p53 is the most frequently mutated tumour-suppressor gene in human cancers. Mutant p53 is thought to contribute to carcinogenesis by the acquisition of gain-of-function properties or through the exertion of dominant-negative (DN) effects over the remaining wild-type protein. However, the context in which the DN effects are observed is not well understood. We have therefore generated 'knock-in' mouse embryonic stem (ES) cells to investigate the effects of expressing a commonly found hot-spot p53 mutant, R246S -- the mouse equivalent of human R249S, which is associated with hepatocellular carcin

Reports that selective estrogen receptor modulators (SERMs) reduce occurrence of only estrogen receptor (ER) positive tumors strongly support the etiological distinction between ER positive and ER negative breast cancers. Based on these evidences three concepts are described: Concept I. The occurrence of ER negative tumor cells might be a consequence of the clonal selection among tumor cells. This would lead to mosaicism in the ER expression. If ER negative cells become the most prevalent clone, the patient will be diagnosed to have an ER negative breast cancer. Since all cancers start as ER p

Hepatocellular carcinoma (HCC) is a highly lethal malignancy with limited treatment options, particularly for patients with advanced stages of the disease. Sorafenib, the standard first-line therapy, faces significant challenges due to the development of drug resistance. Yu et al explored the mechanisms by which lncRNA KIF9-AS1 regulates the stemness and sorafenib resistance in HCC using a combination of cell culture, transfection, RNA immunoprecipitation, co-immunoprecipitation, and xenograft tumor models. They demonstrate that N6-methyladenosine-modified long non-coding RNA KIF9-AS1 acts as

Lung cancer remains the leading cause of cancer-related mortality worldwide, largely due to therapeutic resistance and tumor recurrence. In non-small cell lung cancer (NSCLC), cancer stem cells (CSCs) have emerged as key contributors to tumor persistence and progression. However, accelerated CSC-targeted drug discovery is limited by the lack of scalable and quantitative screening systems capable of capturing stemness-associated phenotypes. Conventional two-dimensional (2D) assays often fail to reflect the architectural and molecular complexity of CSC-enriched tumor populations. Here, we report

Gastric cancer is a significant global health concern due to its high morbidity and mortality. Gastric cancer-associated mesenchymal stem cells (GC-MSCs) significantly contribute to its progression, with circ_0024107 being notably elevated in these cells and essential for their tumor-promoting activities. However, the expression and function of this circRNA in gastric cancer cells as well as its upstream regulators remain unclear. qPCR was used to assess circ_0024107 expression levels. Gain- and loss-of-function experiments evaluated its roles. Transwell assays measured cell migration and inva

The cost of cancer care globally is unsustainable and strategies to reduce the mounting burden of cancer are urgently needed. One approach is the use of preventive therapies to reduce cancer risk; dietary-derived compounds with good safety profiles represent a promising source of potential candidates but translating encouraging preclinical data to successful trials presents significant challenges. Development of curcumin, from the spice turmeric, as a preventive therapy for colorectal cancer (CRC) is hindered by poor understanding of its mechanism of action. Using patient derived xenografts an

Eukaryotic cells can synthesize formyl-methionine (fMet)-containing proteins not only in mitochondria but also in the cytosol to some extent. Our previous study revealed substantial upregulation of N-terminal (Nt)-fMet-containing proteins in the cytosol of SW480 colorectal cancer cells. However, the functional and pathophysiological implications remain unclear. Here, we demonstrated that removal of the Nt-formyl moiety of Nt-fMet-containing proteins (via expressing Escherichia coli PDF peptide deformylase) resulted in a dramatic increase in the proliferation of SW480 colorectal cancer cells. Breast cancer stem cells are mainly responsible for poor prognosis, especially in triple-negative breast cancer (TNBC). In a previous study, we demonstrated that ε-Sarcoglycan (SGCE), a type Ⅰ single-transmembrane protein, is a potential oncogene that promotes TNBC stemness by stabilizing EGFR. Here, we further found that SGCE depletion reduces breast cancer stem cells, partially through inhibiting the transcription of FGF-BP1, a secreted oncoprotein. Mechanistically, we demonstrate that SGCE could interact with the specific protein 1 transcription factor and translocate into the nucleus, whic

Despite the fact that much research has focused on gastric cancer, it is still a worldwide concern, because of the difficulties with factors such as signaling pathway crosstalk and gastric cancer stem cell (GCSC). Placental growth factor (PlGF) is one of these factors, and its tumorigenicity potential still remains a question. As a result, we have investigated the effect of PlGF knockdown on apoptosis and genes involved in the Wnt signaling pathway, and apoptosis in cancer stem cells derived from AGS an MKN-45 gastric cancer cell lines. We isolated GCSCs from MKN-45 and AGS cell lines on a non

