Comparative analysis of skin transcriptome reveals differences of cashmere fineness in different body parts of Inner Mongolia cashmere goats.

OBJECTIVE: The growth and development of secondary hair follicles primarily determine the economic value of cashmere traits, significantly influencing the quality of cashmere fineness. Previous studies have concentrated on the periodic growth regulation of hair follicles in Inner Mongolia cashmere goats (IMCGs), identifying numerous candidate genes that influence cashmere traits. Research on the factors and regulatory mechanisms affecting cashmere fineness in different body parts is currently limited. METHODS: The differences of cashmere fineness traits among different body parts or ages were

Whole-Mount Dissection of the Vestibular Complex and Quantification of Vestibular Ganglion Neurons in Mice.

The vestibular system is crucial for our sense of balance, for detecting head movements, and for regulating gaze and posture. There are five vestibular organs located inside the temporal bone, including two otolith organs (utricle and saccule), which detect linear movement, and three cristae (lateral/horizontal, anterior, and posterior), which detect rotational movement. Each organ contains a sensory epithelium with hair cells that detect head movements and supporting cells that provide structural support, release growth factors and cytokines, and phagocytose dying hair cells. The neurons that

Exosomal miR-199a-3p From Dermal Papilla Cells Regulates Hair Follicle Pigmentation by Targeting POU2F1 in Melanocytes.

To elucidate the paracrine regulatory mechanism of dermal papilla cells (DPCs) on melanocyte (MC) melanogenesis, we focused on exosomal miR-199a-3p as a key mediator. DPCs, MCs, and DPC-Exos were identified by immunofluorescence, Western blot (WB), nanoparticle tracking analysis (NTA), and transmission electron microscopy (TEM). Functional assays showed DPC-Exos were internalized by MCs, enhanced proliferation, decreased apoptosis, and elevated melanin production. miRNA sequencing identified miR-199a-3p as the key exosomal cargo, which was localized to hair follicle bulbs by fluorescence in si

Mesenchymal stem cell exosomes enhance the development of hair follicle to ameliorate androgenetic alopecia.

BACKGROUND: Mesenchymal stem cells (MSCs) and their secretome have significant potential in promoting hair follicle development. However, the effects of MSC therapy have been reported to vary due to their heterogeneous characteristics. Different sources of MSCs or culture systems may cause heterogeneity of exosomes. AIM: To define the potential of human adipose-derived MSC exosomes (hADSC-Exos) and human umbilical cord-derived MSC exosomes (hUCMSC-Exos) for improving dermal papillary cell proliferation in androgenetic alopecia. METHODS: We conducted liquid chromatography-mass spectrometry pr

Analysing the dynamics of a model for alopecia areata as an autoimmune disorder of hair follicle cycling.

Alopecia areata (AA) is a CD8$^{+}$ T cell-dependent autoimmune disease that disrupts the constantly repeating cyclic transformations of hair follicles (HFs). Among the three main HF cycle stages-growth (anagen), regression (catagen) and relative quiescence (telogen)-only anagen HFs are attacked and thereby forced to prematurely enter into catagen, thus shortening active hair growth substantially. After having previously modelled the dynamics of immune system components critically involved in the disease development (Dobreva et al., 2015), we here present a mathematical model for AA which inco

NEO-FFI personality clusters in trichotillomania.

The purpose of this study was to determine whether personality prototypes exist among hair pullers and if these groups differ in hair pulling (HP) characteristics, clinical correlates, and quality of life. 164 adult hair pullers completed the NEO-Five Factor Inventory (NEO-FFI; Costa and McCrae, 1992) and self-report measures of HP severity, HP style, affective state, and quality of life. A latent class cluster analysis using NEO-FFI scores was performed to separate participants into clusters. Bonferroni-corrected t-tests were used to compare clusters on HP, affective, and quality of life vari

Fate mapping of Trps1 daughter cells during cardiac development using novel Trps1-Cre mice.

Tricho-rhino-phalangeal syndrome (TRPS) is a rare congenital disorder that is characterized by abnormal hair growth and skeletal deformities. These result in sparse hair, short stature, and early onset of joint problems. Recent reports have shown that a relatively high proportion of patients with TRPS exhibit a broad range of congenital heart defects. To determine the regulation of Trps1 transcription in vivo, we generated novel transgenic mice, which expressed Cre recombinase under the murine Trps1 proximal promoter sequence (Trps1-Cre). We crossed these mice with Cre reporter mice to identif

The usefulness of biomarkers of alcohol abuse in hair and serum carbohydrate-deficient transferrin: a case report.

