Chemokine-induced fever: intracerebroventricular actions of MIP-1 and MIP-1 alpha in rats.

The central pyrogenic actions in the rat of doublet macrophage inflammatory protein-1 (MIP-1) and MIP-1 alpha were determined by their intracerebroventricular infusion. Doses of 560 pg and 11.2 ng of MIP-1 or 10.0 ng MIP-1 alpha infused into the third cerebral ventricle induced a long lasting fever. However, MIP-1 alpha was much less potent than MIP-1 in terms of intensity and longer latency. Overall, these cytokines are pyrogenic when acting on the walls of the third ventricle; however, a dose 10 times greater than that injected directly into the anterior hypothalamus is required to evoke fev

Interferon-gamma and a factor derived from trypanosomes cause behavioural changes in the rat.

A newly isolated interferon-gamma (IFN-gamma) immunoreactive molecule, "neuronal IFN-gamma", and recombinant lymphocyte-derived IFN-gamma were injected intracerebroventricularly (i.c.v.) through a previously implanted cannula into adult male rats during both the light and the dark phases of the light/dark cycle. The two molecules caused a reduction in both frequency and duration of rearing and locomotion during the dark, but not the light, phase. A molecule isolated from Trypanosoma brucei brucei, a parasite of the same subspecies of trypanosomes which causes African sleeping sickness, can ind

Role of glucocorticoids in the regulation of brain prostaglandin biosynthesis under basal conditions and in response to endotoxin.

Glucocorticoids (GC) are known to inhibit eicosanoid production in various peripheral tissues; however, their role in the regulation of basal and induced prostaglandin (PG) biosynthesis in the brain is still not clear. In the present study we examined the effect of exogenous dexamethasone (dex) or endogenous GC on basal and on bacterial endotoxin (lipopolysaccharide, LPS) induced ex vivo production of PGE2 by the frontal cortex of rat brain. The experimental groups were: 1) intact rats; 2) rats in whom endogenous GC were removed either by surgical or by chemical (metopirone) adrenalectomy (ade

[Activation of delta-opiate receptor simultaneously stimulates ACTH beta-endorphin and prolactin release].

To evaluate a possible physiological and pathological role of brain delta-opiate receptor and endogenous ligands in the control of ACTH, beta-endorphin (beta-EP) and prolactin (PRL) release, a specific agonist of the delta-opiate receptor--[D-Pen2, D-Pen5]-enkephalin (DPDPE) was microinjected into the third cerebral ventricle (1 microgram or 3 micrograms/3 microliters/rat) in conscious freely-moving male rats. Plasma ACTH, beta-EP and PRL concentrations were measured by radioimmunoassay (RIA). Our results clearly indicate that third ventricular injection of DPDPE significantly elevates plasma

Central insulin administration reduces neuropeptide Y mRNA expression in the arcuate nucleus of food-deprived lean (Fa/Fa) but not obese (fa/fa) Zucker rats.

By acting in the brain, insulin suppresses food intake, whereas neuropeptide Y (NPY) has the opposite effect. Since fasting increases NPY gene expression in the hypothalamic arcuate nucleus (ARC) and also lowers circulating insulin levels, we hypothesized that the anorexiant effect of insulin could result from insulin inhibition of NPY gene transcription in the ARC. Therefore, we determined whether the administration of insulin (200 mU per 12 hrs) into the 3rd cerebral ventricle of lean (Fa/Fa) female Zucker rats (n = 5) during 48 hrs of food deprivation reduces the expression of preproNPY mRN

Neurotensin and neurotensin analogues modify the effects of chronic neuroleptic administration in the rat.

