G-quadruplex (G4) DNA, an alternate structure formed by Hoogsteen hydrogen bonds between guanines in G-rich sequences, threatens genomic stability by perturbing normal DNA transactions including replication, repair, and transcription. A variety of G4 topologies (intra- and intermolecular) can form in vitro, but the molecular architecture and cellular factors influencing G4 landscape in vivo are not clear. Helicases that unwind structured DNA molecules are emerging as an important class of G4-resolving enzymes. The BRCA1-associated FANCJ helicase is among those helicases able to unwind G4 DNA i

AIMS: The traditional approach for the management of benign laryngeal lesions is transoral microsurgery. In cases of anatomic particularities, such as insufficient cervical extension, short mandible, temporomandibular joint ankylosis etc., and contraindications for general anesthesia, transoral microsurgery is not possible. In such cases transnasal flexible endoscopic surgery under local anesthesia can be a relevant alternative. The purpose of the study was to assess the possibilities of flexible endoscopic management of benign nonepithelial lesions of the larynx.
MATERIALS AND METHODS: Flexi

During the eukaryotic cell cycle, genetic material must be accurately duplicated and faithfully segregated to each daughter cell. Segregation of chromosomes is dependent on the centromere, a region of the chromosome which interacts with mitotic spindle microtubules during cell division. Centromere function in the budding yeast, Saccharomyces cerevisiae, can be regulated by placing an inducible promotor adjacent to centromere DNA. This conditional centromere can be integrated into chromosome III to generate a conditionally functional dicentric chromosome. Activation of the dicentric chromosome

We have isolated and characterized a plasmid (pChug 20.1) that contains the cDNA of a nuclear uracil DNA glycosylase (UDG) gene isolated from normal human placenta. This cDNA directed the synthesis of a fusion protein (Mr 66,000) that exhibited UDG activity. The enzymatic activity was specific for a uracil-containing polynucleotide substrate and was inhibited by a glycosylase antibody or a beta-galactosidase antibody. Sequence analysis demonstrated an open reading frame that encoded a protein of 335 amino acids of calculated Mr 36,050 and pI 8.7, corresponding to the Mr 37,000 and pI 8.1 of pu

BACKGROUND: Two PARP inhibitors (PARPis), olaparib and talazoparib, have been approved in combination with androgen receptor pathway inhibitors (ARPIs) for metastatic castration-resistant prostate cancer (mCRPC) in Europe, regardless of homologous-recombination repair (HRR) status. However, the mechanism in HRR-negative patients remains unclear.
METHODS: We assessed PARP inhibitors (PARPis; olaparib, talazoparib) alone and with ARPIs (abiraterone, enzalutamide, apalutamide) in prostate cancer cell lines, metastatic patient-derived organoids (mPDOs), and LN-metastatic tissue slice cultures. HR

Exposure to ionizing radiation can occur during medical treatments, from naturally occurring sources in the environment, or as the result of a nuclear accident or thermonuclear war. The severity of cellular damage from ionizing radiation exposure is dependent upon a number of factors including the absorbed radiation dose of the exposure (energy absorbed per unit mass of the exposure), dose rate, area and volume of tissue exposed, type of radiation (e.g., X-rays, high-energy gamma rays, protons, or neutrons) and linear energy transfer. While the dose, the dose rate, and dose distribution in tis

BACKGROUND & AIMS: Targeting the transforming growth factor-β (TGF-β) pathway to reverse the immunologically "cold" tumor microenvironment of pancreatic ductal adenocarcinoma (PDAC) remains clinically unsuccessful, warranting novel therapeutic strategies.
METHODS: We performed multiplex immunohistochemistry on human PDAC samples to correlate cell-type-specific TGF-β pathway activation and CD8+ T-cell abundance and developed a tumor and T-cell coculture to interrogate the TGF-β pathways that promote T-cell-mediated cytotoxicity. We employed newly generated genetically engineered mouse models a

Ovarian cancer is a highly heterogeneous malignancy and remains a major cause of cancer-related mortality, mainly due to late diagnosis and frequent development of therapy resistance. Current diagnostic markers show limited sensitivity for early-stage disease, highlighting the need for novel biomarkers and improved understanding of tumor biology. Epigenetic deregulation is a hallmark of ovarian cancer, characterized by global DNA hypomethylation and focal hypermethylation of tumor suppressor genes. Among hypomethylated genomic regions, long interspersed nuclear elements-1 (LINE-1) represent a

Cisplatin and its analogues are a cornerstone of cancer chemotherapy; however, their therapeutic potential is limited by severe side effects and the development of drug resistance. To address these issues, nanocarriers and/or platinum prodrugs are being evaluated. Previous in vitro studies showed that ultra-small (< 5 nm) iron oxide nanoparticles coated with tartaric and adipic acids (FeAT-NPs) are an efficient delivery system for the cisplatin prodrug cis-diamminetetrachloroplatinum(IV) (cisplatin(IV)). In this study, the in vivo behavior of cisplatin (IV) conjugated with the FeAT-NPs (FeAT-N

