CMR gives clue to "ragged red fibers" in the heart in a patient with mitochondrial myopathy.

Mitochondrial myopathy may manifest either as isolated myopathy or as a neuromuscular multisystemic disease and is caused by genetic defects in the mitochondrial genome resulting in respiratory chain disorders. MELAS, which is characterised by mitochondrial myopathy, encephalopathy, lactic acidosis and stroke-like episodes due to gene mutations in the mitochondrial DNA (adenine-to-guanine transition at nucleotide pair 3243, m.3243A>G), constitutes such a mitochondrial multisystemic disease. Although hypertrophied or dilated cardiomyopathy is quite common in MELAS, there have been no cardiovasc

Endogenous hSNM1B/Apollo interacts with TRF2 and stimulates ATM in response to ionizing radiation.

Human SNM1B/Apollo is involved in the cellular response to DNA-damage, however, its precise role is unknown. Recent reports have implicated hSNM1B in the protection of telomeres. We have found hSNM1B to interact with TRF2, a protein which functions in telomere protection and in an early response to ionizing radiation. Here we show that endogenous hSNM1B forms foci which colocalize at telomeres with TRF1 and TRF2. However, we observed that additional hSNM1B foci could be induced upon exposure to ionizing radiation (IR). In live-cell-imaging experiments, hSNM1B localized to photo-induced double-

Cortical dysplasia of the left temporal lobe might explain severe expressive-language delay in patients with duplication of the Williams-Beuren locus.

We report on a new duplication case of 7q11.23, reciprocal of the Williams-Beuren (WB) deletion. The patient, a 13-year-old girl, was ascertained within an array-CGH screening of patients with epilepsy and neuronal migration defects. Similarly to the first reported patient, she showed serious difficulties in expressive language in the absence of severe mental retardation and marked dysmorphic features. Magnetic resonance imaging (MRI) of the brain revealed an abnormal development of the cerebral cortex in the left temporal lobe, which showed a simplified gyral pattern, and increased cortical t

Small molecule-based reversible reprogramming of cellular lifespan.

Источник пометил публикацию как отозванную

Most somatic cells encounter an inevitable destiny, senescence. Little progress has been made in identifying small molecules that extend the finite lifespan of normal human cells. Here we show that the intrinsic 'senescence clock' can be reset in a reversible manner by selective modulation of the ataxia telangiectasia-mutated (ATM) protein and ATM- and Rad3-related (ATR) protein with a small molecule, CGK733. This compound was identified by a high-throughput phenotypic screen with automated imaging. Employing a magnetic nanoprobe technology, magnetism-based interaction capture (MAGIC), we iden

Comparative proteome analysis of laboratory grown Brucella abortus 2308 and Brucella melitensis 16M.

Brucella species are pathogenic agents that cause brucellosis, a debilitating zoonotic disease that affects a large variety of domesticated animals and humans. Brucella melitensis and Brucella abortus are considered major health threats because of their highly infectious nature and worldwide occurrence. The availability of the annotated genomes for these two species has allowed a comparative proteomics study of laboratory grown B. melitensis 16M and B. abortus 2308 by two-dimensional (2-D) gel electrophoresis and peptide mass fingerprinting. Computer-assisted analysis of the different 2-D gel

A preliminary assessment of the effects of summer transport on behavioral and physiological stress biomarkers in cull sows selected for heat stress tolerance or sensitivity in the F1 generation.

Cull sow transport in hot summer months can cause morbidity and mortality due to heat stress (HS), and there is a need to reduce these risks. Therefore, the study objective was to evaluate whether genomic selection for HS tolerance (TOL) or sensitivity (SEN) in the F1 generation would alter behavioral and physiological indicators of HS in cull sows transported during the summer. We hypothesized that TOL sows would have a reduction in behavioral and physiological stress responses relative to SEN sows during summer transport. One day post-weaning, 14 TOL and 14 SEN primiparous sows (Large White

In vivo adenine base editing of mutant Galc gene ameliorates Krabbe disease progression.

BACKGROUND: Krabbe disease (KD) is caused by mutation of the galactosylceramidase (GALC) gene, leading to deficient sphingolipid metabolism, which is essential for functional myelination. The twitcher (Galctwi/twi) mouse, a KD model with a premature termination codon (PTC) caused by a single-nucleotide G-to-A substitution at the 355th codon of the Galc gene, is a model candidate for treatment with adenine base editors (ABEs). ABEs have emerged exclusively among genome editing systems as viable therapeutic candidates to correct mutant genes. METHODS: To confirm base editing efficiency of ABEs,

Genetics of digital phenotypes of keel bone in layer chickens and correlations with keel bone fractures and deviations.

