De novo design of cysteine proteases.

Despite advances in de novo enzyme design, success has been largely limited to low-energy-barrier model reactions. Amide bonds such as those linking amino acids along the peptide backbone are stable for hundreds of years in neutral aqueous solution because of the high energy barrier to hydrolysis1. Here we describe the de novo design of enzymes that use an activated cysteine nucleophile to hydrolyse the polypeptide backbone in a sequence-dependent manner, with a success rate of 13/69 = 19% and rate enhancements over the background reaction (kcat/kuncat) of up to 3 × 107. The designed proteases

The contractual vacuum: AI procurement and the erosion of good administration.

The integration of artificial intelligence as a service (AIaaS) within public administration fundamentally relocates the state's evidentiary and inferential capacities into proprietary software infrastructures. When public bodies deploy these third-party systems, they introduce an acute systemic risk: a structural "contractual vacuum" emerges wherein public authorities lack the practical data telemetry required to reconstruct, explain, or correct algorithmic administrative actions. This article evaluates the doctrinal limitations of contemporary European and common law public procurement frame

Graph theoretical characterization and QSPR modeling of aluminum and magnesium hydroxide networks through integrated face degree topological indices.

Aluminum hydroxide (gibbsite) and magnesium hydroxide (brucite) rank among the most prevalent hydroxide minerals and function as structural prototypes for diverse layered materials, including phyllosilicates, layered double hydroxides, and emerging two-dimensional halides. An exciting feature of these materials is their novel electronic and spintronic functionalities. Gibbsite adopts a dioctahedral framework with one-third of its octahedral sites vacant and stabilized by hydrogen bonding, whereas brucite forms a fully occupied trioctahedral lattice bound by van der Waals interactions. Such str

PETRA: a pedestal exoskeletal testbed with real-time architecture for upper limb rehabilitation robotics.

More than 85% of stroke survivors present motor impairment in the upper limb, yet access to rehabilitation robotics remains concentrated in high-income countries due to costs exceeding 10,000 USD and architectures dependent on external computers. This economic barrier limits the ability of researchers in low- and middle-income countries (LMICs) to develop and validate new therapeutic strategies. This article presents PETRA (Pedestal Exoskeletal Testbed with Real-time Architecture), an open-source, 3-degree-of-freedom kinematic validation testbed manufactured in PETG for upper limb rehabilitati

Personality in negotiation revisited: Recent advances and future directions.

Influential voices long dismissed individual differences as irrelevant to negotiation research. Recent calls to examine the process in its full complexity invite a renewed look at personality in negotiation. We take stock of the evidence using three complementary lenses: When does personality matter across contexts, outcomes, and phases? With whom does personality matter in interaction, including partner and fit effects? How should researchers design studies and measures that can recover stable personality effects in interdependent settings? Across these lenses, recent advances suggest that tr

Noise analysis and vibration control of flow-induced vibration in the rotating detonation combustor.

The persistent operation of rotating detonation engines (RDEs) is severely constrained by intense combustion chamber vibrations and excessive noise emissions. Existing research on engine noise has been predominantly confined to experimental investigations of noise generation mechanisms and sound pressure levels, leaving critical gaps in predictive modeling. To address this, this study employs a numerical methodology for analyzing transient flow fields within the combustion chamber, enabling systematic characterization of vibration-coupled noise phenomena. Leveraging this computational approach

Functional bipartite invariance in mouse primary visual cortex receptive fields.

Sensory systems support generalization by representing features that persist under input variation; however, identifying the neuronal basis of these invariances remains difficult due to high-dimensional and nonlinear neural computations. Here we leverage the inception loop paradigm, iterating between large-scale recordings, predictive models and in silico experiments with in vivo verification, to characterize neuronal invariances in mouse primary visual cortex (V1). We synthesize varied exciting inputs (VEIs), dissimilar images that drive target neurons. These VEIs revealed a new bipartite inv

Predicting ionic conductivity of sulfide glass solid-state electrolytes using max-flow methodology.

Disordered materials such as glasses are promising candidates as solid-state electrolytes for batteries, but they tend to exhibit low ionic conductivity and current methods for investigating and predicting the ionic conductivity of electrolyte materials are inefficient. Here, we propose a framework that summarizes the topological properties of the electrolyte material's atomic structure with a weighted graph based on periodic boundary conditions. We then compute a variation of the max-flow algorithm, a commonly used model in continuum models of transportation and liquid flow, on this summary a

Critical structural features of HDAC11 inhibitors.

Histone deacetylase 11 (HDAC11), the sole member of the class IV HDACs, has gained significance as a key regulator of a wide range of physiological and pathological processes. The structural characterization of HDAC11 remains challenging due to the absence of an experimentally determined crystal structure. Proper analysis of reported HDAC11 inhibitors can yield valuable insights into the key structural fingerprints essential for inhibition. Using a diverse range of descriptor sets, including various 2D descriptors and fingerprint-based descriptors, it becomes possible to effectively identify c

Deep Learning-Based Image Classification of Pupae from 11 Lepidoptera Pest Species.

The morphological identification of lepidopteran pest pupae has long been a difficult task. To explore automated solutions, this study established a standardized, multi-angle image dataset of pupae from 11 economically important lepidopteran pests. We then systematically evaluated six deep learning models, including both convolutional neural networks and Transformer architectures. The results show that all models successfully learned to distinguish the vast majority of species, with Vit-Small achieving the highest accuracy (98.71 ± 0.16%) and the highest F1-score (98.69 ± 0.20%). This confirms

Towards a novel toxicity prediction pipeline, combining PBPK models and mechanistic cellular adversity models.