Smooth muscle (SM) is a spontaneously contractile tissue that provides physical support and function to organs such as the uterus. Uterine smooth muscle-related neoplasia comprise common well-differentiated benign lesions called leiomyomas (ULM), and rare, highly aggressive and pleomorphic tumors named leiomyosarcomas (ULMS). MicroRNAs (miRNAs) are small non-coding RNAs that play essential roles in normal cellular development and tissue homeostasis that can be used to accurately subclassify different tumor types. Here, we demonstrate that miRNAs are required for full smooth muscle cell (SMC) d

Acute myeloid leukemia (AML) is an aggressive hematological malignancy that is sustained by leukemia-initiating cells (LICs). Although protein tyrosine phosphatase 4A2 (PTP4A2), also known as phosphatase of regenerating liver 2 (PRL2), is highly expressed in AML, the mechanisms by which PTP4A2 promotes leukemogenesis are largely unexplored. In this study, we demonstrate that PTP4A2 promotes AML by inhibiting the p53 tumor suppressor pathway in LICs. Using KMT2A-MLLT3-driven AML as a model, we found that PTP4A2 deficiency activates p53 and induces LIC apoptosis and senescence, thereby extending

Hepatocellular carcinoma (HCC), the most prevalent form of primary liver cancer, is driven by cancer stem cells (CSCs) and an immunosuppressive tumor microenvironment, which may underlie the limited efficacy of immune checkpoint blockade therapy. Here, we report that HEY1 plays a crucial role in sustaining HCC stemness and undergoes polyubiquitylation during liver CSC differentiation. Mechanistically, USP28 interacts with HEY1 and deubiquitinates its lysine 87 residue, thereby stabilizing HEY1 and enhancing the stem-like properties of liver cancer cells. Moreover, IKKβ phosphorylates HEY1 at s

Acute myeloid leukemia (AML) is the most common acute leukemia in adults. Although advances in targeted agents have greatly improved the prognosis of patients with AML in recent years, those who fail to achieve remission or relapse after remission are still in urgent need of novel therapeutic strategies. The hypoxia signaling pathway is involved in various biological processes, and hypoxia-inducible factor alpha (HIF-α) is considered a potential therapeutic target in AML. The bone marrow microenvironment is known to be in a state of chronic hypoxia, which is important for hematopoietic stem ce

The role of splicing dysregulation in cancer is underscored by splicing factor mutations; however, its impact in the absence of such rare mutations remains poorly understood. Prompted by the finding that splicing uniquely resolved genetic subtypes of cancer, we developed an unsupervised computational workflow called OncoSplice to comprehensively define tumor molecular landscapes. In adult and pediatric acute myeloid leukemia (AML), OncoSplice identified the spectrum of driver genetics from splicing profiles alone, defined more than a dozen previously unreported molecular subtypes recurrent acr

Currently, tumor treatment strategies centered on Ca2+ homeostasis imbalance primarily target mature tumor cells while neglecting the eradication of cancer stem cells (CSCs), which results in the long-standing unresolved challenge of tumor relapse in later stages of treatment. To tackle the challenge, a calcium peroxide nanoparticle (CPN)-based strategy aimed at inducing an intracellular calcium ion storm in CSCs for efficient cancer therapy was proposed in this study. Typically, hyaluronic acid (HA)-stabilized hollow mesoporous calcium peroxide nanoparticles (HMCPN@HA4) were synthesized via a

Type-I photosensitizers are advantageous for treating hypoxic tumors compared to singlet oxygen (1O2)-based photodynamic therapy (PDT) agents. However, efficient and selective Type-I reactive oxygen species (ROS) generators remain scarce. Here, Limantrafin (Notch1 transcription inhibitor) conjugated BODIPY photosensitizer is reported that selectively produces superoxide radicals (O₂•⁻) upon green light irradiation and exhibits a significantly higher phototherapeutic index (PI) than its structural analog. Theoretical studies and delayed photoluminescence measurements indicate that efficient int

BACKGROUND: Cancer stem cells (CSCs) contribute to bladder cancer (BCa) progression and treatment resistance. Ferroptosis, an iron-dependent cell death, influences CSC maintenance, but its post-translational regulation remains unclear. This study investigated how the deubiquitinase OTUB1 regulated ferroptosis and CSC-like traits in BCa through interaction with SLC7A11.
METHODS: OTUB1 expression was assessed in clinical BCa tissue pairs (tumour vs adjacent normal) collected from patients undergoing surgical resection and in cell lines. Gain- and loss-of-function models were established in BCa

Medulloblastoma (MB) is the most common malignant childhood brain tumour1,2, yet the origin of the most aggressive subgroup-3 form remains elusive, impeding development of effective targeted treatments. Previous analyses of mouse cerebella3-5 have not fully defined the compositional heterogeneity of MBs. Here we undertook single-cell profiling of freshly isolated human fetal cerebella to establish a reference map delineating hierarchical cellular states in MBs. We identified a unique transitional cerebellar progenitor connecting neural stem cells to neuronal lineages in developing fetal cerebe
