omega-Conotoxin GVIA and nifedipine inhibit the depolarizing action of the fungal metabolite, destruxin B on muscle from the tobacco budworm (Heliothis virescens).

Recent studies on a group of fungal metabolites, collectively called the destruxins, have suggested that these compounds activate calcium influx in insect skeletal muscle. In this study, we have investigated the sensitivity of destruxin B to the voltage-dependent calcium channel antagonists; omega-conotoxin GVIA, nifedipine, diltiazem and methoxyverapamil on skeletal muscle from the lepidopteran insect pest, tobacco budworm (Heliothis virescens). At a concentration of 1.7 microM, destruxin B caused a rapid decrease in the transmembrane resting potential. The effect of destruxin B on insect mus

[New aspects of anticonvulsive therapy in severe pre-eclampsia and eclampsia].

Parenterally administered magnesium is a reliable and effective treatment to prevent and control convulsions associated with preeclampsia and eclampsia. Because of several side effects in the newborn, especially in the premature child, benzodiazepines are not recommended for prolonged medication. Clomethiazole has also been used with good acceptance, but clinical experience is limited. The anticonvulsive property of magnesium is not clearly understood. Beside a well known peripheral action by neuromuscular blockade through very high serum concentrations, a central mechanism is postulated alrea

Between bodily changes and performance: some possible reasons for slowing with age.

A number of models are outlined which attempt to account for relationships between various bodily, and especially neural, changes and the widespread slowing of performance observed in old age. Muscular and neuromuscular changes can account for some types of slowing, but most types are attributable to central mechanisms. The accumulation of small increments of time in a long chain of cells from sensory input to motor output is shown to be an inadequate explanation. Correlations between reaction time, EEG alpha cycle-time and age are in line with the widespread tendency for increases of reaction

Comparative analysis of processed EEG indices and entropy-based metrics for assessing anesthetic depth: a scoping review - PRISMA-ScR.

BACKGROUND: Processed electroencephalography (EEG) monitors such as the bispectral index (BIS) and patient state index (PSI), are used clinically to estimate anesthetic depth, yet their algorithmic design obscures how closely these indices reflect underlying neural complexity. Entropy-based analyses, grounded in information theory, provide a quantitative framework for characterizing EEG signal irregularity and have been proposed as physiologically interpretable alternatives. However, the relationship between these commercial indices and theoretical entropy measures remains unclear. OBJECTIVES

Barriers and levers to using an app for motor function assessment by therapists in hospitals: A qualitative study of the MFM-Play app.

BACKGROUND: The Motor Function Measure (MFM) is an important evaluation tool used to monitor the progression of most neuromuscular diseases. However, the MFM is subject to inter-rater variability, and patients must produce their best effort, which can be difficult for children. In response to these issues, we developed a digital app named MFM-Play, designed to optimise completion of the MFM in a fun manner. The objective of this study was to explore barriers and levers to the MFM-Play's use by therapists. METHOD: We conducted a qualitative study with rehabilitation professionals who had teste

Death following high-dose AAV9 gene therapy in a patient with advanced SMA-PME.

Spinal muscular atrophy with progressive myoclonic epilepsy (SMA-PME) is an ultra-rare, fatal autosomal-recessive disorder caused by ASAH1 mutations, with no curative treatment. We report the first-in-human intravenous administration of an AAV9 vector carrying the human ASAH1 coding sequence in a 15-year-old female with advanced SMA-PME (heterozygous ASAH1 c.456A>C and c.918-2A>G mutations). The patient presented with severe neuromuscular impairment, deafness, and recurrent myoclonic status epilepticus, without detectable anti-AAV9 neutralizing antibodies. A marked clinical deterioration warra

The influence of menstrual cycle phase on neuromuscular performance and subjective perception of effort in elite football players.

This study aimed to investigate the potential fluctuations in neuromuscular performance and subjective perception of effort during three different phases of the menstrual cycle. Fifteen elite female football players (age: 23.47 ± 6.14) volunteered to participate in the study. Tympanic temperature, saliva hormone measurement, urine concentration of the luteinizing hormone and a calendar tracker were used to verify the following phases of the menstrual cycle: early follicular phase (EFP), late follicular phase (LFP), and mid-luteal phase (MLP). For each phase, mean concentric velocity and the ra

An examination into factors that impact hip and groin pain in ice hockey players.

Ice hockey is a demanding sport that requires high degrees of flexibility, strength, and neuromuscular control. Ice hockey players are at an elevated risk of hip and groin injuries. Thus, it is important that clinicians understand factors that may influence hip and groin pain in this population. The purpose of this study is to explore the factors of strength, pain, special tests, and symptoms of the hip and groin in male ice hockey players based on age and position. We utilized a cross-sectional study design to collect Copenhagen Hip and Groin Outcome Score (HAGOS), the Visual Analog Pain Scal

Startle sign events induced by mechanical manipulation during surgery for neuroma localization: a retrospective cohort study.

BACKGROUND: Chronic pain from peripheral neuromas is difficult to manage and often requires surgical excision, though intraoperative identification of neuromas can be challenging due to anatomical ambiguity. Mechanical manipulation of the neuroma during surgery can elicit a characteristic "startle sign", which can help guide surgical management. However, it is unknown how anesthetic management affects detection of the startle sign. METHODS: We performed a retrospective cohort study of 73 neuroma excision surgeries performed recently at Massachusetts General Hospital. Physiological changes in

Regenerative biomarkers for Duchenne muscular dystrophy.

Skeletal muscle has an extraordinary capacity to regenerate after injury and trauma. The muscle repair mechanism is a complex process orchestrated by multiple steps. In neuromuscular disorders such as Duchenne muscular dystrophy (DMD), the pathological consequences of the lack of dystrophin and the loss of the dystrophin-associated protein complex are dramatic, with a progressive cascade of events, such as continual influx of inflammation, repeated cycles of degeneration and impaired regeneration. Thus, muscle regeneration is a hallmark of the disease and careful monitoring of regenerative pro
