Cells of origin of the branches of the facial nerve: a retrograde HRP study in the rabbit.

The origin of different branches of the facial nerve in the rabbit was determined by using retrograde transport of HRP. Either the proximal stump of specific nerves was exposed to HRP after transection, or an injection of the tracer was made into particular muscles innervated by a branch of the facial nerve. A clear somatotopic pattern was observed. Those branches which innervate the rostral facial musculature arise from cells located in the lateral and intermediate portions of the nuclear complex. Orbital musculature is supplied by neurons in the dorsal portion of the complex, with the more r

Subcortical afferent and efferent connections of the superior colliculus in the rat and comparisons between albino and pigmented strains.

Subcortical connections of the superior colliculus were investigated in albino and pigmented rats using retrograde and anterograde tracing with horseradish peroxidase (HRP), following unilateral injection of HRP into the superior colliculus. Afferents project bilaterally from the parabigeminal nuclei, the nucleus of the optic tract, the posterior pretectal region, the dorsal part of the lateral posterior-pulvinar complex and the ventral nucleus of the lateral lemniscus; and ipsilaterally from the substantia nigra pars reticulata, the pars lateralis of the ventral lateral geniculate nucleus, th

Guidance of neuronal growth cones in the grasshopper embryo. I. Recognition of a specific axonal pathway by the pCC neuron.

The selective affinities that growth cones display for specific axonal surfaces give rise to stereotyped patterns of selective fasciculation. Previous studies on cell recognition by neuronal growth cones in the grasshopper embryo led to the proposal and initial experimental testing of the labeled-pathways hypothesis. Here we report on a further experimental analysis of this hypothesis, using the first 3 longitudinal axon fascicles, which initially contain only the axons of 7 identified neurons. We describe and experimentally test the selective affinity of the pCC growth cone for the MP1 and dM

Organization of motoneuronal pools in the rostral spinal cord of the sea robin, Prionotus carolinus.

The functional organization of the motoneurons in the spinal cord of the sea robin, Prionotus carolinus, was studied by means of retrograde transport of horseradish peroxidase (HRP). This species has a complex pectoral apparatus which includes not only a webbed fin, but also three independently mobile fin rays. The motoneurons in the rostral spinal cord fall into two longitudinal columns: dorsal and ventral. The motoneurons of the ventral column innervate the appendicular musculature of the pectoral apparatus. Within the ventral motor column of the rostral spinal cord, four distinct motoneuron

Pathways for orofacial pain sensation in the trigeminal brain-stem nuclear complex of the Macaque monkey.

Eleven Macaque monkeys underwent a variety of lesions in the trigeminal afferent system; namely, tractotomy, rhizotomy, and radiofrequency destruction of various components of the bulbospinal trigeminal nuclear complex. Behavioral responses were evaluated before and after the lesions using a quantitative paradigm which measured lever-pressing in response to electrical stimulation of the dental pulp or facial skin, and by assessing adversive responses to facial cutaneous and intraoral pin-scratch. Thresholds for lever-pressing in response to cutaneous facial stimulation were elevated by tractot

The olivocerebellar projection to the uvula in the mouse.

The organization of the olivocerebellar projection in the mouse was studied with the use of microinjections of horseradish peroxidase (HRP) or HRP conjugated to wheat germ agglutinin (WGA-HRP). Injections were made in medial, intermediate, and lateral sites along the width of the uvula. The purpose of this investigation was to determine the subnuclear origin of olivary afferents to different mediolateral regions of the uvula. Injections made in or adjacent to the midline of the uvula resulted in the retrograde labeling of cells, bilaterally in the caudal portion of the medial accessory olive (

Retrograde and anterograde-transneuronal degeneration in the parabigeminal nucleus following tectal lesions in developing rats.

Degenerative effects in the parabigeminal nucleus were studied in adult rats that had received lesions of the superior colliculus at different postnatal ages. Unilateral lesions lead to complete degeneration of the dorsal and ventral divisions of the parabigeminal nucleus, which connect reciprocally with the damaged tectum, and produce effects on the middle division which receives afferents from the ipsilateral and projects to the contralateral tectum. On the side contralateral to the lesion there was a reduction in the cross-sectional area of the division as a consequence of retrograde degene

The ponto-cerebellar projection in the rat: differential projections to sublobules of the uvula.

Microinjections of horseradish peroxidase (HRP) were made in different sublobules of the uvula (lobule IX, a, b, and c of Larsell, 1952) of the cerebellar cortex in the rat. These injections resulted in retrograde labeling of cells located in the pontine nuclei. Sublobule IXa receives a predominant input from a single column of cells situated in a dorsointermediate position in the caudal pontine nuclei. Sublobules IXb and IXc receive a pontine projection from two different columns of cells, one medial and one lateral. The location of the labeled cells in the lateral part of the caudal pons sug

Lipomas of the corpus callosum and epilepsy.

An analysis of four cases of lipomas of the corpus callosum with epilepsy, and a review of the literature, have led to the following conclusions: (1) Epilepsy as an almost constant feature is often severe, nearly always partial, and begins before the age of 15. (2) Pathophysiology of the seizures appears to be essentially an interhemispheric disconnection (rather than the classic theory that seizures depend upon an infiltration of the cingulate gyri by fibrous tissue growing out from the capsule of the lipoma). This disconnection is responsible for a facilitatory and disinhibitory action that

Potentials evoked by alvear tract in hippocampal CA1 region of rats. I. Topographical projection, component analysis, and correlation with unit activities.

