Discrimination between nanocurcumin and free curcumin using graphene quantum dots as a selective fluorescence probe.

Accurate-controlled sized graphene quantum dots (GQDs) have been used as an analytical nanoprobe for detecting curcumin as a function of the photoluminescent quenching upon increasing concentrations of the analyte. Regarding the importance of curcumin nanoparticles in nutraceutical food, the analytical method described herein was also proven for the discrimination of curcumin remaining in free solution from that encapsulated into water-soluble nanomicelles of ca. 11 nm. This recognition is based on the displacement of GQD emission when interacting with both curcumin species. Maximum emission w

PVP-templated highly luminescent copper nanoclusters for sensing trinitrophenol and living cell imaging.

Copper nanoclusters (CuNCs) exhibit susceptibility to oxidation in the subnanometer size range. In this work, a facile and green protocol is reported for the successful synthesis of water soluble CuNCs, with poly(vinylpyrrolidone) as a template and ascorbic acid as a mild reducing agent. The as-prepared CuNCs exhibit a green fluorescence and high quantum yield (QY = 44.67%) in water, which is the highest among the reported water soluble CuNCs. The origin of their highly luminescent nature was also investigated. In addition, the obtained CuNCs show good tolerability to high ionic strength, supe

A new lysosome-targetable fluorescent probe with a large Stokes shift for detection of endogenous hydrogen polysulfides in living cells.

Hydrogen polysulfides (H2Sn, n > 1) influence a variety of important biological functions and activities associated with hydrogen sulfide (H2S). The development of probes for rapid, selective, and sensitive detection of H2Sn still remains a great challenge. Lysosome plays crucial roles in various physiological processes among living cells, which arouse high interest in detecting endogenous lysosome-targetable H2Sn. To the best of our knowledge, there is no lysosome-targetable probe for H2Sn has been reported. In this work, a new lysosome-targetable probe NIPYNF, based on the scaffold of imidaz

A rapid and sensitive fluorescent probe for detecting hydrogen polysulfides in living cells and zebra fish.

Hydrogen polysulfides (H2Sn, n>1) plays crucial roles in many biological processes, while it remains a challenge for rapid and selective detection of H2Sn. We designed and synthesized a turn-on fluorescent probe (JCCF) for detecting H2Sn based on a new julolidine-coumarinocoumarin scaffold. H2Sn could trigger a dramatic fluorescence enhancement (52-fold) with a fast response time and a low detection limit of 98.3 nM (S/N = 3). Moreover, JCCF was successfully applied to image H2Sn in living cells and zebra fish with low cytotoxicity.

A fluorescent aminosugar to rapidly screen and study RNA binders.

RNA targeted high-throughput assays that allow for rapid detection of high affinity binding ligands are important in RNA recognition studies. A number for fluorescent dyes have been reported that can assist in rapidly identifying nucleic acid (RNA) binding elements without the need for immobilization of RNA or the ligand. A number of these dyes are planar aromatic molecules that bind non-specifically to nucleic acids and often distort their parent nucleic acid structures leading to ambiguity in the interpretation of results. In this light, we report here, the use of an aminoglycoside (neomycin

A ratiometric multicolor fluorescence biosensor for visual detection of alkaline phosphatase activity via a smartphone.

Herein we designed a selective and smartphone-based strategy for visual detection of alkaline phosphatase (ALP) by utilizing the property of amino-functionalized copper (II)-based metal-organic frameworks (NH2-Cu-MOFs) with oxidase mimic property and fluorescence property. Surprisingly, the oxidase mimic property of NH2-Cu-MOFs can work well at a high pH value 8.0. Thus, a cascade reaction between ALP and NH2-Cu-MOFs was realized for the construction of a ratiometric multicolor sensing platform through the controllable catalytic activity of NH2-Cu-MOFs by pyrophosphate (PPi) and ALP. The catal

Full-Range pH Stable Au-Clusters in Nanogel for Confinement-Enhanced Emission and Improved Sulfide Sensing in Living Cells.

