[Usefulness of umbilical cord mercury concentrations as biomarkers of fetal exposure to methylmercury].

In epidemiological studies on the health effect of methylmercury (MeHg) exposure, maternal-hair mercury concentration has been used as an exposure biomarker because of its ease of collection and capability to recapture the exposure history. However, artificial hair-waving reduces the mercury concentration and there is little agreement about which part of the hair strand properly represents fetal exposure. We presented an overview of the studies addressing umbilical cord and mercury in PubMed and evaluated the usefulness of umbilical cord mercury concentrations as biomarkers of fetal exposure t

Maternal placental growth factor mediates the association between PM2.5 and its constituents with adverse pregnancy outcomes.

Maternal placental growth factor (PLGF) is essential for fetal growth, and emerging evidence links PM2.5 exposure to adverse pregnancy outco … n analysis was used to estimate the mediation effects of maternal PLGF levels on these association. Maternal exposure to PM2.5 and its constituents was associated with an increased risk of premature rupture of membrane, preterm birth (PTB), and low birth weight (LBW) during entire pregnancy. Among the constituents, organic matter (OM) demonstrated the greatest adve …

Maternal influences on cord blood lead levels.

We constructed models of umbilical cord blood lead (PbB), with and without the addition of maternal PbB at delivery and earlier in pregnancy, to determine which factors explaining cord PbB depended upon maternal PbB and which did not. We prospectively studied women of low-to-middle socioeconomic status who lived in the Valley of Mexico from 12 weeks of pregnancy to delivery. We measured maternal venous PbB during pregnancy and at delivery, and umbilical cord PbB (1-38 micrograms/dl, 0.05-1.83 mumol/l). We used multiple regression analyses to model cord PbB and a logit analysis to model the mat

Industrial air pollution and low birth weight in New Mexico, USA.

In recent decades, the low birth weight (LBW) rate in New Mexico has consistently exceeded the Unites States average. Maternal exposure to air pollution during pregnancy may be a significant contributor to LBW in offspring. This study investigated the links between maternal residential exposure to air pollution from industrial sources and the risk of LBW in offspring. The analysis included 22,375 LBW cases and 233,340 controls. It focused on 14 common chemicals listed in the Toxic Release Inventory (TRI) and monitoring datasets, which have abundant monitoring samples. The Emission Weighted Pro…

Impact of prenatal exposure to metallic elements on neural tube defects: Insights from human investigations.

Metallic elements play a pivotal role in maternal and fetal health. Metals can cross the placental barrier and be absorbed by fetuses, where they may affect closure of the neural tube during embryonic development. Neural tube defects (NTDs), which result from aberrant closure of the neural tube three to four weeks post-conception, have a multifactorial and complex etiology that combines genetic variants and environmental exposure. Recent advances in population-level association studies have investigated the link between maternal environmental exposure and NTDs, particularly the influence of me

Ethanol exposure during pregnancy persistently attenuates cranially directed blood flow in the developing fetus: evidence from ultrasound imaging in a murine second trimester equivalent model.

BACKGROUND: Ethanol (EtOH) consumption during pregnancy can lead to fetal growth retardation, mental retardation, and neurodevelopmental delay. The fetal brain initiates neurogenesis and vasculogenesis during the second trimester, and depends on maternal-fetal circulation for nutrition and growth signals. We used high-resolution in vivo ultrasound imaging to test the hypothesis that EtOH interferes with fetal brain-directed blood flow during this critical developmental period. METHODS: Pregnant mice were lightly anesthetized on gestational day 12 with an isoflurane/oxygen mixture. We assessed

Traffic-related air pollution, preterm birth and term birth weight in the PIAMA birth cohort study.

BACKGROUND: Maternal exposure to air pollution has been associated with adverse pregnancy outcomes. Few studies took into account the spatial and temporal variation of air pollution levels. OBJECTIVES: To evaluate the impact of maternal exposure to traffic-related air pollution during pregnancy on preterm birth and term birth weight using a spatio-temporal exposure model. METHODS: We estimated maternal residential exposure to nitrogen dioxide (NO(2)), particulate matter (PM(2.5)) and soot during pregnancy (entire pregnancy, 1st trimester, and last month) for 3853 singleton births within the …

Evaluation of the evidence on acetaminophen use and neurodevelopmental disorders using the Navigation Guide methodology.

BACKGROUND: Acetaminophen is the most commonly used over-the-counter pain and fever medication taken during pregnancy, with > 50% of pregnant women using acetaminophen worldwide. Numerous well-designed studies have indicated that pregnant mothers exposed to acetaminophen have children diagnosed with neurodevelopmental disorders (NDDs), including autism spectrum disorder (ASD) and attention-deficit/hyperactivity disorder (ADHD), at higher rates than children of pregnant mothers who were not exposed to acetaminophen. METHODS: We applied the Navigation Guide methodology to the scientific literat

Mechanisms and risk factors for perinatal allergic disease.

Allergies are among the top causes of chronic disease in children. Their pathogenesis classically involves T helper 2 (Th2)-type inflammatio … ied by epigenetic, metabolic, dietary, and microbiome-mediated effects. Here, we review how diverse maternal exposures and mediators signal across the placenta and through nursing perinatally to promote future tolerance or enhance reactivity against allergens. Improved understanding of the mechanisms predisposing for allergic disease in early life can guide the dev …

Association of exposure to second-hand smoke during childhood with blood DNA methylation.

