Virus-like particle vaccines: from antigen architecture to antigen presentation and adaptive immune responses.

INTRODUCTION: Virus-like particles (VLPs) mimic key structural features of native viruses while lacking a replication-competent viral genome, combining favorable safety with multivalent antigen presentation and strong immunogenicity. Yet, how VLP design influences the quality and persistence of adaptive immunity remains incompletely understood. AREAS COVERED: This review examines relevant literature identified through targeted searches on recent advances in VLP engineering, with emphasis on particle architecture, antigen-display strategies, and structural determinants of immune recognition. W…

The Architecture of Deep Phenotyping in Asthma: Integrating Molecular, Metabolic, and Neuro-Hormonal Endotypes.

Asthma is increasingly recognized as a heterogeneous syndrome where traditional management fails, particularly given spirometry's limitations in assessing small airway dysfunction. This review synthesizes the transition from clinical phenotyping to deep molecular endotyping, establishing a framework for precision medicine. We highlight the insufficiency of absolute eosinophil counts, proposing eosinophil cationic protein (ECP) and eosinophil-derived neurotoxin (EDN) as superior activation metrics. Furthermore, we explore Type 2 drivers (IL-4/IL-13, periostin) and epithelial alarmins like TSLP.

Crumpled Nanosheet architecture of a Ni3B2O6@V4C3 hybrid for electrochemical detection of Chlorothalonil fungicide.

The widespread use of the highly toxic fungicide chlorothalonil (CTL), which is lethal to fish at ppb levels and harms amphibians and soil ecosystems, necessitates robust monitoring tools. This work presents a high-performance electrochemical sensor based on a novel nickel borate/vanadium carbide (Ni3B2O6/V4C3) nanocomposite. The synergistic integration of catalytically active Ni3B2O6 (NiBO) with conductive V4C3 (VC) creates an interface that enhances charge transfer for CTL reduction. To our knowledge, this study provides the most comprehensive electrochemical investigation of CTL to date, ut

Microglial P2Y12 signaling phase-shifts glymphatic circadian rhythms and alters sleep architecture in mice.

Microglia regulate neural circuits and vascular-glial interfaces, yet whether microglial purinergic signaling coordinates glymphatic function and brain state remains unclear. We tested the role of the microglial Gi-coupled receptor P2Y12 using acute pharmacological inhibition with MRS2395 in adult mice. Glymphatic influx was quantified across circadian time by cisterna-magna tracer infusion and whole-brain fluorescence imaging, and related to microglial density and morphology assessed by Iba1 immunofluorescence and Sholl analysis in cortex, hippocampus, and amygdala. Sleep-wake architecture an…

Spatial architecture of tertiary lymphoid structures represents an independent prognostic dimension in hepatocellular carcinoma.

BACKGROUND AND AIMS: Tertiary lymphoid structures (TLS) are associated with heterogeneous outcomes in hepatocellular carcinoma (HCC), but whether their anatomical context determines clinical impact remains unknown. We hypothesized that spatial compartmentalization of TLS influences their prognostic significance. METHODS: We developed SpatialDecoder, a deep learning pipeline that simultaneously segments TLS, classifies three maturation subtypes (Agg, Fol I, Fol II), and assigns spatial compartments (intratumoral, peritumoral, capsular), achieving accurate tissue segmentation (mean Dice similar

Heteroepitaxial Control of Thickness, Strain, and Domain Architecture in Few-Layer Ferroelectric Tin Monochalcogenides.

Thin-film epitaxy and epitaxial strain have been widely exploited to tune domain configurations, switching behavior, and ferroic properties in conventional three-dimensional ferroelectric thin films; however, its application to controlling the properties of two-dimensional (2D) ferroelectrics has remained largely unexplored. Here, using SnX (X = Se, S) as a model system, we demonstrate heteroepitaxial control of thickness, strain state, and domain architecture in few-layer ferroelectric SnX via growth on monolayer MoS2 van der Waals (vdW) templates. Compared with conventional growth, MoS2-temp…

Large-scale genomic analysis of jumbo phages: coevolution, genome architecture, and host interaction mechanisms.