There is a strong need for animal-free methods that properly predict drug adversity. Especially drug-induced liver injury (DILI) is a common adverse effect that is difficult to predict with current methodology. In this study, we developed a data-driven computational adversity model for predicting the relative number of apoptotic cells in vitro, based on stress pathway activity invoked by exposure of liver cells to nitrofurantoin, diclofenac, and ketoconazole. Next, we adapted this adversity model to represent an in vivo situation and developed physiologically-based pharmacokinetic (PBPK) model

Metalloid Tin Clusters, SnntBum, with α-Sn-Like Structural Motifs.

Metalloid tin clusters represent an intriguing class of compounds that bridge molecular chemistry and the solid state, yet access to diverse examples remains limited. Herein, we present a mild, modular hydrostannolysis-type strategy that enables the synthesis of tin-based clusters stabilized by fairly small tert-butyl substituents, thereby overcoming the ligand-imposed structural limitations of established methodologies. The approach gave access to a series of clusters of different core sizes, including Sn11tBu12, featuring a [1.1.1]propellane-type backbone, as well as larger Sn15tBu14 and Sn1

Handling uncertainty in optimization enabled the deep Q network for traffic sign and criticality detection.

OBJECTIVE: This research aims to develop an efficient model for Traffic Sign Recognition (TSR) with uncertainty quantification. METHODS: The process involves pre-processing, sign localization, TSR, and criticality detection. After pre-processing with a Gaussian filter, sign localization is carried out using Segmentation U-Net (SegU-Net). Following TSR, a Deep Q Network (DQN) is trained using Gradient Descent-Teamwork Optimization Algorithm-based Deep Q Network (GD-TOA based DQN), which incorporates Gradient Descent model into Teamwork Optimization Algorithm (TOA), is used to classify traffic

Reproducibility and Validation Challenges in AI-Driven Scaffold Design for Bone Regeneration.

Artificial intelligence (AI) is increasingly applied to bioink formulation and bioprinting process control in tissue engineering (TE). Yet, translational progress remains constrained by small datasets, limited cross-platform validation, and weak links between computational predictions and biological outcomes. This review critically evaluates experimentally validated AI applications across scaffold-based bone regeneration, spanning materials design, fabrication control, and biological assessment. Physics-informed models are often more robust and generalizable than purely data-driven approaches,

A study on ultrasound imaging for thyroid detection and classification using machine learning and deep learning techniques.

Thyroid diseases are increasingly prevalent and have a major influence on endocrine and metabolic functions. Ultrasound imaging is popular for early diagnosis and categorizing thyroid abnormalities because it is non-invasive, inexpensive, and real-time. From this paper, we planned to study the recent developments in computer-aided diagnosis (CAD) systems in detecting and classifying thyroid diseases based on ultrasound images. Researchers are focused on traditional image processing algorithms, machine learning techniques, and deep learning techniques to improve accuracy. Modern classifiers lik

Uncertainty quantification and global sensitivity analysis of a patient-specific mandibular finite element model using Latin hypercube sampling and sparse polynomial chaos expansion.

Most mandibular finite element models are deterministic and overlook the influence of biological and modeling variability. This study proposes an automated, uncertainty-aware computational workflow for a patient-specific mandibular FE model using Latin Hypercube Sampling and sparse Polynomial chaos expansion. We investigated 24 input parameters related to masticatory muscle loading, material properties, and geometric discretization over 2400 simulations. Global and regional sensitivity analyses were performed using both Spearman rank correlations and variance-based Sobol indices to identify th

Exploring hyperfine coupling in molecular qubits.

Molecular qubits represent a promising avenue for advancing quantum sensing and computing technologies, yet significant challenges remain in optimising their performance. Hyperfine coupling has a critical influence on molecular qubit properties. While previous studies have exhaustively investigated this phenomenon, a comprehensive understanding of the underlying mechanisms across different systems remains elusive. A benchmark test was performed using DFT to assess which methodology worked best to accurately predict hyperfine coupling constants in molecular qubits predominantly composed of VIV

Generation of Pure Spin Current with Insulating Antiferromagnetic Materials.

The generation of pure spin currents is critical for low-dissipation spintronic applications, yet existing methods relying on spin-orbit coupling or ferromagnetic interfaces face challenges in material compatibility and operational robustness. We propose a paradigm-shifting approach to generate symmetry-protected pure spin currents by applying mechanical stress on insulating antiferromagnetic materials, i.e., the pure piezospintronic effect. We first classify magnetic point groups enabling pure piezospintronic effects. A novel first-principles method is developed to compute the spin dipole mom

Aerodynamic and Acoustic Power in Infant Cry.

OBJECTIVES: Acoustic and aerodynamic powers in infant cry are not scaled downward with body size or vocal tract size. The objective here was to show that high lung pressures and impedance matching are used to produce power levels comparable to those in adults. STUDY DESIGN AND METHODOLOGY: A computational model was used to obtain power distributions along the infant airway. The parameters were cross-sectional areas of the larynx canal and the velar region, which tend to be variable in infant cry when there is no specific vowel structure. RESULTS: Aerodynamic power can reach 1.0 W with 70 cm