BACKGROUND: Poultry is a global industry with laying hens that are genetically optimized for high egg yield. Keel bone fractures can affect up to 80% of laying hens, posing welfare and production problems. Therefore, genetic selection to reduce keel fractures is important. However, the lack of a reliable, automated, and heritable phenotypes for keel bones makes this a challenging task. The aim of this study was to (1) develop automated analyses of radiographic images to phenotype keel bones, and (2) investigate whether the proposed phenotypes are heritable and genetically correlated with the p

Imaging in children with ataxia-telangiectasia-The radiologist's approach.

Ataxia-telangiectasia (A-T) is a syndromic inborn error of immunity (IEI) characterized by genomic instability, defective reparation of the DNA double-strand breaks, and hypersensitivity to ionizing radiation disturbing cellular homeostasis. The role of imaging diagnostics and the conscious choice of safe and advantageous imaging technique, as well as its correct interpretation, are crucial in the diagnostic process and monitoring of children with A-T. This study aimed at defining the role of a radiologist in the early diagnosis of A-T, as well as in detecting and tracking disease complication

Resistance to Thyroid Hormone Beta or Thyroid-Stimulating Hormone (TSH)-Secreting Pituitary Adenoma: A Diagnostic Dilemma With a THRB Variant of Uncertain Significance.

Elevated thyroid hormone concentrations with nonsuppressed thyroid-stimulating hormone (TSH) present a challenging diagnostic problem. Resistance to thyroid hormone beta (RTHβ), analytical interference, and a TSH-secreting pituitary adenoma (TSHoma) are important considerations. We describe a woman with persistent elevation of free thyroxine (FT4) and nonsuppressed or elevated TSH for more than five years, autoimmune thyroiditis, a heterozygous THRB c.284G>C (p.Gly95Ala) variant classified as a variant of uncertain significance (VUS), and a 4.9 × 4.4 mm pituitary microadenoma. Cohesin prevents local mixing of condensed euchromatic domains in living human cells.

The human genome is folded into chromatin loops by the cohesin complex, forming functional chromatin domains that underlie transcription and DNA replication/repair. However, how cohesin organizes these domains in living cells, especially in active euchromatin, remains elusive. Here, to address this question, we combined single-nucleosome imaging/tracking and super-resolution three-dimensional structured illumination microscopy with euchromatin-specific labeling of histone variant H3.3. Using this nanoscopic approach, we revealed that euchromatin forms condensed domains that are constrained by

Malignant vs. Non-malignant Annotations on TCGA Breast Cancer Whole Slide Images for AI Analysis.

Identification of malignant and non-malignant regions in breast cancer whole slide images (WSIs) is essential for understanding tumor heterogeneity and histopathological evaluation. Annotated data allows deep learning models to learn features and analyze histological structures. In this study, we obtained 50 breast cancer WSIs from The Cancer Genome Atlas (TCGA) and had an expert pathologist manually annotate malignant (n = 1,882) and non-malignant (n = 374) regions using QuPath. The annotated regions were independently reviewed by a second pathologist, achieving an inter-observer agreement of

Condensation of MATCAP promotes CENP-E-dependent chromosome congression in mitosis.

Accurate segregation of mitotic chromosomes requires pre-anaphase alignment driven by microtubule-based motor proteins. Tubulin detyrosination is essential to guide CENP-E-driven chromosome congression in mitosis. However, the mechanisms of action and physiochemical properties of the detyrosinases for decoding CENP-E motility remain elusive. Here we show that microtubule-associated tyrosine carboxypeptidase (MATCAP) undergoes intrinsically disordered region (IDR)-dependent liquid-liquid phase separation (LLPS) on microtubules to constitute the tubulin detyrosination machinery. These biomolecul

The trypanosome flagellum as model for parasitology, cell biology and ciliopathies.

Cilia and flagella exhibit widely conserved structures and functions across species. In humans, defects in these organelles are responsible for diseases called ciliopathies and many model organisms are used to study them. In this review, we will discuss one of them, the parasite Trypanosoma brucei, which is particularly well-suited to investigate general aspects of cilia and flagella, such as construction or protein localisation. Its flagellum remains present throughout the cell cycle, offering the opportunity to monitor flagellum maintenance and assembly within the same cell. This model organ