1. The field potentials and unit activities evoked by the alvear tract (AT) in CA1 region of the dorsal hippocampus of rats were studied under sodium pentobarbital anesthesia. 2. The localized activity evoked anterior to an AT stimulus began as a compound action potential, followed by a slower negative wave, and ended in a long-lasting, slow positive wave. Observed with a 64-electrode recording array, topographical projections of the AT in CA1 were seen as parallel strips inclined at an angle of 5-30 degrees medially from the sagittal plane. 3. Three overlapping components in the averaged evok

Projections to the inferior olive of the cat. I. Comparisons of input from the dorsal column nuclei, the lateral cervical nucleus, the spino-olivary pathways, the cerebral cortex and the cerebellum.

The present experiments compare the projections from the dorsal column nuclei (DCN), the lateral cervical nucleus (LCN), the spino-olivary pathways (SO), the motor cortex (Ms1) and the cerebellum (CB) to the inferior olive of the cat. A differential labeling strategy was used for these comparisons. It was found that projections from the contralateral DCN and LCN and the ipsilateral SO overlap extensively with each other in the dorsal accessory division of the inferior olive and the caudal half of the medial accessory olive. Projections from the contralateral motor cortex partially overlap thes

Lateral cervical nucleus in the cat: functional organization and characteristics.

1. The lateral cervical nucleus (LCN) was investigated with extracellular recordings in the anesthetized cat. A total of 556 LCN units were characterized; the locations of most of these were histologically verified. Half of these had receptive fields on the rostral third of the ipsilateral body surface including the face; 14% had fields on the thorax or abdomen, 33% had fields on the hindlimb or tail, and about 3% had receptive fields larger than one limb. 2. The LCN was observed to be somatotopically organized in experiments using angled microelectrode penetrations. Hindlimb units were dorsol

Properties of intraoral mechanoreceptors represented in the mesencephalic nucleus of the fifth nerve in the cat.

1. The activity of neurones in the mesencephalic nucleus of the fifth nerve that respond to forces applied to the teeth were recorded using extracellular microelectrodes; the properties of these neurones have been studied. 2. Electrophysiological evidence consistent with the view that primary afferent intraoral mechanoreceptor fibres have their cell bodies in the trigeminal mesencephalic nucleus is presented. 3. Two groups of intraoral mechanoreceptor neurones were found. The first group, the periodontal mechanoreceptor neurones, which have been described by previous workers, responded to elec

A new major projection from locus coeruleus: the main source of noradrenergic nerve terminals in the ventral and dorsal columns of the spinal cord.

Almost all catecholamine (CA)-containing nerve terminals in the ventral column, intermediate grey and ventral half of the dorsal column disappeared after bilateral stereotaxic lesions of nucleus locus coeruleus, as revealed by fluorescence histochemistry. Some of the CA nerve terminals in the dorsal half of the column seemed to be unaffected by the lesions, as well as the CA terminals innervating the thoracic sympathetic lateral column and the band of nerve terminals crossing the midline and innervating the central grey. This coeruleo-spinal pathway in the rat is located in the anterior funicu

Projection-defined hypothalamic outputs differentially regulate thermogenesis and lipolysis.

The ventromedial hypothalamus (VMH) is a key regulator of energy homeostasis, linking central neural activity to peripheral metabolic function. Within the VMH, neurons expressing steroidogenic factor-1 (SF-1; VMHSF1) are essential for regulating energy expenditure, yet the projection-defined pathways through which they differentially control distinct adipose depots remains unclear. Here, we use projection-specific optogenetic and chemogenetic manipulations in SF1-Cre mice to identify two anatomically and functionally distinct VMHSF1 output pathways with complementary metabolic roles. Axon guidance pathways and the control of gene expression.

Axons need to be properly guided to their targets to form synaptic connections, and this requires interactions between highly conserved extracellular and transmembrane ligands and their cell surface receptors. The majority of studies on axon guidance signaling pathways have focused on the role of these pathways in rearranging the local cytoskeleton and plasma membrane in growth cones and axons. However, a smaller body of work has demonstrated that axon guidance signaling pathways also control gene expression via local translation and transcription. Recent studies on axon guidance ligands and r

Ectoderm barcoding reveals neural and cochlear compartmentalization.

Placodes and the neural crest are defining features of vertebrates. In this study, we investigate their lineages in mice using in utero approaches. We demonstrated that nanoinjection at embryonic day 7.5 targeted the ectoderm, including the future nervous system, placodes, and neural crest, allowing highly efficient manipulation of the future nervous system and inner ear. By using heritable DNA barcodes and high-throughput next-generation single-cell lineage tracing, we elucidated convergent differentiation pathways and identified distinct nervous system-, neural crest-, and otic placode-deriv

Coxsackievirus A16 infection induces neural cell and non-neural cell apoptosis in vitro.

Coxsackievirus A16 (CA16) is one of the main causative pathogens of hand, foot and mouth disease (HFMD). Viral replication typically results in host cell apoptosis. Although CA16 infection has been reported to induce apoptosis in the human rhabdomyosarcoma (RD) cell line, it remains unclear whether CA16 induces apoptosis in diverse cell types, especially neural cells which have important clinical significance. In the current study, CA16 infection was found to induce similar apoptotic responses in both neural cells and non-neural cells in vitro, including nuclear fragmentation, DNA fragmentatio
