The statistics of refractive error maps: managing wavefront aberration analysis without Zernike polynomials.

The refractive error of a human eye varies across the pupil and therefore may be treated as a random variable. The probability distribution of this random variable provides a means for assessing the main refractive properties of the eye without the necessity of traditional functional representation of wavefront aberrations. To demonstrate this approach, the statistical properties of refractive error maps are investigated. Closed-form expressions are derived for the probability density function (PDF) and its statistical moments for the general case of rotationally-symmetric aberrations. A close

[Target-controlled infusion. Clinical relevance and special features when using pharmacokinetic models].

Since its commercial introduction in 1996, target-controlled infusion (TCI) has become an established technique for administration of intravenous anaesthetics. Modern TCI systems, however, are characterized by an increasing number of additional options and features, such as the choice between different pharmacokinetic models and modes of application, which may confuse the less experienced user. This review describes the differences between pharmacokinetic models, modes of application and the effect of covariates as well as the consequences for dosing. The aim is to explicate for the user of mo

Using empirical likelihood to combine data: application to food risk assessment.

This article introduces an original methodology based on empirical likelihood, which aims at combining different food contamination and consumption surveys to provide risk managers with a risk measure, taking into account all the available information. This risk index is defined as the probability that exposure to a contaminant exceeds a safe dose. It is naturally expressed as a nonlinear functional of the different consumption and contamination distributions, more precisely as a generalized U-statistic. This nonlinearity and the huge size of the data sets make direct computation of the proble

Zonal matrix iterative method for wavefront reconstruction from gradient measurements.

PURPOSE: To present an alternate method to Zemike decomposition (modal) of wavefront reconstruction using iterative implicit solution to the finite difference equations (zonal). METHODS: Different reconstruction methods, modal and zonal, were compared and the advantages of each method were analyzed. RESULTS: Although the modal or Zemike method allows for quantitative interpretation of some of the aberrations, it is cumbersome for use with fine details and may lead to errors for eyes with keratoconus or other rapidly varying aberration. The zonal method produces a very high-resolution map tha

Air quality modeling of interpollutant trading for ozone precursors in an urban area.

Emission trading is a market-based approach designed to improve the efficiency and economic viability of emission control programs; emission trading has typically been confined to trades among single pollutants. Interpollutant trading (IPT), as described in this work, allows for trades among emissions of different compounds that affect the same air quality end point, in this work, ambient ozone (O3) concentrations. Because emissions of different compounds impact air quality end points differently, weighting factors or trading ratios (tons of emissions of nitrogen oxides (NO(x)) equivalent to a

Statistical issues of quality control in organised breast cancer screening.

BACKGROUND: European guidelines for breast-cancer screening recommend an integrated approach of mammography screening with subsequent assessment and biopsy, if required, in one screening unit under permanent quality control, for which target values are released. Although the calculation of the respective rates (e.g. for participation, assessment, biopsy, or cancer detection) appears trivial, the statistical assessment of their compatibility with the target values is less obvious. This is especially true if subjects with a positive diagnostic result leave the screening-assessment chain prematur

Relative infertility: modeling clinical paradoxes.

OBJECTIVES: To assume that a cause of relative infertility will decrease the monthly chance of conception (fecundability) in a dose-dependent manner and, by a mathematical model, to identify common clinical observations and paradoxes that are explainable within this hypothesis. DESIGN: An empirically based assumption of a population-mean fecundability of 0.2 and the accumulating probability of pregnancy equations and projections were used to examine over time the effects of diminishing such fecundability to one half, one fifth, and one twentieth of normal, and then reversing this effect with

Hemifacial microsomia: developmental consequence of perturbation of the auriculofacial cartilage model?

The cause of hemifacial microsomia (HFM) is currently the subject of much investigation. Despite a large body of clinical and experimental data, little is certain other than the heterogeneity of this malformation complex. Here we suggest that some of the cases previously designated as being multifactorial in origin may be interpreted instead as resulting from a single-gene mutation, by applying a stochastic single-gene model. A variety of models of the pathogenesis of HFM have been described, including the proposal that local embryonic haemorrhage is a causal mechanism. More recently, it has b

Practical implications of modes of statistical inference for causal effects and the critical role of the assignment mechanism.

Causal inference in an important topic and one that is now attracting serious attention of statisticians. Although there exist recent discussions concerning the general definition of causal effects and a substantial literature on specific techniques for the analysis of data in randomized and nonrandomized studies, there has been relatively little discussion of modes of statistical inference for causal effects. This presentation briefly describes and contrasts four basic modes of statistical inference for causal effects, emphasizes the common underlying causal framework with a posited assignmen

A simple method for estimating the contraceptive prevalence required to reach a fertility target.

