Enzymatically Crosslinked Hybrid Hydrogels from Ultrasound-Decellularized Spinal Cord Meninges and Hydrophilic Silk Fibroin for Neural Tissue Engineering.

Hydrogels combining the biochemical complexity of the native extracellular matrix (ECM) with the tunable properties of protein-based biomaterials are promising for neural tissue engineering. In this study, decellularized spinal cord meninges (dSCM) were combined with water-soluble hydrophilic silk fibroin (hSF) and enzymatically crosslinked using a horseradish peroxidase/H2O2 system to develop composite hydrogels. A detergent-free, sonication-assisted decellularization method effectively removed cellular components while preserving matrix integrity, reducing residual double-stranded DNA to bel…

Structural and mechanical insights into the extracellular matrix of the aging human lacrimal gland for tissue engineering applications.

Aqueous-deficient dry eye disease (ADDE) frequently results from structural and functional impairment of the lacrimal gland. Regenerative approaches require biomaterials that replicate the extracellular matrix (ECM) composition and biomechanical properties of native lacrimal gland tissue. However, quantitative data on the ECM composition and large-strain biomechanics of the human lacrimal gland remain limited. In this study, histological and immunofluorescence-based ECM analyses were combined with large-strain mechanical testing and quantitative enzyme-linked immunosorbent assay (ELISA)-based

Advancements in Spinal Cord Injury Treatment: Integrating Drug Delivery, Biophysical Stimulation, Cell-Based Therapies, and Tissue Engineering Approaches.

Spinal cord injuries, whether resulting from traumatic or nontraumatic events, have severe and lasting detrimental effects on individuals, significantly impacting their overall health, mobility, and quality of life. The limited regenerative capacity of the spinal cord is mainly due to neuronal damage, the presence of inhibitory molecules, an impaired immune response, and the formation of glial scars, all of which create a hostile environment for neural repair and functional recovery. The majority of SCIs are caused by traffic accidents and falling objects. The current global treatments used fo

A lubcan cross-linked polyethylene glycol dimethyl ether hydrogel for hyaluronic acid replacement as soft tissue engineering fillers.

The structure of soft tissues is often destroyed by injury and aging. Injectable fillers eliminate the need for surgery and enhance repair. Hyaluronic acid-based hydrogels are commonly employed for their effectiveness and biocompatibility. However, hyaluronidase breaks them down quickly. Lubcan, a naturally sourced microbial extracellular polysaccharide, has demonstrated significant water absorption and retention capabilities, as well as lubricating properties comparable to those of hyaluronic acid. In this study, a novel injectable and implantable hydrogel was created from lubcan by adding po

Fabrication and characterization of 3D printed agarose/poly(ethylene glycol) diacrylate/hydroxyapatite nanocomposite hydrogel for cartilage tissue engineering.

Damage to the articular cartilage can be caused by osteoarthritis (OA), exercise, cancer, and etc. A major challenge is cartilage's limited ability to self-repair after injury. In this study, the new cartilage scaffold was prepared by two biocompatible materials, agarose and poly(ethylene glycol) diacrylate (PEGDA), using a 3D printer. In this work used Taguchi-based statistical optimization to determine the optimal agarose concentration for 3D scaffolds. A key innovation was the incorporation of nanohydroxyapatite (nHA), which improved the hydrogel's viscosity and shear-thinning behavior, pri

Fabrication and Characterization of a Porous TiO2-Modified PEEK Scaffold with Enhanced Flexural Compliance for Bone Tissue Engineering.

Bone pathologies are becoming increasingly prevalent with an aging population, often necessitating bone grafting procedures. The current gold standard for grafting uses autologous tissue; however, this approach carries limitations such as donor site morbidity. Consequently, there is a growing interest in alternative biomaterials. Polyetheretherketone (PEEK), a thermoplastic with bone-like mechanical properties, has shown promise, although its limited bioactivity remains a critical constraint. Various functionalization strategies have been employed to enhance the biological performance of other

Preclinical Assessment in Juvenile Sheep of an Allogeneic Bone Tissue Engineering Product with Wharton's Jelly Mesenchymal Stromal Cells.

Secondary osteonecrosis (ON) is a common complication in paediatric cancer survivors. Combining multipotent mesenchymal stromal cells (MSCs) with core decompression surgery halts disease progression and stimulates bone regeneration. However, the success of advanced therapy medicinal products (ATMPs) requires versatile "off-the-shelf" tissue engineering products (TEPs). This study evaluated the safety and efficacy of TEPs loaded with allogeneic MSCs from Wharton's jelly (WJ-MSCs) in a large-animal model of bone regeneration to support a paediatric investigational plan for ON patients. Alveolar Bone Tissue Engineering Using Deciduous Dental Pulp Stem Cells in a Patient with Cleft Lip and Palate.

The use of tissue bioengineering strategies in dentistry has gained relevance. Many studies indicate that stem cells associated with biomaterials can regenerate intraoral tissues and have been applied to patients with cleft lip and palate (CLP). One of the treatments is alveolar bone reconstruction through bone grafts, where the bone is removed from the donor site and placed in the alveolar cleft. The use of stem cells from deciduous dental pulp, associated with a hydroxyapatite and collagen scaffold, can eliminate the need for autologous bone grafts, reducing pain and morbidity at the donor s

A Multifunctional Tissue-Engineering Hydrogel Aimed to Regulate Bacterial Ferroptosis-Like Death and Overcoming Infection Toward Bone Remodeling.

Infection is the most common complication after orthopedic surgery and can result in prolonged ailments such as chronic wounds, enlarged bone defects, and osteomyelitis. Iron, which is essential for bacterial metabolism and immune cell functions, is extremely important. Bacteria harness iron from nearby cells to promote biofilm formation, ensuring their survival. Iron deficiency within the infection microenvironment (IME) consequently hampers macrophage function, enabling further dissemination of the infection and hindering macrophage polarization to the M2 phenotype. Therefore, a novel approa
