Multi-omics identifies RNF149 as a key molecular marker mediating neuron-glia interaction in temporal lobe epilepsy.

Epilepsy is a complex central nervous system disease with a high incidence and a significant social health burden. Although there are many antiepileptic drugs, about 30% of patients are insensitive to existing drug treatments, and it is urgently required to identify reliable molecular markers and therapeutic targets. Traditional research has focused on a single omics level, and it is difficult to establish a complete connection from genetic variation to changes in cell function. In this study, a multi-level analysis framework integrating transcriptome, proteome, Mendelian randomization, and si

Polyphenols as Systems Modulators of the Gut-Brain-Liver Axis: A Narrative Review of Neuro-Metabolic and Immunomodulatory Networks.

Dietary polyphenols are gaining attention as regulators of gut-brain-liver (GBL) signaling in metabolic disease. The objective of this narrative review is to examine how polyphenols affect neuro-metabolic and immune pathways across the gut-brain-liver axis in obesity, type 2 diabetes, and metabolic dysfunction-associated steatotic liver disease. The review covers literature from 2000 to 2025 indexed in PubMed, Scopus, and Web of Science, with emphasis on mechanistic, multi-omics, preclinical, and human studies. Most polyphenols show poor absorption in the gastrointestinal tract and depend on l

Gut microbiota-driven metabolites modulate the development of stress-related mental disorders.

Stress-related mental disorders, including depression, anxiety, and post-traumatic stress disorder, represent a major global health burden, yet their underlying biological mechanisms and effective therapeutic strategies remain incompletely understood. Growing evidence from both preclinical and clinical studies indicates that alterations in gut microbiota composition and function are closely associated with the onset and progression of these disorders. A central role of the gut microbiota is the biotransformation of dietary and host-derived substrates into diverse metabolites that enter systemi

Non-communicating abomasal (gastric) diverticulum in a 7-day-old bull calf: a case report.

A gastric diverticulum (GD) is defined as an outpouching of the gastric wall and is reported rarely in humans, cats and dogs. A congenital GD (true GD) is a full-thickness diverticulum involving all gastric layers. A 7-day-old bull calf with a history of sudden death was presented for post-mortem examination to the Purdue Animal Disease Diagnostic Laboratory. On gross examination, arising from the wall of the caudal aspect of the greater curvature of the abomasum was a soft to firm hollow mass with no gross evidence of communication with the abomasal lumen. Histological sections of the mass re

Gut microbiota and its metabolites with thyroid diseases: functions and mechanisms.

The gut microbiota, a complex and dynamic ecosystem of microorganisms residing in the gastrointestinal tract, has emerged as a critical regulator of human physiology, extending its influence far beyond digestion. Recent advances have illuminated the existence of a bidirectional communication network known as the "gut-thyroid axis" which posits that the intestinal microbial community and its vast repertoire of bioactive metabolites are fundamental modulators of thyroid health and disease pathogenesis. This comprehensive academic review synthesizes current evidence to elucidate the multifaceted

Single-cell transcriptomics and a boundary wound model reveal temporal decoupling and microenvironmental heterogeneity in radiation-induced fibrosis.

BACKGROUND: Radiotherapy is a key treatment method for cancer, but high-dose treatment can lead to damage to normal tissues and chronic fibrosis. There is a lack of systematic temporal analysis of the dynamic progression from early damage signals to late extracellular matrix (ECM) deposition. METHODS AND MATERIALS: We reanalyzed publicly available scRNA-seq count matrices from GSE211713 to explore microenvironmental remodeling during fibrosis. Then, we established an in vivo local irradiation (12 Gy) mouse model with fully irradiated wounds and boundary-spanning wounds. For tissue repair and

Hepatocellular Carcinoma in Metabolic Dysfunction-Associated Steatotic Liver Disease Beyond Fibrosis: Metabolic, Microbial, and Immune Determinants.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is an increasingly important cause of hepatocellular carcinoma (HCC). Fibrosis and cirrhosis remain the best-validated predictors of HCC, yet a substantial minority of MASLD-related tumors are diagnosed without cirrhosis. This narrative review evaluates what 'beyond fibrosis' should mean for risk assessment rather than treating fibrosis as dispensable. We first separate the proportion of HCC cases that are noncirrhotic from the much lower absolute incidence of HCC in the noncirrhotic MASLD population. We then synthesize human, an

Microbiome-targeted Alzheimer's interventions via gut-brain axis.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder with limited treatment options, underscoring the need for novel therapeutic targets. The gut-brain axis has emerged as a critical bidirectional communication system, with growing evidence establishing gut dysbiosis as a causal factor in AD pathogenesis. This dysbiosis, characterized by a reduction in beneficial microbes and an increase in pro-inflammatory taxa, compromises intestinal and blood-brain barrier integrity, promoting systemic inflammation and the translocation of neurotoxic agents like lipopolysaccharide (LPS). Con

Bird-Scented Nests as a Mechanism for Olfactory Homing in a Burrow-Nesting Seabird.

AbstractNavigation to and from a familiar site is a common animal behavior called homing. Many species use olfactory environmental landmarks or scents deposited in the environment by themselves or their conspecifics for homing. Birds regularly commute to and from their nest, and olfaction, an underappreciated sense in birds, may facilitate this behavior. Burrow-nesting seabirds, for example, rely on olfaction to locate their breeding colony and to identify their burrow, but the specific chemical information they use is unclear. We examined the chemical profiles of the colony landscape and its

Chronic, Battery-Free, Fully Implantable Multimodal Spinal Cord Stimulator for Pain Modulation in Small Animal Models.

