BACKGROUND: The treatment of patients with Hodgkin lymphoma (HL) who develop disease progression after undergoing allogeneic stem cell transplantation (allo-SCT) remains challenging.
METHODS: The authors assessed outcomes in 184 adult patients with HL who developed disease recurrence or progression after a matched related or unrelated allo-SCT at European Society for Blood and Marrow Transplantation-participating centers between 2010 and 2014.
RESULTS: Eighty patients who received brentuximab vedotin (BV) salvage therapy were compared with 104 patients who did not. Patients in the BV group w

Anti-CD30 therapy for Hodgkin lymphoma led to transient loss of detectable CD4+ T-cell HIV RNA and a decrease in residual plasma viremia. Targeting nonviral markers expressed on HIV-1 transcriptionally active cells may lead to reduced measures of HIV-1 persistence.

The hallmark of classical Hodgkin lymphoma (cHL) is the presence of giant, mostly multinucleated Hodgkin-Reed-Sternberg (HRS) cells. Whereas it has recently been shown that giant HRS cells evolve from small Hodgkin cells by incomplete cytokinesis and re-fusion of tethered sister cells, it remains unsolved why this phenomenon particularly takes place in this lymphoma and what the differences between these cell types of variable sizes are. The aim of the present study was to characterize microdissected small and giant HRS cells by gene expression profiling and to assess differences of clonal gro

A 38-year-old woman was diagnosed with cutaneous anaplastic T-cell lymphoma that proved refractory to methotrexate, bexarotene, denileukin diftitox, interferon γ-1b, interferon α-2b, vorinostat, and pralatrexate. She was therefore started on the newly approved monoclonal anti-CD30 antibody brentuximab vedotin. Treatment with brentuximab 1.8 mg/kg IV every 3 weeks quickly led to disappearance of her cutaneous tumors. The day after her second brentuximab infusion she developed word-finding difficulties and unsteady gait. Due to further neurologic deterioration, she was admitted to an outside hos

BACKGROUND: The immunopathogenesis of type 1 diabetes mellitus is associated with T-cell autoimmunity. To be fully active, immune T cells need a co-stimulatory signal in addition to the main antigen-driven signal. Abatacept modulates co-stimulation and prevents full T-cell activation. We evaluated the effect of abatacept in recent-onset type 1 diabetes.
METHODS: In this multicentre, double-blind, randomised controlled trial, patients aged 6-45 years recently diagnosed with type 1 diabetes were randomly assigned (2:1) to receive abatacept (10 mg/kg, maximum 1000 mg per dose) or placebo infusio

We report a case of rheumatoid vasculitis (RV) that responded well to abatacept, a cytotoxic T lymphocyte-associated antigen 4 (CTLA4)-immunoglobulin fusion protein. A 38-year-old woman developed RV despite treatment with methotrexate and tumor necrosis factor (TNF) inhibitors. The effects of steroid therapy, immunoabsorption plasmapheresis, and interleukin-6 inhibitor were insufficient, however, administration of abatacept rapidly improved her clinical symptoms with almost normalization of the immunological findings. This is the first published case report of the successful treatment of RV wi

After virus infection, cytotoxic T lymphocytes (CTLs) divide rapidly to eradicate the pathogen and prevent the establishment of persistence. The magnitude of an antiviral CTL response is thought to be controlled by the initiation of a cell cycle program within lymphoid tissues. However, it is presently not known whether this division program proceeds during migration or is influenced locally at sites of viral infection. We demonstrate that antiviral CTLs remain in cell cycle while transiting to infected tissues. Up to one third of virus-specific CTLs within blood were found to be in cell cycle

Single-walled carbon nanotubes (CNTs) emit heat when they absorb energy from near-infrared (NIR) light. Tissue is relatively transparent to NIR, which suggests that targeting CNTs to tumor cells, followed by noninvasive exposure to NIR light, will ablate tumors within the range of NIR. In this study, we demonstrate the specific binding of antibody-coupled CNTs to tumor cells in vitro, followed by their highly specific ablation with NIR light. Biotinylated polar lipids were used to prepare stable, biocompatible, noncytotoxic CNT dispersions that were then attached to one of two different neutra

Coeliac disease (CD) is a multifactorial disease for which there is an intensive search for genetic risk factors. Some authors found an association between the CTLA-4 region and CD. In the present work, we investigate the possible implication of the CTLA-4 region as a genetic risk factor for CD, through two statistical approaches: the maximum likelihood score (MLS) test in a large Italian sample of affected sib-pairs using polymorphic genetic markers on chromosome 2, and the transmission disequilibrium test (TDT) in continental Italian and Tunisian families using the CTLA-4 exon 1 49 A/G polym

Susceptibility to Graves' disease (GD) is determined by environmental and genetic factors. The genetic susceptibility to GD is conferred by genes in the human leukocyte antigen (HLA), and several other genes unlinked to HLA are thought to contribute to the development of GD. Three recent papers described the association of GD with the CTLA-4 gene. CTLA-4 is a candidate gene for T-cell mediated autoimmune diseases because it is a negative regulator of T-cell proliferation. As CTLA-4 association with GD may be influenced by the racial composition of the population, it is important to study it in