The effects of intracerebroventricular neurotensin and the neurotensin analogues neuromedin N and [D-Trp 11]neurotensin on the behavioural responses to chronic neuroleptic administration were investigated in the rat. Chronic (18 weeks) administration of a low dose (12.5 mg/kg, i.m., every 3 weeks) of fluphenazine decanoate alone failed to elicit the vacuous chewing mouth movements (VCMs) which have previously been reported following higher doses of this drug, but VCMs were seen in neuroleptic-treated animals following the additional administration of neurotensin. A higher dose of fluphenazine

Conditioned taste aversion is disrupted by prolonged retrograde effects of intracerebral injection of tetrodotoxin in rats.

Acquisition of conditioned taste aversion (CTA) is disrupted when 10 ng tetrodotoxin (TTX) is injected into both parabrachial nuclei of rats immediately after saccharin drinking and before LiCl poisoning (Ivanova & Bures, in press). Further analysis of this finding showed that parabrachial TTX injection (a) elicited retrograde amnesia also when applied 1, 2, or 4 days but not 8 days after CTA acquisition; (b) did not abolish CTA produced by 2 or 3 saccharin-LiCl pairings; (c) did not cause persistent increase of quinine threshold; and (d) elicited anterograde CTA amnesia when applied 1 but not

[Emetic and antiemetic properties of regulatory peptides].

Emetic and antiemetic properties of opioid peptides, substance "P", beta-lipotropin, and ACTH1-39 have been investigated in experiments on cats. It was shown that morphine, enkephalin, beta-endorphin and DADLE caused vomiting in animals, which was blocked by naloxone. Substance "P", gamma- and des-tyr-gamma-endorphin manifested antiemetic properties similar to those of naloxone. Selective antagonists of delta-opioid receptors ICI 154, 129 blocked emetic action of delta-agonist DADLE but did not prevent vomiting caused by mu-agonist morphine. It is suggested that the vomiting mechanisms of endo

Comparative behavioral effects of anticholinergic agents in cats: psychomotor stimulation and aggression.

The effect on behavior of eight anticholinergic agents: atropine, scopolamine, trihexyphenidyl, biperiden, homatropine, eucatropine, hexocyclium and propantheline, injected into the cerebral ventricle (ICV) of the cat was investigated and compared. The anticholinergic agents evoked: (1) psychomotor stimulation such as miaowing, loud calling, restlessness, impelling locomotion, jumping, vacant staring, apprehension and loss of interest of the surroundings; (2) aggression, hissing, threat, attack, defense, fighting with paws and flight; (3) autonomic responses including mydriasis, tachypnea, dys

Pertussis toxin blocks the outward currents evoked by opiate and alpha 2-agonists in locus coeruleus neurons.

Pertussis toxin, a substance known to inactivate the inhibitory guanine nucleotide regulatory unit of adenylate cyclase, was injected into the lateral cerebral ventricle of rats; intracellular recordings were made from locus coeruleus neurons in brain slices 1-3 days later. Morphine (an opiate agonist) and clonidine (an alpha 2-agonist) produced the expected outward currents (and associated hyperpolarization and inhibition of firing) in controls whereas the effects of both agonists were blocked in animals pretreated with pertussis toxin. These results are consistent with the hypothesis that op

Participation of cyclophosphamide-resistant T cells in murine lymphocytic choriomeningitis.

A severe inflammatory process can be induced by transferring lymphocytic choriomeningitis virus (LCMV)-immune spleen cell populations into cyclophosphamide (Cy)-suppressed, LCMV-infected recipients. The extent of cellular invasion can be quantified accurately by counting cells obtained from the cisterna magna. The underlying lymphocyte-target cell interactions are apparently very specific, with massive cellular invasion being dependent on the donor and recipient sharing class I major histocompatibility complex glycoproteins. Unlike the situation found by others for delayed-type hypersensitivit

Centrally administered PGE2 inhibits gastric secretion in the rat by releasing vasopressin.