The detection of carbohydrate-deficient transferrin (CDT) in serum is widely accepted to identify chronic alcohol consumption over the previous two weeks, but minor ethanol metabolites detected in hair often complete the information obtained. In particular, ethylglucuronide and cocaethylene (a marker of simultaneous intake of cocaine and alcohol) allow correct interpretation of data obtained in forensic cases. We refer to a negative CDT value obtained from a serum sample collected during hospitalization of a man admitted for cardiac arrest who died about 14 h later. Clinical analysis performed

Polymorphisms upstream of the melanocortin-1 receptor coding region are associated with human pigmentation variation in a Brazilian population.

OBJECTIVE: We describe an association of two SNPs, rs3212345:C>T and rs3212346:G>A, located approximately 2.5 kb upstream of the melanocortin-1 receptor (MC1R) translation initiation codon, with pigmentation phenotype variation in a Southeast Brazilian miscegenated population. METHODS: One hundred thirty-eight genetically unrelated subjects, with multicolor phenotype, were selected from the southeast region of Brazil. Skin, hair and eye color, and tanning ability were rated. Genotypes for each SNP (rs3212345:C>T and rs3212346:G>A) were determined. A logistic regression analysis was performed

Heat treatment increases the incidence of alopecia areata in the C3H/HeJ mouse model.

Alopecia areata (AA) is a common autoimmune disease characterized by non-scarring hair loss. Previous studies have demonstrated an association between AA and physiological/psychological stress. In this study, we investigated the effects of heat treatment, a physiological stress, on AA development in C3H/HeJ mice. Whereas this strain of mice are predisposed to AA at low incidence by 18 months of age, we observed a significant increase in the incidence of hair loss in heat-treated 8-month-old C3H/HeJ mice compared with sham-treated mice. Histological analysis detected mononuclear cell infiltrati

[Cutaneous signs of Noonan's syndrome. Apropos of a case with ulerythema ophyogenes, disseminated pilar and sudoral keratosis and progressive alopecia].

A case of typical Noonan syndrome (NS) with eye abnormalities, pterygium colli, cryptorchid testes, lymphoedema and asymmetrical cardiac sep … been reported, but our patient exhibited progressive alopecia which is very rare in NS. Biochemical hair analysis did not show any abnormalities of aminoacids. All these features were suggestive of keratosis follicularis spinulosa decalvans. It therefore seems very likely that NS is associated with keratinization disorders but ulerythema ophryogenes might only …

MitoQ enhances CYP19A1 expression to stimulate WNT/β-catenin signaling pathway for promoting hair growth in androgenetic alopecia.

Increased sensitivity to androgens and androgen receptors is the underlying cause of androgenetic alopecia (AGA), a hereditary disease. Our study investigated the preventive effects of MitoQ on dihydrotestosterone (DHT)-induced mitochondrial dysfunction and subsequent hair loss from three perspectives: in vivo, in vitro, and network pharmacology. A mouse model of AGA was used to assess the effectiveness of MitoQ intervention. Seventy-five drug targets and 367 disease targets were identified through network pharmacology analysis. Molecular docking analysis revealed that the androgen receptor (A

The psychosocial burden of androgenetic alopecia in Saudi Arabia: A cross-sectional study.

BACKGROUND: The most prevalent cause of hair loss is androgenetic alopecia. Improved therapeutic modalities may result from knowing how stress affects hair growth, incorporating stress-coping methods into the treatment of hair loss disorders, and developing new pharmacotherapeutic approaches. This study aimed to examine the psychosocial burden of androgenetic alopecia in Saudi Arabia. MATERIALS AND METHODS: A cross-sectional online survey study was conducted among patients with androgenetic alopecia in Saudi Arabia in February 2023. A previously developed questionnaire was adapted from a prev

Molecular detection and species identification of dermatophytes by SYBR-Green real-time PCR in-house methodology using hair samples obtained from dogs and cats.