INTRODUCTION: By recent estimates, 40% of children worldwide are exposed to second-hand smoke (SHS), which has been associated with adverse health outcomes. While numerous studies have linked maternal smoking during pregnancy (MSDP) to widespread differences in child blood DNA methylation (DNAm), research specifically examining postnatal SHS exposure remains sparse. To address this gap, we conducted epigenome-wide meta-analyses to identify associations of postnatal SHS and child blood DNAm. METHODS: Six cohorts from the Pregnancy And Childhood Epigenetics (PACE) Consortium (total N = 2,695),

The burden of mild intellectual disability attributed to prenatal exposure to methylmercury in China, 2017.

Methylmercury (MeHg) is a neurodevelopmental toxicant that is widespread in the environment and food. Considering the presence of multiple sources of MeHg exposure in the environment, the burden attributable to different exposure sources needs to be determined. This study aimed to estimate the burden of mild intellectual disability (MID) caused by in-utero exposure to MeHg and identify the attributable burden related to MeHg exposure from different sources in China. We applied the hair mercury concentrations from studies to evaluate the burden of MID associated with maternal MeHg exposure and

Assessing lead exposure in U.S. pregnant women using biological and residential measurements.

There is strong scientific evidence for multiple pathways of human exposure to lead (Pb) in residential settings, particularly for young children; however, less is known about maternal exposure during pregnancy and children's exposure during early lifestages. A robust, multi-faceted secondary analysis was conducted using data collected by the National Institute of Child Health and Human Development in the 2009-2014 National Children's Study Vanguard Studies. Descriptive statistics summarized Pb concentrations of maternal blood, maternal urine, and house dust vacuum samples collected during pre…

Associations between residential proximity to agricultural land use as pesticides exposure and birth outcomes.

Pesticides are widely used globally. Due to their widespread use, exposure to pesticides is of concern. In addition to occupational exposure, residential exposure during pesticide application is a concern for those living in or near agricultural areas. The objective of this study was to analyze the association between residential proximity to agricultural pesticide use and birth outcomes. The association between residential proximity to agricultural land use as pesticide exposure and birth outcomes was explored in a birth cohort including 283 pregnant women from a medical center in Hualien dur

Exposure to environmental chemicals and perinatal psychopathology.

Women are nearly twice as likely to develop mood disorders compared with men, and incidence is greatest during reproductive transitions, including pregnancy and postpartum. Because these periods are characterized by dramatic hormonal and physiologic changes, there is heightened susceptibility to external factors, such as exposure to environmental toxicants, which may play a role in maternal psychopathology. The purpose of this scoping review was to provide an overview of studies conducted in humans and animal models on the effects of nonoccupational exposure to environmental chemicals on mater

In utero exposure to persistent and nonpersistent endocrine-disrupting chemicals and anogenital distance. A systematic review of epidemiological studies†.

Anti-androgenic endocrine-disrupting chemicals (EDCs) can cross the placenta to modify early offspring sexual dimorphic markers. These changes are linked to anogenital distance (AGD), which is an androgen-sensitive anthropometric parameter used as a biomarker of perineal growth and caudal migration of the genital tubercle. This review aimed to summarize strength of evidence for associations of in utero exposure to EDCs with AGD and to identify gaps and limitations in the literature so as to inform future research. We performed an electronic search of English literature in September 2019 in med

Human biomonitoring to evaluate exposure to toxic and essential trace elements during pregnancy. Part B: Predictors of exposure.

Maternal exposure to toxic and essential trace elements represents a surrogate of exposure to the unborn child. Variables of exposure as sociodemographic, lifestyles and diet may contribute to different exposure of pregnant women to specific trace elements. Blood, urine and cord blood samples of 53 pregnant women of the HEALS-EXHES cohort, recruited in Reus (Catalonia, Spain) between 2016 and 2017, were analysed for the concentrations of As, Cd, Co, Cr, Cu, Hg, Mn, Ni, Pb, Se and Zn. Univariate and multivariate models were built in order to assess associations between element concentrations in…

Prenatal paracetamol use and asthma in childhood: A systematic review and meta-analysis.

OBJECTIVES: Some studies have suggested that prenatal paracetamol exposure might associate with the risk of child asthma. However, other studies have not confirmed this result. Therefore, we conducted a meta-analysis to investigate their relationship. METHODS: Two authors searched Pubmed and Embase databases up to June 2016. The strength of the association was calculated with the OR and respective 95% CIs. The random-effects model was chosen to calculate the pooled OR. RESULTS: A total of 13 articles of more than 1,043,109 individuals were included in the meta-analysis. A statistically signi

Utility of a physiologically-based pharmacokinetic model for investigating time-dependent variability in the fetal-to-maternal plasma concentration ratio of digoxin.

The purpose of this study was to elucidate the mechanisms underlying the wide variability in fetal-to-maternal (F/M) concentration ratios of digoxin by comprehensively characterizing maternal and fetal pharmacokinetics using a physiologically based pharmacokinetic (PBPK) model that explicitly incorporates pregnant women, the placenta, and the fetus, with particular emphasis on placental transfer processes and time-dependent concentration dynamics. Maternal–fetal digoxin pharmacokinetics were simulated using a maternal–placental–fetal PBPK model implemented in Simcyp™. Placental transfer was de

Canadian Arctic Contaminants and Their Effects on the Maternal Brain and Behaviour: A Scoping Review of the Animal Literature.

Background: Environmental toxicants such as methylmercury, polychlorinated biphenyls, and organochlorine pesticides are potentially harmful pollutants present in contaminated food, soil, air, and water. Exposure to these ecologically relevant toxicants is prominent in Northern Canadian populations. Previous work focused on toxicant exposure during pregnancy as a threat to fetal neurodevelopment. However, little is known about the individual and combined effects of these toxicants on maternal health during pregnancy and post-partum. Methods: A scoping review was conducted to synthesize the curr
