Population ecology and biogeochemical implications of ssDNA and dsDNA viruses along a permafrost thaw gradient.

Anthropogenic-driven climate change is accelerating permafrost thaw, threatening to release vast carbon stores through increased microbial activity. While microbial roles are increasingly studied, the contributions of viruses remain largely unexplored, in part due to soil-associated technical challenges that have hindered their detection and characterization. Here, we applied an optimized virion enrichment workflow along a permafrost thaw gradient, identifying 9,963 viral populations (vOTUs), including single- and double-stranded DNA viruses, with 99.9% novelty compared to other soils. A pectic polysaccharide from wax gourd peel improves ethanol-induced gastric ulcer injury in mice via the PI3K/AKT signaling pathway.

The pectic polysaccharide WGPP-1a (46.8 kDa) has been obtained from wax gourd peel (WGP) which is a popular soup ingredient and Chinese herbal medicines in China. Monosaccharide composition, methylation analysis, FT-IR and NMR demonstrate that WGPP-1a is composed of rhamnogalacturonan I (RG-I) and homogalacturonan (HG) domains with mass ratios of 4:1. The RG-I domains contain a highly branched structure that is substituted primarily with β-D-1,4-galactan, α-L-1,5-arabinan and β-D-1,4-glucan. In ethanol-induced gastric ulcer mice, WGPP-1a reduced ulcer area, restored mucosal glycoproteins. In v

Ethanol-Water Co-Solvent Extraction Optimizes the Yield and Bioactivity of Polysaccharides and Essential Oils From Morel Mushroom.

Morel mushrooms are valued for their bioactive polysaccharides and essential oils, yet optimal extraction methods remain underexplored. This study systematically compared four solvent systems (water, ethanol, water-ethanol, and ethanol-water) for the simultaneous extraction of polysaccharides and essential oils. The results demonstrated that the ethanol-water method achieved better extraction efficiency for both polysaccharides (6.99% yield) and essential oils (37.81% yield). In comparison, the conventional methods had much lower yields; for example, the water method only extracted 6.52% polys

Structural characterization and immunomodulatory activity of a glycogen from Meretrix lyrata.

Marine shellfish are not only important food ingredients in human diets, but also their tissues contain polysaccharides with significant bioactivities and pharmacological effects. In this study, a glucan (MLP60-1) was successfully isolated from the soft tissue of Meretrix lyrata. Structural characterization through monosaccharide compositions, methylation analysis, acid and enzymatic hydrolysis, mass spectrometry, and nuclear magnetic resonance spectroscopy suggested that MLP60-1 is a highly branched glycogen defined by a branching frequency of approximately one branch per 3.3 glucose residues

Lipid-lowering effects of bile Arisaema polysaccharides on high-fat diet-induced hyperlipidemia: An integrated analysis of metabolomics, lipidomics and microbiome.

Bile Arisaema, a traditional Chinese medicine, has been previously identified by our team to possess antipyretic properties attributed to its polysaccharide component. Recently, we have confirmed that bile Arisaema played a significantly lipid-lowering effect on hyperlipidemia rats. Building upon this discovery, the present study aimed to investigate the unexplored hypolipidemic potential of its polysaccharide component and elucidate the underlying mechanisms. A soluble polysaccharide fraction devoid of free proteins, named BAPs, was extracted from bile Arisaema using a combination of hot wate

Aromatic residue Trp146 mediates dynamic interplay between type IVa pili and exopolysaccharide in social motility of Myxococcus xanthus.

The social motility (S-motility) of Myxococcus xanthus is driven by the intricate interplay between type IVa pili (T4aP) and exopolysaccharide (EPS). Although T4aP retraction furnishes the mechanical force necessary for cellular movement, EPS serves as a matrix for T4aP anchoring and instigates pilus retraction. Despite this interdependence, the molecular determinants governing specific T4aP-EPS recognition and interaction have yet to be fully elucidated. Utilizing a comprehensive approach, our study identifies glucosamine as the key monosaccharide within EPS that interacts with the major pili

Integral kidney function assessment in pediatric patients with glycogen storage diseases.

