro, human immortalized keratinocyte (HaCaT) cells were treated with H2O2 to simulate oxidative stress conditions observed in vitiligo. Through specific knockdown of the NPY receptor (NPY2R or NPY4R) achieved by siRNA transfection, combined with techniques such as Western

Dexamethasone Attenuates Oncostatin M Production via Suppressing of PI3K/Akt/NF-κB Signaling in Neutrophil-like Differentiated HL-60 Cells.

Oncostatin M (OSM) plays a role in various inflammatory reactions, and neutrophils are the main source of OSM in pulmonary diseases. However, there is no evidence showing the mechanism of OSM production in neutrophils. While dexamethasone (Dex) has been known to exert anti-inflammatory activity in various fields, the precise mechanisms of OSM downregulation by Dex in neutrophils remain to be determined. Here, we examined how OSM is produced in neutrophil-like differentiated HL-60 cells. Enzyme-linked immunosorbent assay, real-time polymerase chain reaction, and Western blot analysis were utili

Sustainable Primary Cell Banking for Topical Compound Cytotoxicity Assays: Protocol Validation on Novel Biocides and Antifungals for Optimized Burn Wound Care.

Thorough biological safety testing of topical therapeutic compounds and antimicrobials is a critical prerequisite for appropriate cutaneous wound care. Increasing pathogen resistance rates to traditional antibiotics and antifungals are driving the development and registration of novel chemical entities. Although they are notably useful for animal testing reduction, the gold standard in vitro cytotoxicity assays in continuous cell lines (HaCaT keratinocytes, 3T3 fibroblasts) may be discussed from a translational relevance standpoint. The aim of this study was thus to establish and validate a su

Photoprotective properties of new derivatives of kinetin.

The chronic exposure of skin to ultraviolet (UV) radiation causes adverse dermal reactions, such as erythema, sunburn, photoaging, and cancer, by altering several signalling pathways associated with oxidative stress, inflammation, and DNA damage. One of the possible UV light protection strategies is the use of dermal photoprotective preparations. The plant hormone kinetin (N6-furfuryladenine; KIN) exhibits antioxidant and anti-senescent effects in human cells. Topically applied KIN also reduced some of the clinical signs of photodamaged skin. To improve the biological activities of KIN, severa

Interleukin-37 overexpression enhances the therapeutic efficacy of adipose-derived mesenchymal stem cells in atopic dermatitis by activating autophagy and improving skin barrier dysfunction.

Adipose-derived mesenchymal stem cells (ADSCs) have emerged as a promising therapeutic option because of their immunomodulatory and tissue-repair capabilities in the treatment of Atopic dermatitis (AD). However, their efficacy is limited by cellular senescence and functional decline. Interleukin-37 (IL-37), an anti-inflammatory cytokine, has shown potential in mitigating inflammation and enhancing skin barrier function. This study investigated whether IL-37 overexpression in ADSCs could augment their therapeutic effects in AD and the underlying mechanism. Lentiviral-mediated IL-37 overexpressi

Enhanced Clearing of Wound-Related Pathogenic Bacterial Biofilms Using Protease-Functionalized Antibiotic Nanocarriers.

Biofilms are prevalent in chronic wounds and once formed are very hard to remove, which is associated with poor outcomes and high mortality rates. Biofilms are comprised of surface-attached bacteria embedded in an extracellular polymeric substance (EPS) matrix, which confers increased antibiotic resistance and host immune evasion. Therefore, disruption of this matrix is essential to tackle the biofilm-embedded bacteria. Here, we propose a novel nanotechnology to do this, based on protease-functionalized nanogel carriers of antibiotics. Such active antibiotic nanocarriers, surface coated with t

In vitro micro-physiological immune-competent model of the human skin.

Skin allergy, in particular, allergic contact dermatitis and irritant contact dermatitis, are common occupational and environmental health problems affecting the quality of life of a significant proportion of the world population. Since all new ingredients to be incorporated into a product are potential skin allergens, it is essential that these ingredients be first tested for their allergenic potential. However, despite the considerable effort using animal models to understand the underlying mechanism of skin sensitization, to date, the molecular and cellular responses due to skin contact wit