Head and neck squamous cell carcinoma (HNSCC) and esophageal carcinoma (ESCA) are anatomically contiguous, frequently co-occurring malignancies that collectively account for substantial global cancer mortality. With half of patients progressing to recurrent or metastatic disease, alternative therapeutic strategies are urgently needed. Antibody-drug conjugates (ADCs) offer promise, but their clinical impact requires identification of robust target antigens and elucidation of resistance mechanisms. Through single-cell transcriptomic analysis followed by experimental validation, we identified TRO

Targeted therapy for advanced colorectal cancer (CRC) remains a significant unmet clinical need. Here, we investigate the mechanism of the anti-TROP2 antibody-drug conjugate IMMU132, delivering SN-38 to induce TOP1-mediated DNA damage and cytotoxicity. We further discover that it concurrently suppresses the PERK-eIF2α-ATF4 axis of the unfolded protein response, a key adaptive survival pathway activated by therapy-induced endoplasmic reticulum (ER) stress. This dual action of direct killing and stress adaptation disruption may dismantle a key resistance mechanism. Furthermore, combining IMMU132

BACKGROUND: Myeloid epithelial reproductive tyrosine kinase (MERTK) receptor is overexpressed in cancers and is associated with poor prognosis. RGX-019-MMAE, a novel humanized IgG1-MMAE antibody-drug conjugate (ADC) (Inspirna, Inc), selectively binds to MERTK with high affinity, resulting in internalization and degradation of the receptor. It then induces cytotoxicity through the release of the payload, MMAE (monomethyl auristatin E), which disrupts mitosis. METHODS: MERTK protein expression was analyzed in 818 AML patients using Reverse Phase Protein Arrays (RPPA). Expression was also assesse

Balancing efficacy with off-target toxicity remains a critical challenge in the development of antibody-drug conjugates (ADCs). Emerging ADC technologies, including the design of safer payloads and optimized drug-linkers, offer new avenues to improve both safety and efficacy. Amatoxins bind efficiently to eukaryotic RNA polymerase II at nanomolar concentrations, potently inhibiting transcription and subsequent protein synthesis. In this study, we designed, synthesized, and evaluated a series of novel amatoxin-based bicyclic peptide payloads. From these, three payload-linkers incorporating a Va

PURPOSE: Optimizing metaplastic breast cancer (MpBC) management remains an unmet clinical need. Current literature on MpBC response toantibody drug-conjugates is limited to individual case reports and case series. This study investigates outcomes in metastatic MpBC treated withsacituzumab govitecan (SG) and characterizes actionable targets to inform future studies.
METHODS: Patients (pts) with metastatic triple negative breastcancer (TNBC) treated at the Massachusetts General Hospital Cancer Center between 2000 and 2025 were assessed for MpBC and receipt of SG. MpBCcases were interrogated for

The therapeutic landscape for HER2-positive breast cancer (BC) is evolving rapidly with the advent of antibody-drug conjugates such as trastuzumab deruxtecan (T-DXd). Recently, positive results from the Destiny Breast 011 (DB11) randomized clinical trial were reported, seemingly supporting T-DXd followed by paclitaxel, trastuzumab, and pertuzumab as a potential new neoadjuvant treatment option for high-risk, HER2-positive early breast cancer (eBC). However, in our view, several elements of the trial's design and conduct may have affected the results both qualitatively and quantitatively. These

BACKGROUND: Avoiding unnecessary dose reductions is important for patients with advanced breast cancer (ABC) receiving trastuzumab deruxtecan (T-DXd). Nausea and vomiting, the most common adverse events of T-DXd, frequently necessitate dose reductions, which may impact treatment benefit.
METHODS: This retrospective exploratory study investigated the impact of triple antiemetic regimen (TAR) prophylaxis on T-DXd dose preservation over time. Data from 143 human epidermal growth factor receptor 2 (HER2)-positive or HER2-low ABC patients who received ≥2 T-DXd cycles were stratified based on TAR u

Disitamab vedotin (RC48), a HER2-targeted antibody-drug conjugate, has shown promising activity in locally advanced or metastatic urothelial carcinoma (la/mUC), but clinically useful biomarkers remain lacking. In this retrospective single-center real-world study, we evaluated the efficacy of RC48 and the clinical relevance of tumor miR-145 expression in 136 patients with HER2-positive la/mUC treated between June 2022 and October 2024. Clinical outcomes included objective response rate (ORR), disease control rate (DCR), progression-free survival (PFS), and overall survival (OS). The ORR and DCR

PURPOSE: Ovarian cancer (OC) is frequently diagnosed at late stages after metastasis and chemorefractive leading to poor survival outcomes. There is a critical need for earlier detection and novel antigen-targeted therapies to improve patient survival at all stages. TROP-2 is a transmembrane glycoprotein overexpressed in many cancers and presents a promising target for OC. Recently, antibody-drug conjugates such as sacituzumab govitecan (SG) and datopotamab deruxtecan have been approved for various TROP-2-positive malignancies such as metastatic breast, lung, or urothelial cancer. However, dia