The sensitive and selective detection of hydrogen sulfide is of great importance due to its crucial role in pathological and physiological processes. Herein, we report a novel fluorescent platform, AuNCs@GC, for selective detection of hydrogen sulfide in living cells by impregnating the Au nanoclusters (AuNCs) into a biocompatible cationic polymer matrix, glycol-chitosan (GC) nanogel. The confinement effect significantly increased the emissive Au(I) units, resulting in a 6-fold enhancement of quantum yield (from 6.38% to 36.42%). In addition, the prepared positively charged AuNCs@GC nanogel ex

Fluorescent Probe DCVJ Shows High Sensitivity for Characterization of Amyloid β-Peptide Early in the Lag Phase.

The aggregation of intrinsically disordered and misfolded proteins in the form of oligomers and fibrils plays a crucial role in a number of neurological and neurodegenerative diseases. Currently, most probes and biophysical techniques that detect and characterize fibrils at high resolution fail to show sensitivity and binding for oligomers. Here, we show that 9-(dicyano-vinyl)julolidine (DCVJ), a class of molecular rotor, binds amyloid beta (Aβ) early aggregates, and we report the kinetics as well as packing of the oligomer formation. The binding of DCVJ to Aβ40 increased its emission intensit

Mono and dihydroxy coumarin derivatives: Copper chelation and reduction ability.

UNLABELLED: Due to the limited array of the currently available copper chelators, research of such compounds continues to be of clinical interest. Notably, o-dihydroxycoumarins have been previously shown to be potent iron chelators under neutral conditions. Within this study, the interaction of a series of natural coumarins and their synthetic analogs with copper has been evaluated in order to obtain structure-activity relationships under different pathophysiological pH conditions. Both competitive and non-competitive methods have been employed. Analysis of cupric ion reduction has also been p

Switch-on fluorescent strategy based on crystal violet-functionalized CdTe quantum dots for detecting L-cysteine and glutathione in water and urine.

The concentration of L-cysteine (Cys) and glutathione (GSH) is closely related to the critical risk of various diseases. In our study, a new rapid method for the determination of Cys and GSH in water and urine samples has been developed using a fluorescent probe technique, which was based on crystal violet (CV)-functionalized CdTe quantum dots (QDs). The original QDs emitted fluorescence light, which was turned off upon adding CV. This conjugation of CV and QDs could be attributed to electrostatic interaction between COO- of mercaptopropionic acid (MPA) on the surface of QDs and N+ of CV in aq

A Fluorescent Switch Sensor for Glutathione Detection Based on Mn-doped CdTe Quantum Dots - Methyl Viologen Nanohybrids.

In the work, a fluorescence switch sensor consists of Mn-doped CdTe quantum dots (QDs) - methyl viologen (MV(2+)) nanohybrid is fabricated. In the sensor, MV(2+) plays a role in turning the QDs fluorescence to the "OFF" state due to the efficient electron transfer process while glutathione (GSH) could turn "ON" the native QDs fluorescence by effectively releasing QDs from the QDs-MV(2+) nanohybrids. In addition, the recovery level of QDs fluorescence is closely related to the amount of GSH. Based on this phenomenon, a reliable and convenient GSH quantitative determination method is established

An ascorbic acid sensor based on cadmium sulphide quantum dots.

We present a Förster resonance energy transfer (FRET)-based fluorescence detection of vitamin C [ascorbic acid (AA)] using cadmium sulphide quantum dots (CdS QDs) and diphenylcarbazide (DPC). Initially, DPC was converted to diphenylcarbadiazone (DPCD) in the presence of CdS QDs to form QD-DPCD. This enabled excited-state energy transfer from the QDs to DPCD, which led to the fluorescence quenching of QDs. The QD-DPCD solution was used as the sensor solution. In the presence of AA, DPCD was converted back to DPC, resulting in the fluorescence recovery of CdS QDs. This fluorescence recovery can

Fluorescent "turn-on" detecting CN(-) by nucleophilic addition induced Schiff-base hydrolysis.

A new chemosensor Sz based on Schiff-base group as recognition site and naphthalene as the fluorescence signal group was designed and synthesised. It could fluorescent "turn-on" detect cyanide (CN(-)) via a novel mechanism of nucleophilic addition induced Schiff-base hydrolysis. Adding the CN(-) into the solution of Sz could induce Sz to emit blue fluorescence at 435 nm instantly. Moreover, Sz could also colorimetric detect CN(-). Upon the addition of CN(-), the Sz showed dramatic color change from yellow to colorless. These sensing procedures could not be interfered by other coexistent compet