The classical dermatophytes diagnosis is based on mycological culture and microscopy observation both human and animal hair, skin, and nail samples. The aim of this work was to develop the new in-house real-time PCR with pan-dematophyte reaction for detection and identification of the main dermatophytes directly from hair samples, providing a simple and rapid diagnosis of dermatophytosis in dogs and cats. An in-house SYBR-Green real-time PCR was designed and used for detecting a DNA fragment encoding chitin synthase 1 (CHS1). A total of 287 samples were processed by culture, microscopic examin

High dose craniospinal irradiation as independent risk factor of permanent alopecia in childhood medulloblastoma survivors: cohort study and literature review.

PURPOSE: Our aim was to determine the main risk factors related to the occurrence of permanent alopecia in childhood medulloblastoma (MB) survivors. METHODS: We retrospectively analyzed the clinical features of all consecutive MB survivors treated at our institute. We divided the patients into 3 groups depending on the craniospinal irradiation (CSI) dose received and defined permanent alopecia first in terms of the skin region affected (whole scalp and nape region), then on the basis of the toxicity degree (G). Any relationship between permanent alopecia and other characteristics was investig

Primary cicatricial alopecia: recent advances in understanding and management.

Primary cicatricial alopecias (PCA) are a rare group of disorders, in which the hair follicle is the main target of destructive inflammation resulting in irreversible hair loss with scarring of affected lesions. The most typical clinical manifestation of PCA is the loss of visible follicular ostia. The histopathological hallmark of a fully developed lesion is the replacement of the hair follicle structure by fibrous tissue. PCA could share similar clinical manifestations and eventually lead to "burn-out" alopecia. Some subsets are hardly distinguishable histopathologically and the mechanisms t

Gene for arrhythmogenic right ventricular cardiomyopathy with diffuse nonepidermolytic palmoplantar keratoderma and woolly hair (Naxos disease) maps to 17q21.

BACKGROUND: Arrhythmogenic right ventricular cardiomyopathy (ARVC) is a heart muscle disease of unknown etiology that causes arrhythmias, heart failure, and sudden death. Diagnosis can be difficult, and this hampers investigation of its molecular basis. Forms of ARVC in which gene penetrance and disease expression are greater should facilitate genetic study. We undertook a clinical and genetic investigation of Naxos disease, originally described by Protonotarios in 1986. This disease constitutes the triad of ARVC, diffuse nonepidermolytic palmoplantar keratoderma, and woolly hair. METHODS AND

The assessment of Hair Transplantation Outcomes in Cicatricial Alopecia.

BACKGROUND: Cicatricial alopecia (CA) is scarring alopecia due to a diverse group of disorders which destroy the hair follicle, replace it with scar tissue causing permanent hair loss. Spontaneous regrowth of hair in CA is rarely seen and thus it has a negative impact on the self-esteem of the patient. However, hair transplantation (HT) has emerged as an effective modality of treatment to replace lost hair for cosmetic benefit. MATERIALS AND METHODS: This was a prospective analysis of patients affected with CA, who underwent HT from January 2015 to December 2017. Demography of patients along

[Evoked acoustic oto-emissions: effects of atropine].

The Evoked Otoacoustic Emissions (EOE) are a sinusoidal wave complex coming from the contractile properties of the cochlea and particularly of the outer hair cells. This activity is influenced by the centrifugal pathways whose transmitter is acetylcholine. In order to study the effects of the functional block of the cochlea, ten young normal subjects were examined recording the EOE before and ten mins after the i.v. infusion of 1 mg of atropine. The statistical analysis of the results has shown a significant decrease only in EOE appearance threshold in absence of modifications of the saturatio

Immunohistochemical analysis of T-cell subsets in the peribulbar and intrabulbar infiltrates of alopecia areata.

In 11 patients with untreated alopecia areata in the progressive stage of the disease, an in situ analysis of the inflammatory infiltrate of the hair bulbs was performed by means of different monoclonal antibodies. Most of the peribulbar cells reacted with the pan T-cell antibodies OKT 3 and Lyt 3. Staining for T-cell subsets revealed that the proportion of OKT 4+ cells was about fourfold higher than that of OKT 8+ cells. Almost all of the T cells were OKIa1+, indicating that they were in an activated state. In four of the 11 cases, both subsets of T lymphocytes were also found to infiltrate t