INTRODUCTION: Glycogen storage diseases (GSDs) are a group of hereditary metabolic disorders with variable clinical manifestations, depending on the enzyme and organ affected. Renal dysfunction, including hyperfiltration, proteinuria, and renal tubular acidosis (RTA), is a known complication, particularly in GSD types of Ia and Ib. METHODS: This cross-sectional study evaluated renal function in 17 pediatric patients with different GSD types using an integral kidney assessment (IKA). The comprehensive evaluation included biochemical and urinary analyses, glomerular filtration rate calculations

Acute physiological effects on macromolecule digestion following oral ingestion of the enzyme blend Elevase® in individuals that had undergone an ileostomy, but were otherwise healthy-a randomized, double blinded, placebo-controlled exploratory study.

Digestive enzymes can selectively degrade proteins, carbohydrates and lipids; and their supplementation alongside food may accelerate the breakdown of complex food matrices, facilitate greater nutrient absorption, decrease food sensitivities and aid in the management of certain disease states. Several intrinsic and extrinsic factors govern food digestion and for every individual this phenomenon is unique. This study was conducted as a randomized, crossover, placebo-controlled design where each participant served as their own control. This post-hoc analysis investigated the impact of a dietary

PEG-based-ultrasound-microwave synergistic extraction of mucilage polysaccharides from chia seed: Structural characterization and bioactivity.

In this study, the effects of microwave, ultrasound, ultrasound-microwave synergy, and polyethylene glycol (PEG)-200 on the chia seed mucilage's (CSM) structural, thermal, and functional properties have been evaluated by modern spectroscopic techniques, crystal diffraction, and thermal analyses. The results showed that CSM, extracted by PEG-200 aqueous solution has the largest particles (1120-1231 μm), and the smallest size (44.06-317.19 μm) has been observed in microwave extraction. Furthermore, microwave exposure seems to impact the primary structure of CSM profoundly, evidenced by shifts an

Back to the Basics of SARS-CoV-2 Biochemistry: Microvascular Occlusive Glycan Bindings Govern Its Morbidities and Inform Therapeutic Responses.

Consistent with the biochemistry of coronaviruses as well established over decades, SARS-CoV-2 makes its initial attachment to host cells through the binding of its spike protein (SP) to sialylated glycans (containing the monosaccharide sialic acid) on the cell surface. The virus can then slide over and enter via ACE2. SARS-CoV-2 SP attaches particularly tightly to the trillions of red blood cells (RBCs), platelets and endothelial cells in the human body, each cell very densely coated with sialic acid surface molecules but having no ACE2 or minimal ACE2. These interlaced attachments trigger th

D-Mannose reduces cellular senescence and NLRP3/GasderminD/IL-1β-driven pyroptotic uroepithelial cell shedding in the murine bladder.

Aging is a risk factor for disease via increased susceptibility to infection, decreased ability to maintain homeostasis, inefficiency in combating stress, and decreased regenerative capacity. Multiple diseases, including urinary tract infection (UTI), are more prevalent with age; however, the mechanisms underlying the impact of aging on the urinary tract mucosa and the correlation between aging and disease remain poorly understood. Here, we show that, relative to young (8-12 weeks) mice, the urothelium of aged (18-24 months) female mice accumulates large lysosomes with reduced acid phosphatase

Decreased nutrient digestibility due to viscosity is independent of the amount of dietary fibre fed to growing pigs.

Fibre content and its effect on chyme viscosity are associated with changes in the digestive system of humans and pigs. It is unclear if fibre content and viscosity affect digestive function independently or interactively. We evaluated apparent ileal digestibility (AID) of nutrients and intestinal function in thirty-six ileal-cannulated barrows fed for 29 d either maize-soyabean meal (MSBM) or high-fibre MSBM + 30 % distillers dried grains with solubles (MSBM + DDGS) modified to three levels of viscosity by adding 5 % non-viscous cellulose (CEL), 6·5 % medium-viscous carboxymethylcellulose (MC

Integrated Multiomics Reveals Glucose Use Reprogramming and Identifies a Novel Hexokinase in Alcoholic Hepatitis.