OBJECTIVE: Homologous recombination (HR) comprises series of interrelated pathways that repair double-stranded DNA breaks and inter-strand crosslinks. It provides support for DNA replication to recover stalled or broken replication forks. Compared with homologous recombination proficiency (HRP), cancers with homologous recombination deficiency (HRD) are more likely to undergo cell death when treated with DNA-damaging agents, such as platinum agents, and have better disease control.
METHODS: Patients diagnosed with stage III/IV ovarian cancer, early stages with recurrence, who received adjuvan

Xeroderma pigmentosum group C (XPC) protein is a sensor of helix-distorting DNA lesions, the function of which is to trigger the global genome repair (GGR) pathway. Previous studies demonstrated that XPC protein operates by detecting the single-stranded character of non-hydrogen-bonded bases opposing lesion sites. This mode of action is supported by structural analyses of the yeast Rad4 homologue that identified critical side chains making close contacts with a pair of extrahelical nucleotides. Here, alanine substitutions of the respective conserved residues (N754, F756, F797, F799) in human X

The multifunctional protein kinase CK2 is involved in several aspects of the DNA damage response (DDR) in mammals. To gain insight into the role of CK2 in plant genome maintenance, we studied the response to genotoxic agents of an Arabidopsis CK2 dominant-negative mutant (CK2mut plants). CK2mut plants were hypersensitive to a wide range of genotoxins that produce a variety of DNA lesions. However, they were able to activate the DDR after exposure to γ irradiation, as shown by accumulation of phosphorylated histone H2AX and up-regulation of sets of radio-modulated genes. Moreover, functional as

This critical and selective review synthesizes the accumulating body of biological evidence supporting a process we term epigenetic-genetic coupling as a mechanistic basis for Lamarckian inheritance of somatically acquired adaptations. We propose that evolutionary processes in mammals and higher vertebrates can involve deaminase-driven, reverse transcriptase-mediated, RNA-templated targeted homologous recombination. We contrast well-established examples of "Soft", reversible epigenetic inheritance with historical and contemporary evidence suggestive of stable, DNA-integrated "Hard" Lamarckian

BACKGROUND: Spermatozoa have the task of delivering an intact paternal genome to the oocyte and supporting successful embryo development. The detection of sperm DNA fragmentation (SDF) has been emerging as a complementary test to conventional semen analysis for male infertility evaluation, but the mechanism leading to SDF and its impact on assisted reproduction remain unclear. Therefore, the study identified and analyzed the differentially expressed proteins of sperm with high and low SDF.
METHODS: Semen samples from men attended the infertility clinic during June 2020 and August 2020 were an

Thirteen years ago, we pointed out that ovarian transitional cell carcinomas (TCCs) and conventional high-grade serous carcinomas (HGSCs) had similar genetic alterations and clinical behavior. Consequently, ovarian TCC is now classified as a morphologic variant of HGSC. Defective homologous recombination, resulting from genetic or epigenetic inactivation of DNA damage repair genes, such as BRCA1/2, occurs in approximately 50% of the HGSCs. Although BRCA mutations have been associated with HGSCs with solid, pseudoendometrioid or transitional (SET) features, little is known about the role of non

PURPOSE: DNA polymerase theta (Polθ) is the central component of DNA double-strand break repair through microhomology-mediated end joining (MMEJ). Targeting Polθ for cancer therapy has been proposed due to its tumor-selective expression, as well as its role in the survival of homologous recombination-deficient (HRD) cancer cells. Additionally, inhibition of Polθ has been shown to potentiate the effects of DNA damage, such as that caused by poly (ADP-ribose) polymerase (PARP) inhibition. In this study, we present the pharmacologic profile of ART6043, a potent, selective, allosteric, small-molec

RecQ helicases are ubiquitous genome maintenance proteins that can resolve complex DNA structures during replication, recombination, and repair. Eukaryotic RecQ helicases are known to interact with replication protein A (RPA), leading to the stimulation of their unwinding activity. In higher eukaryotes, this interaction is mediated through the acidic residues of RecQ helicases and the N-terminal OB fold (DBD-F) of RPA70. In kinetoplastid parasites, the RPA70 homolog lacks this N-terminal DBD-F domain, raising the possibility of alternative interaction mechanisms. Here, we investigate the physi

It has been shown that the pathogenic variants (PVs) of the DNA Damage Response (DDR) genes, whether of a germinal or somatic nature, represent a predictive biomarker of high sensitivity to treatment with inhibitors of the enzyme poly-ADP-ribose polymerase (PARP) in patients with hormone-resistant metastatic prostate cancer (HRPCa). Moreover, the detection of PVs of the Homologous Recombination Repair (HRR) genes in PCa patients can help to define the patient's prognosis and the choice of the therapeutic procedure. Among men with metastatic PCa, the frequency of PVs in HRR genes ranges from 11

FENs (flap endonucleases) and related FEN-like enzymes [EXO-1 (exonuclease-1), GEN-1 (gap endonuclease 1) and XPG (xeroderma pigmentosum complementation group G)] are a family of bivalent-metal-ion-dependent nucleases that catalyse structure-specific hydrolysis of DNA duplex-containing nucleic acid structures during DNA replication, repair and recombination. In the case of FENs, the ability to catalyse reactions on a variety of substrates has been rationalized as a result of combined functional and structural studies. Analyses of FENs also exemplify controversies regarding the two-metal-ion me
