Evaluation of a newly developed direct agglutination test (DAT) for serodiagnosis and sero-epidemiological studies of visceral leishmaniasis: comparison with IFAT and ELISA.

A newly developed direct agglutination test (DAT) for visceral leishmaniasis, IFAT and ELISA were applied to sera of patients with visceral leishmaniasis, African and American trypanosomiasis, other parasitic infections and healthy controls. The sensitivities of the 3 tests were comparable (96.3% to 100%); excluding patients with African and American trypanosomiasis, the specificities of DAT and IFAT were 100% and ELISA 87.3%. When trypanosomiasis sera were included, the specificities were 72.6%, 94.3% and 79.4% in DAT, IFAT and ELISA respectively. In 273 sera from a leishmaniasis endemic area…

Isolation of human serum resistant Trypanozoon from cattle in Nigeria.

Four trypanosome stocks of the subgenus Trypanozoon were isolated from a herd of cattle in the Jos Plateau of Nigerian during a study on the epizootiology of trypanosomiasis. One of the stocks was repeatedly highly resistant to normal human serum (Blood Incubation Infectivity Test), indicating potential human infectivity. These observations suggest that the domestic cattle might act as reservoir hosts for the causative agent of human trypanosomiasis.

The situation of sleeping sickness in Angola: a calamity.

Although nearly one-fifth of the Angolan population is at risk of becoming infected with trypanosomiasis, only 6% currently have access to surveillance and treatment because of the war and its resultant destruction of the country's infrastructure. The paper outlines the history of human African trypanosomiasis (HAT) control activities in Angola and sums up what measures need to be taken to re-establish them.

Novel compounds to combat trypanosomatid infections: a medicinal chemical perspective.

INTRODUCTION: The current therapeutic arsenal against the kinetoplastids Trypanosoma brucei, Trypanosoma cruzi and Leishmania spp. is clearly inadequate and underscores the urgent need to develop new effective, safe and cost-effective drugs. AREAS COVERED: Accordingly, this review of patented products and processes using anti-kinetoplastid agents provides insight into the identification of novel or more refined drugs. In this review, we describe products developed in recent years for the treatment of human African trypanosomiasis, American trypanosomiasis and leishmaniasis from a medicinal ch…

A role for hepcidin in the anemia caused by Trypanosoma brucei infection.

Trypanosomiasis is a parasitic disease affecting both humans and animals in the form of Human African Trypanosomiasis and Nagana disease, respectively. Anemia is one of the most common symptoms of trypanosomiasis, and if left unchecked can cause severe complications and even death. Several factors have been associated with the development of this anemia, including dysregulation of iron homeostasis, but little is known about the molecular mechanisms involved. Here, using murine models, we study the involvement of hepcidin, the key regulator of iron metabolism and an important player in the deve…

Conventional therapy and promising plant-derived compounds against trypanosomatid parasites.

Leishmaniasis and trypanosomiasis are two neglected and potentially lethal diseases that affect mostly the poor and marginal populations of developing countries around the world and consequently have an important impact on public health. Clinical manifestations such as cutaneous, mucocutaneous, and visceral disorders are the most frequent forms of leishmaniasis, a group of diseases caused by several Leishmania spp. American trypanosomiasis, or Chagas disease, is caused by Trypanosoma cruzi, a parasite that causes progressive damage to different organs, particularly the heart, esophagus, and lo…

Sensitivity and specificity of HAT Bioline Abbott® 2.0 and HAT Sero K-Set CORIS® 2.0 for human African trypanosomiasis in the Kasai-Oriental province, Democratic Republic of the Congo: a retrospective diagnostic accuracy study.

BACKGROUND: Human African trypanosomiasis (HAT) due to Trypanosoma brucei gambiense has declined substantially in recent years, with many endemic areas now reporting very low prevalence levels as countries move toward elimination. In the Democratic Republic of the Congo (DRC), rapid diagnostic tests (RDTs) are increasingly used in surveillance and elimination programs; however, their performance in low-prevalence settings remains critical. This study evaluated and compared the diagnostic accuracy of two second-generation RDTs; HAT Bioline Abbott® 2.0 and HAT Sero K-Set CORIS® 2.0; using trypan…

[Age group distribution of Glossina palpalis palpalis females in plantations and shallow water of the forest region of Côte d'Ivoire. Relation to the prevalence of sleeping sickness].

In epidemiologically dangerous biotopes of foci with high prevalence of Human African Trypanosomiasis (HAT), different female age groups (nulliparous, young parous, old parous) were observed in the same proportions. On the contrary, in areas without HAT or in low prevalence foci (< 0.2%), these proportions significantly differed. Female age group distribution in epidemiologically dangerous biotopes could thus be a good indication by which to guide the control of Human African Trypanosomiasis (HAT).

Methods Applied to the Diagnosis of Cattle Trypanosoma vivax Infection: An Overview of the Current State of the Art.