BACKGROUND & AIMS: We recently showed that alcoholic hepatitis (AH) is characterized by dedifferentiation of hepatocytes and loss of mature functions. Glucose metabolism is tightly regulated in healthy hepatocytes. We hypothesize that AH may lead to metabolic reprogramming of the liver, including dysregulation of glucose metabolism. METHODS: We performed integrated metabolomic and transcriptomic analyses of liver tissue from patients with AH or alcoholic cirrhosis or normal liver tissue from hepatic resection. Focused analyses of chromatin immunoprecipitation coupled to DNA sequencing was per

Generation of 13C-Labeled MUC5AC Mucin Oligosaccharides for Stable Isotope Probing of Host-Associated Microbial Communities.

Stable isotope probing (SIP) has emerged as a powerful tool to address key questions about microbiota structure and function. To date, diverse isotopically labeled substrates have been used to characterize in situ growth activity of specific bacterial taxa and have revealed the flux of bioavailable substrates through microbial communities associated with health and disease. A major limitation to the growth of the field is the dearth of biologically relevant "heavy" labeled substrates. Mucin glycoproteins, for example, comprise an abundant source of carbon in the gut, oral cavity, respiratory t

Synthesis of polyfluoroalkyl sp2-iminosugar glycolipids and evaluation of their immunomodulatory properties towards anti-tumor, anti-leishmanial and anti-inflammatory therapies.

Immunomodulatory glycolipids, among which α-galactosylceramide (KRN7000) is an iconic example, have shown strong therapeutic potential in a variety of conditions ranging from cancer and infection to autoimmune or neurodegenerative diseases. A main difficulty for those channels is that they often provoke a cytokine storm comprising both pro- and anti-inflammatory mediators that antagonize each other and negatively affect the immune response. The synthesis of analogues with narrower cytokine secretion-inducing capabilities is hampered by the intrinsic difficulty at controlling the stereochemical

O2 sensing-associated glycosylation exposes the F-box-combining site of the Dictyostelium Skp1 subunit in E3 ubiquitin ligases.

Skp1 is a conserved protein linking cullin-1 to F-box proteins in SCF (Skp1/Cullin-1/F-box protein) E3 ubiquitin ligases, which modify protein substrates with polyubiquitin chains that typically target them for 26S proteasome-mediated degradation. In Dictyostelium (a social amoeba), Toxoplasma gondii (the agent for human toxoplasmosis), and other protists, Skp1 is regulated by a unique pentasaccharide attached to hydroxylated Pro-143 within its C-terminal F-box-binding domain. Prolyl hydroxylation of Skp1 contributes to O2-dependent Dictyostelium development, but full glycosylation at that pos

Food and functional dyspepsia: a systematic review.

BACKGROUND: Functional dyspepsia (FD) is a debilitating functional gastrointestinal disorder characterised by early satiety, post-prandial fullness or epigastric pain related to meals, which affects up to 20% of western populations. A high dietary fat intake has been linked to FD and duodenal eosinophilia has been noted in FD. We hypothesised that an allergen such as wheat is a risk factor for FD and that withdrawal will improve symptoms of FD. We aimed to investigate the relationship between food and functional dyspepsia. METHODS: Sixteen out of 6451 studies identified in a database search o

[Fructose and fructose intolerance].

Although fructose was discovered in 1794, it was realised in recent decades only that its malabsorption can lead to intestinal symptoms while its excessive consumption induces metabolic disturbances. Fructose is a monosaccharide found naturally in most fruits and vegetables. Dietary intake of fructose has gradually increased in the past decades, especially because of the consumption of high fructose corn syrup. With its 16.4 kg/year consumption, Hungary ranks secondly after the United States. Fructose is absorbed in the small intestine by facilitated transport mediated by glucose transporter p

Natural antisense transcript for hyaluronan synthase 2 (HAS2-AS1) induces transcription of HAS2 via protein O-GlcNAcylation.

Changes in the microenvironment organization within vascular walls are critical events in the pathogenesis of vascular pathologies, including atherosclerosis and restenosis. Hyaluronan (HA) accumulation into artery walls supports vessel thickening and is involved in many cardiocirculatory diseases. Excessive cytosolic glucose can enter the hexosamine biosynthetic pathway, increase UDP-N-acetylglucosamine (UDP-GlcNAc) availability, and lead to modification of cytosolic proteins via O-linked attachment of the monosaccharide β-N-GlcNAc (O-GlcNAcylation) from UDP-GlcNAc by the enzyme O-GlcNAc tran