Spinal cord stimulation (SCS) for chronic pain management is an invasive therapy involving surgical implantation of electrodes into spinal epidural space. While the clinical value and mechanistic action of the therapy is debated considerably in recent years, preclinical chronic studies employing rodent models can provide invaluable insights regarding the balance between efficacy and complications as well as mechanistic understanding of SCS therapy. However, current rodent compatible devices require tethered power delivery or bulky batteries, severely limiting the ability to probe long-term eff

Liquid biopsy in orthopedic trauma: exosomes as functional mediators and mechanistic indicators in post-traumatic complications.

Severe orthopedic trauma initiates complex pathophysiological cascades that frequently lead to life-threatening complications including acute compartment syndrome, fat embolism syndrome, deep vein thrombosis, and fracture non-union. Traditional biomarkers provide only retrospective indicators of tissue damage, lacking the sensitivity and specificity needed for early complication detection. Exosomes, nanoscale extracellular vesicles carrying proteins, lipids, and nucleic acids, have emerged as critical mediators of intercellular communication that actively participate in trauma pathophysiology.

In-depth behavioral characterization of a rat model of Schaaf-Yang syndrome.

Schaaf-Yang syndrome (SYS, OMIM #615547) is a rare neurodevelopmental disorder caused by truncating variants in the maternally imprinted MAGEL2 gene. It is characterized by intellectual disability, autism spectrum disorder, joint contractures, and feeding difficulties. Although MAGEL2 is deleted in most cases of Prader-Willi syndrome (PWS, OMIM #176270), SYS presents with more severe symptoms, suggesting pathogenic effects of truncated MAGEL2 beyond a mere loss of function. This study expands the behavioral characterization of a novel rat model ("Magel2Pmut rats") which carries a truncating mu

Multi-country and intersectoral assessment of cluster congruence between pipelines for genomics surveillance of foodborne pathogens.

Different laboratories employ different Whole-Genome Sequencing (WGS) pipelines for Food and Waterborne disease (FWD) surveillance, casting doubt on the comparability of their results and hindering optimal communication at intersectoral and international levels. Through a collaborative effort involving eleven European institutes spanning the food, animal, and human health sectors, we aimed to assess the inter-pipeline clustering congruence across all resolution levels and perform an in-depth comparative analysis of cluster composition at outbreak level for four important foodborne pathogens: L

Examining the Uncertainties Surrounding Exosome Therapy in Androgenetic Alopecia: A Call for Evidence-Based Practice.

Hair loss, a pervasive and often distressing condition, affects a substantial number of individuals globally. Although conventional treatments such as hair transplantation, topicals, oral medications, and injectables exist, they have limitations, including the necessity for repeated treatments, potential adverse effects, and cost barriers. Exosome therapy, an innovative and burgeoning option within regenerative medicine, offers a novel approach to hair loss treatment. Exosomes are small vesicles that are produced from the membranes of late-endosomes and secreted by cells, playing a crucial rol

Short- and Long-Term Neurobehavioral Effects of Developmental Exposure to Valproic Acid in Zebrafish.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by impairments in social interaction and communication, anxiety, hyperactivity, and interest restricted to specific subjects. In addition to the genetic factors, multiple environmental factors have been related to the development of ASD. Animal models can serve as crucial tools for understanding the complexity of ASD. In this study, a chemical model of ASD has been developed in zebrafish by exposing embryos to valproic acid (VPA) from 4 to 48 h post-fertilization, rearing them to the adult stage in fish water. For th

Effects of (S)-3,4-DCPG, an mGlu8 receptor agonist, on hippocampal long-term potentiation at perforant pathway-dentate gyrus synapses in prenatal valproic acid-induced rat model of autism.

Autism spectrum disorder (ASD) is a pervasive neurodevelopmental condition characterized by social interaction deficits, communication impairments, repetitive behaviors, and sensory sensitivities. While the etiology of ASD is multifaceted, abnormalities in glutamatergic neurotransmission and synaptic plasticity have been implicated. This study investigated the role of metabotropic glutamate receptor 8 (mGlu8) in modulating long-term potentiation (LTP) in a rat model of ASD induced by prenatal valproic acid (VPA) exposure. To induce an animal model with autism-like characteristics, pregnant rat

Nanoparticles Based on Natural Lipids Reveal Extent of Impacts of Designed Physical Characteristics on Biological Functions.

Nanoparticles based on lipids (LNPs) are essential in pharmaceuticals and intercellular communication, and their design parameters span a diverse range of molecules and assemblies. In bridging the gap in insight between extracellular vesicles (EVs) and synthetic LNPs, one challenge is understanding their in-cell/in-body behavior when simultaneously assessing more than one physical characteristic. Herein, we demonstrate comprehensive evaluation of LNP behavior by using LNPs based on natural lipids (N-LNPs) with designed physical characteristics: size tuned using microfluidic methods, surface fl

Embryonic signals mediate extracellular vesicle biogenesis and trafficking at the embryo-maternal interface.

BACKGROUND: Extracellular vesicles (EVs) are membrane-coated nanoparticles secreted by almost all cell types in living organisms. EVs, as paracrine mediators, are involved in intercellular communication, immune response, and several reproductive events, including the maintenance of pregnancy. Using a domestic animal model (Sus scrofa) with an epitheliochorial, superficial type of placentation, we focused on EV biogenesis pathway at the embryo-maternal interface, when the embryonic signaling occurs for maternal recognition and the maintenance of pregnancy. RESULTS: Transmission electron micros