BACKGROUND: Jumbo phages are phages with comparatively large genome sizes. Jumbo phages have been identified in various microbial communities. However, their diversity, genome structure, potential function, and their interactions with hosts and other phages are largely unknown due to insufficient genomic data. RESULTS: We collected 59,652,008 putative viral genomes from seven habitats by using 38 public metagenome datasets, an integrated public viral genome database (IGN), and pig gut viral genome databases. We obtained 10,754 jumbo phage genomes with sizes ranging from 200 to 831 kb. Most (94

Conserved macromolecular architecture of poplar secondary cell walls revealed by ssNMR and atomistic modeling.

The macromolecular architecture of plant secondary cell walls governs wood's mechanical and biochemical properties, yet its natural intra-species variability remains poorly characterized. Here, we combined ¹³C solid-state NMR (ssNMR), multivariate statistical analysis, and molecular modeling to profile nanoscale structure across 13 genetically diverse Populus trichocarpa genotypes grown in ¹³C-enriched atmospheres. SsNMR-derived phenotypes spanning composition, structure, mobility, and inter-polymer proximities reveal a conserved architecture, with a subtle yet coordinated variation organizing…

Multifunctional Single-Transistor Architecture Enabled by Nonvolatile Band Rearrangement in 2D Ferroelectric Tunneling Junction.

A defining challenge for next-generation nanoelectronics is integrating diverse functionalities within a single device while strictly minimizing power consumption. Two-dimensional (2D) ferroelectrics, with their atomic-scale nonvolatility, offer a promising platform for such architectural innovation. Here, we theoretically propose a multifunctional single-transistor (1T) architecture embedding a 2D ferroelectric tunneling junction at the source-channel interface. Polarization reversal deterministically drives a nonvolatile band rearrangement, toggling the interface between a gapless type-III a…

A Multiobjective Evolutionary Algorithm Based on Bipopulation With Uniform Sampling for Neural Architecture Search.

Neural architecture search (NAS) automates neural network design, improving efficiency over manual approaches. However, efficiently discovering high-performance neural network architectures that simultaneously optimize multiple objectives remains a significant challenge in NAS. Existing methods often suffer from limited population diversity and inadequate exploration of the search space, particularly in regions with extreme complexity values. To address these challenges, we propose a multiobjective evolutionary algorithm (MOEA)-BUS, an innovative MOEA based on bipopulation with uniform samplin…

Deep learning-based classification of colonoscopic images using an attention-enhanced ConvNeXt V2 architecture.

INTRODUCTION: Proper interpretation of the colonoscopic images is important to early detect and diagnose colorectal diseases like polyps and inflammatory bowel diseases. However, the complex visual patterns and high intra-class similarity of such images make the automated classification task challenging. METHODS: In this work, we propose an attention enhanced deep learning framework using ConvNeXt V2 for robust multi-class classification of colonoscopic images. The proposed method employs a Convolutional Block Attention Module (CBAM) in ConvNeXt V2 architecture to improve the feature represen…

Mapping the cortical architecture of sleep deprivation: insights from fMRI, neurotransmission, and metabolic activity.

BACKGROUND: Sleep deprivation (SD) produces profound cognitive deficits, yet its integrative effects on large-scale cortical organization, metabolic alterations, and cognitive function remain unclear. METHODS: Based on meta-analytic seeds, lesion network mapping was applied to connectome data from 1000 healthy individuals to generate an SD-related cortical weight map (SD-CWM). Thirty participants then completed a 24-hour acute SD protocol, during which resting-state and memory-task-evoked brain activities, untargeted metabolomics, and memory performance were measured under both rested wakeful

A computational model to describe multi-regional brain architecture during neurodegeneration in Alzheimer's disease.

We previously proposed an MRI-based machine learning model to describe the mesoscopic architecture of the human brain to aid in classifying subjects as having non-AD related pathology (nADrp) or AD related pathology (ADrp), including mild cognitive impairment (MCI) and Alzheimer's disease (AD). The method, developed on data from patients scanned at 1.5T showed high performance, but did not generalise well to scans obtained from 3T MRI. In the current work we overcome the problem and extend the approach to patients scanned longitudinally, and at different field strengths. Retrospective T1-MRI d…