This report describes and applies a procedure for estimating the increase in contraceptive prevalence that would have to occur to achieve specified future reductions in a population's fertility. This target-setting methodology is based on a previously developed analytic model for the relationship between fertility and its proximate determinants. The basic aggregate version of the estimation procedure, which is described in detail, requires relatively few input data, but it allows changes in the mix of contraceptive methods in future years. In extended versions of the model, age specificity and

LGTM: Gaussian process modulated neural topic modeling for longitudinal microbiome.

Longitudinal microbiome data are key to understanding the dynamics of microbial communities and their relationships with the host and environment. However, analysis of such data is challenging due to high dimensionality, compositionality, irregular sampling and temporal dependencies on external covariates. Existing analytical approaches typically address only subsets of these challenges, limiting their ability to yield biologically interpretable insights. We introduce LGTM, a probabilistic modeling framework that combines flexible non-linear longitudinal modeling with interpretable topic-based

Modeling the emergence of divergent mutants of SARS-CoV-2, "Omicron-like events": A time-to-event analysis.

BACKGROUND: During the COVID-19 pandemic, several divergent mutants including the Omicron (B.1.1.529) BA.1 variant emerged, having a distinct mechanism of emergence compared with pre-existing variants of concern. Apart from playing a major role in causing recurrent epidemic waves, the highly divergent mutants also contributed to changing the fate of the pandemic by exhibiting large differences in phenotypic characteristics among even closely related variants. Given that several different variants emerged during the pandemic, the present study aimed to quantitatively evaluate the risk of emerge

Unveiling the Multifaceted Dynamics of Breast Cancer: A Copula Regression Approach to Modeling and Predicting Outcomes.

This study examines the application of flexible copula regression models to analyze the complex interdependencies among clinical variables in breast cancer data. As the most commonly diagnosed cancer and the second leading cause of cancer-related deaths among women worldwide, breast cancer presents both clinical and analytical challenges. Unlike traditional multivariate approaches, copulas offer greater flexibility in capturing complex, nonlinear, and asymmetric dependencies between mixed-type outcomes. The present study examines copula-based regression models to investigate the joint behavior

Robust Heterogeneity Adjustment for Gaussian Graphical Model With Latent Variables.

Graphical models serve as fundamental tools for encoding conditional dependence structures in multivariate biological data, with latent variable Gaussian graphical models playing a pivotal role in capturing complex dependencies in the presence of unobserved confounding variables. However, practical implementations often face two critical challenges: systematic heterogeneity arising from unobserved subpopulations (e.g., tumor subtypes, cell clusters, or patient stratifications) and outliers (e.g., technical artifacts or rare phenotypic variations), both of which can substantially distort the un

Time-Dependent Predictive Accuracy Metrics in the Context of Interval Censoring and Competing Risks.

Evaluating the performance of a prediction model is a common task in medical statistics. Standard accuracy metrics require the observation of the true outcomes. This is typically not possible in the setting with time-to-event outcomes due to censoring. Interval censoring, the presence of time-varying covariates, and competing risks present additional challenges in obtaining those accuracy metrics. In this study, we propose two methods to deal with interval censoring in a time-varying competing risk setting: a model-based approach and the inverse probability of censoring weighting (IPCW) approa

Bayesian Power-Based Sample Size Determination for Diagnostic Accuracy Studies.

In diagnostic accuracy studies, it is crucial to estimate the required sample size beforehand for the appropriate evaluation of diagnostic measures (e.g., sensitivity and/or specificity). In the development of pharmaceuticals and medical devices, although frequentist approaches have been used for sample size determination, Bayesian approaches have gained attention recently. We propose a Bayesian method for sample size determination using two kinds of priors (i.e., analysis prior and design prior) based on "Bayesian power." In the proposed method, an analysis prior and a design prior were intro

Optimal Dynamic Treatment Regimes for High-Dimensional Accelerated Failure Time Model.

The increasing prevalence of high-dimensional covariates presents a significant challenge to precision medicine. To overcome this, we propose a novel multi-stage optimal dynamic treatment regime method specifically designed for high-dimensional accelerated failure time models. Our approach, which aims to maximize individual survival time, employs backward induction with counterfactual survival times as the optimization objective at each stage. Parameter estimation leverages weighted counterfactual survival times to account for censoring, followed by the slope determination by analysis of resid

Identification of Changes in Gene Expression.

Evaluating the change in gene expression is a common goal in many research areas, such as in toxicological studies, which are particularly important in pre-clinical research. In practice, the analysis is often based on multiple t-tests evaluated at the observed time points of the experiment, which limits the accuracy of determining the precise time at which the gene changes in expression. If a parametric approach is chosen, the analysis is often restricted to identifying the onset of an effect, but not its length. In this paper, we propose a parametric method to identify the time frame during