The pituitary mechanism of the central inhibitory effect of PGE2 on gastric secretion was investigated in rats. The intravenous (i.v.) injection of neurointermediate lobe extracts but not of anterior lobe extracts inhibited gastric secretion in pylorus-ligated rats. The i.c.v. administration of 3 micrograms of PGE2 increased the plasma level of vasopressin but had no effect on plasma beta-endorphin/beta-lipotropin. The i.v. administration of 10 micrograms d(CH2)5Tyr(Me) AVP, a vasopressin antagonist, prevented the inhibition of gastric secretion induced by i.c.v. administration of 3 micrograms

Inhibitory alpha 2-adrenergic mechanism regulating gastric secretion in pylorus-ligated rats.

Several alpha-adrenoceptor agonists given intracerebroventricularly or subcutaneously to rats were assessed for their effects on gastric secretion under condition of pylorus ligation. Intracerebroventricular injection of alpha-adrenoceptor agonists reduced gastric secretion, in the following order (relative potency, clonidine = 1): oxymetazoline (100) greater than clonidine (1) greater than methoxamine (0.024) greater than phenylephrine (0.003). The antisecretory effects of oxymetazoline and clonidine given intracerebroventricularly were antagonized with yohimbine administered by the same rout

Perfusion of the fourth cerebral ventricle with the synthetic opioid peptide, FK 33-824, induces dose-related bradycardia and naloxone-reversible respiratory depression in the awake dog.

The importance of natural opioids in the central control of blood pressure, heart rate and respiratory function was evaluated with a synthetic analogue of Met-enkephalin, FK 33-824, which is more resistant to enzymatic degradation than the natural opioids. Increasing concentrations (20, 100, 200 and 400 micrograms/ml), perfused through the 4th cerebral ventricle of the conscious dog, induced a concentration-related depression of respiratory function - as reflected in arterial pCO2 and pO2 and heart rate. Naloxone (40 micrograms/ml) was able to reverse respiratory depression whereas bradycardia

Delta versus mu receptors: cardiovascular and respiratory effects of opiate agonists microinjected into nucleus tractus solitarius of cats.

The cardiovascular and respiratory responses to relatively specific mu or delta agonists microinjected (0.5 microliter/kg) into the region of the nucleus of tractus solitarius (NTS) were examined in anesthetized cats. Blood pressure, heart rate, and respiratory rate were monitored for 30 min after the microinjection of opioid compounds or saline vehicle. The delta agonist, (D-Ala2,D-Leu5)-enkephalin (10-100 nmol/kg) elicited dose-dependent decreases in blood pressure, heart rate, and respiratory rate which were naloxone reversible. In contrast the mu agonists, morphine (10-54 nmol/kg) and morp

Effect of delta sleep inducing peptide (DSIP) and arginine vasotocin (AVT) on sleep and motor activity in the rat.

The effect of Delta Sleep Inducing Peptide (DSIP) and arginine vasotocin (AVT) on motor activity and sleep was investigated in the rat. Motor activity was not significantly reduced during the 24 h following systemic DSIP administration (40-160 nmol/kg i.p.), nor were sleep and the power of the EEG delta-band significantly increased in the 2 hours following injection or infusion of DSIP into the lateral or third ventricle (7-24 nmol). Injection or infusion of AVT into the lateral or third ventricle (10(-15)-10(-19) mol) did not enhance sleep and the power of the EEG delta-band. Nevertheless, th

Mirtazapine, an α2 Antagonist-Type Antidepressant, Reverses Pain and Lack of Morphine Analgesia in Fibromyalgia-Like Mouse Models.

Treatment of fibromyalgia is an unmet medical need; however, its pathogenesis is still poorly understood. In a series of studies, we have demonstrated that some pharmacological treatments reverse generalized chronic pain but do not affect the lack of morphine analgesia in the intermittent cold stress (ICS)-induced fibromyalgia-like pain model in mice. Here we report that repeated intraperitoneal treatments with mirtazapine, which is presumed to disinhibit 5-hydroxytriptamine (5-HT) release and activate 5-HT1 receptor through mechanisms of blocking presynaptic adrenergic α2 and postsynaptic 5-H
