Improvements induced by retinal gene therapy with voretigene neparvovec depend on visual cortical hemispheric dominance mechanisms.

BACKGROUND: RPE65-associated retinal degeneration (RPE65-RD) causes severe visual deficits. Gene therapy with AAV2-hRPE65v2 is a breakthrough but it is currently unknown which visual pathways benefit from treatment and if cortical mechanisms can amplify retinal improvements. METHODS: In this within-subject design, ten patients with biallelic RPE65-RD underwent sub-retinal injection of AAV2-hRPE65v2. Psychophysical full-field stimulus threshold determination and functional magnetic resonance imaging were performed before and 12 months after treatment. Population receptive fields (pRF) were com

Medical management of musculoskeletal pain in professional wheelchair basketball players: an evidence-based guide.

Wheelchair basketball (WB) has evolved from a recreational activity to an internationally recognized sport. It is useful in enhancing psychophysical well-being in athletes with disabilities. However, WB professional players' medical management, especially regarding musculoskeletal pain (MSP), poses unique challenges and the related scientific literature still seems fragmentary. An extensive review of the PubMed, Cochrane, and Embase databases was performed to identify the available evidence regarding the medical management of WB players MSP. It emerges that WB players often face musculoskeleta

Assessment of two olfactory training methods for post-COVID-19 loss of olfaction: Classical and intensive.

OBJECTIVES: To determine whether intensive olfactory training provides better chances of recovery than classic protocols in persistent dysosmia after Covid-19. INTRODUCTION: In the literature, olfactory training holds pride of place in the management of post-infection olfactory disorder, with a classic 4-odor protocol. On the other hand, few studies have assessed the benefit of more intensive training. MATERIALS AND METHODS: This prospective randomized clinical trial (No. 2020-A01397-32) assessed olfactory training for persistent dysosmia due to COVID-19, with 5 weeks' to 12 months' progress

Can we tell where our bodies end and the external world begins? Evidence for precise three-dimensional internal body models.

Distinguishing our body from the external world is crucial for self-perception and environmental interaction. Yet, the accuracy with which we perceive this boundary remains underexplored. Here, we developed a psychophysical protocol to assess how accurately individuals perceive their body boundaries. Participants were asked whether the midpoint between two tactile stimuli was inside or outside their perceived body boundary. Three-dimensional scans provided objective anatomical boundaries, allowing psychometric functions to be fitted. Results revealed remarkable overall precision, often within

High-Resolution Models of Human Cochlea for a Study of Neural Activation.

With more than one million implanted devices, the cochlear implant (CI) is the most successful neuroprosthesis and one of the most successful achievements of medical technology in general. While CI users usually regain the ability to understand and use spoken language with or without visual aids, CI performance is widely variable after implantation. After reconstructing anatomically realistic cochlea models from high-resolution X-ray microtomography scans of eight human temporal bone specimens contrast-enhanced with osmium tetroxide, we conducted an in-depth analysis of the stimulus amplitudes

Unveiling the dark side of eating disorders: evidence on the role of dark triad and body uneasiness in youth.

Eating disorders comprise an array of mental disturbance with profound implications for individuals' psychophysical and societal well-being. Extensive research has elucidated the role of the Big Five personality traits in explaining individual differences in the risk of eating disorders, overshadowing alternative personality taxonomies, such as the Dark Triad - DT (i.e., Machiavellianism, psychopathy, and narcissism). Accordingly, the current study aimed to address the association between DT and the risk of eating disorders, also exploring the potential involvement of body uneasiness as captur

Measuring the visual angle of polarization-related entoptic phenomena using structured light.

The ability to perceive polarization-related entoptic phenomena arises from the dichroism of macular pigments held in Henle's fiber layer of the retina and can be inhibited by retinal diseases, such as age-related macular degeneration, which alters the structure of the macula. Structured light tools enable the direct probing of macular pigment density and retinal structure through the perception of polarization-dependent entoptic patterns. Here, we directly measure the visual angle of an entoptic pattern created through the illumination of the retina with a structured state of light and a perc

Biological relevance and methodological implications of unexpected hearing thresholds in a diving bird.

Many animals alternate between different media, such as air and water, thanks to specific adaptations. Among birds, penguins (Sphenisciformes) have the most extreme morphological, physiological, and behavioural adaptations to their amphibious lifestyle. Their auditory perception of sound, potentially matching different impedances in air and under water, is largely unknown particularly in terms of whether their underwater adaptations may have affected their in-air hearing capacity. In this context, we investigated the hearing ability of four captive Humboldt penguins (Spheniscus humboldti) in a

The body interior in anorexia nervosa: from interoception to conceptual representation of body interior.

BACKGROUND: Body image disorders are well documented in anorexia nervosa (AN); however, knowledge of interoceptive awareness (IA) in this population remains poor. This descriptive study investigated whether and how the representation of the interior of the body may have an impact on IA. METHODS: The representations and knowledge of the body interior were evaluated with a drawing task in 34 women with AN and 34 healthy controls (HCs). A lexicometric analysis was performed on the vocabulary used to describe the drawn body parts in a structured interview. It was assumed that the conceptual repre

Image-dependence of the detectability of optogenetic stimulation in macaque inferotemporal cortex.

Artificial activation of neurons in early visual areas induces perception of simple visual flashes.1,2 Accordingly, stimulation in high-level visual cortices is expected to induce perception of complex features.3,4 However, results from studies in human patients challenge this expectation. Stimulation rarely induces any detectable visual event, and never a complex one, in human subjects with closed eyes.2 Stimulation of the face-selective cortex in a human patient led to remarkable hallucinations only while the subject was looking at faces.5 In contrast, stimulations of color- and face-selecti

Harnessing neuroplasticity.

Twenty years ago, cognitive impairments were recognized as an unmet treatment need in schizophrenia. Basic science discoveries in neuroplasticity had led to cognitive training approaches for dyslexia. We wondered whether a similar approach could target working memory deficits in schizophrenia by harnessing plasticity in the auditory cortex. Our per protocol experimental therapeutics studies tested the hypothesis that sharpening auditory cortical representations would result in better verbal learning and memory. We also later studied the effects of intensive training of basic social cognitive o

Attention to visual motion suppresses neuronal and behavioral sensitivity in nearby feature space.

BACKGROUND: Feature-based attention prioritizes the processing of the attended feature while strongly suppressing the processing of nearby ones. This creates a non-linearity or "attentional suppressive surround" predicted by the Selective Tuning model of visual attention. However, previously reported effects of feature-based attention on neuronal responses are linear, e.g., feature-similarity gain. Here, we investigated this apparent contradiction by neurophysiological and psychophysical approaches. RESULTS: Responses of motion direction-selective neurons in area MT/MST of monkeys were record