Bovine trypanosomiasis caused by Trypanosoma vivax is a relevant disease in domestic ungulates in Latin America, causing different types of livestock losses, particularly in African and South American countries, leading to loss of millions of dollars/year related to dairy and meat production. In addition, T. vivax trypanosomiasis requires intensive veterinary care. While vector control is a feasible measure to manage disease spreading, the search for accurate diagnostic tools still represents a gap in routine veterinary practices and a challenge for the scientific community. The parasite is me…

Inducible nitric oxide synthase and nitrotyrosine in the central nervous system of mice chronically infected with Trypanosoma brucei brucei.

Human African trypanosomiasis, or sleeping sickness, evolves toward a meningoencephalitic stage, with a breakage in the blood-brain barrier, perivascular infiltrates, and astrocytosis. The involvement of nitric oxide (NO) has been evoked in the pathogenic development of the illness, since NO was found to be increased in the brain of animals infected with Trypanosoma brucei (T. b.) brucei. An excessive NO production can lead to alterations of neuronal signaling and to cell damage through the cytotoxicity of NO and its derivatives, especially peroxynitrites. In African trypanosomiasis, the sites…

Nitric oxide production in vervet monkeys (Cercopithecus aethiops) infected with Trypanosoma brucei.

A retrospective study of nitrate concentration in serum and cerebrospinal fluid (CSF) from vervet monkeys (Cercopithecus aethiops) infected with Trypanosoma brucei was undertaken. Serum nitrate was significantly elevated in parasitaemic animals. CSF nitrate detection correlated with the presence of parasites in the CNS. The results provide evidence for the production of nitric oxide (NO) in response to infection in a primate model of human African trypanosomiasis and provide the basis for the use of such a model in studies of the immunopathological effects of NO in human trypanosomiasis.

N-myristoyltransferase inhibitors as new leads to treat sleeping sickness.

African sleeping sickness or human African trypanosomiasis, caused by Trypanosoma brucei spp., is responsible for approximately 30,000 deaths each year. Available treatments for this disease are poor, with unacceptable efficacy and safety profiles, particularly in the late stage of the disease when the parasite has infected the central nervous system. Here we report the validation of a molecular target and the discovery of associated lead compounds with the potential to address this lack of suitable treatments. Inhibition of this target-T. brucei N-myristoyltransferase-leads to rapid killing o…

Plasma luteinizing hormone levels in response to gonadotropin-releasing hormone agonist and clonidine in Trypanosoma congolense-infected female goats.

Trypanosomiasis, a parasitic disease of humans and animals, occurs over a wide area of Africa and imposes a large socioeconomic burden on the people. In the present study, we investigated whether trypanosomiasis-induced reproductive disorders were due to pituitary or hypothalamic dysfunction by determining plasma luteinizing hormone (LH) response to gonadotropin-releasing hormone (GnRH) agonist or clonidine in Trypanosoma congolense-infected female goats. With GnRH agonist administration, the total amount of LH secreted over a 140-min sampling period on day 23 and day 60 postinfection was cons…

A traveler with neurobrucellosis.

An Indian traveler developed fever and neurological symptoms after a visit to East Africa. He was treated with suramin, melarsoprol and prednisolone for presumed East African trypanosomiasis. His condition deteriorated and cerebral lesions developed. Neurobrucellosis was diagnosed. Combination antibiotic therapy led to gradual clinical improvement and regression of the brain lesions. Misdiagnosis of East African trypanosomiasis followed by treatment with potentially lethal medication should be avoided by not relying on insufficient evidence during the diagnostic process.

Control of Trypanosoma evansi infection is IgM mediated and does not require a type I inflammatory response.

Very recent reports have documented that Trypanosoma evansi, the etiological agent of the livestock disease "surra," can cause human trypanosomiasis. In contrast to trypanosomes causing human African trypanosomiasis, T. evansi has a wide geographic distribution and host range, yet information about the immunobiological aspects of T. evansi trypanosomiasis is limited. Here, we show that, although T. evansi causes the induction of tumor necrosis factor (TNF), interferon-gamma, and nitric oxide during the early stage of infection, none of these molecules are crucial for parasitemia control and su…

Sleeping sickness and tsetse awareness: a sociological study among the Tambo and Lambya of the northern Luangwa Valley, Zambia.

Data on awareness of tsetse flies and knowledge of trypanosomiasis were collected in May, 1988, in the Luangwa Valley of Isoka District in the Northern Province of Zambia. One thousand and nine hundred adult males and females were interviewed. There was a high level of fly awareness among all the respondents, regardless of duration of residence and age groups. Malaria was considered as the most serious illness in the community, and hence overshadowed the impact of trypanosomiasis in the community. Disease awareness gravity cannot be attributed to individual's socio-economic levels.

Understanding infectious diseases: modelling approaches for the trypanosomiases.

In recent years, modellers have been enchanted by the prospect of using mathematical equations to describe the transmission of trypanosomiasis and other infectious diseases. Models have been proposed but their value to controlling disease has not been properly understood. Many of these models have been constrained by the limitations of classical modelling techniques. Advances in computer hardware and software offers the prospect of describing disease systems using computer simulations, databases and knowledgebases. These developments point towards better models for the understanding and predic…
