Biochemical mechanisms in asbestos toxicity.

The alarming hazardous nature of asbestos makes it the foremost among toxic fugitive dusts. The biochemical mechanisms responsible for the diverse biological effects of asbestos, such as fibrosis, asbestos bodies, pleural plaques, respiratory difficulty, cancer, and cytotoxicity, are being studied in this laboratory. As asbestosis progresses in guinea pigs, along with reticulum formation, lysosomal enzymes are released from membrane-bound latent state to active free form, initiating degradative changes. Considerable alterations take place in the pulmonary metabolic machinery. Mitochondria in l

Biochemical and pathological changes in tissues of Friesian cattle during the experimental induction of copper deficiency.

1. Copper deficiency was induced in five Friesian cattle offered a semi-synthetic diet containing less than 1 mgCu/kg. Changes in blood and liver Cu contents and in the Cu-containing enzymes, ferroxidase I (caeruloplasmin; EC 1.16.3.1) and monoamine oxidase (EC 1.4.3.4) of plasma and cytochrome oxidase (EC 1.9.3.1) of liver and skeletal muscle were monitored during Cu depletion. 2. Rapid decreases in blood and liver Cu and plasma ferroxidase I activity were found at least 80 d before the first appearance of overt clinical signs of deficiency. Plasma monoamine oxidase and liver cytochrome oxida

DESCRIPTION, REDESCRIPTION, AND LIFE CYCLES OF PARORCHIS LAFFERTYI N. SP. AND PARORCHIS CATOPTROPHORI (TREMATODA: DIGENEA: PHILOPHTHALMIDAE) FROM THE CALIFORNIA HORN SNAIL, CERITHIDEOPSIS CALIFORNICA (GASTROPODA: POTAMIDIDAE).

We describe Parorchis laffertyi n. sp., a trematode infecting the California horn snail, Cerithideopsis californica (Haldeman, 1840), as a first intermediate host. The first intermediate host stages of this species were previously cryptic with Parorchis catoptrophori Dronen & Blend, 2008, which also infects C. californica. For both species, we describe the parthenita colonies, soldier rediae, reproductive rediae, cercariae, metacercariae, and sexual adults by using DNA sequencing and material from natural and experimental infections. Our study of all life stages was conducted with material ori

Characterizing mixed strongyle infections in foals and broodmares using cytochrome c oxidase subunit I deep amplicon sequencing.

BACKGROUND: Mixed strongyle infections represent the most prevalent equine parasitosis and can result in life-threatening disease, especially in young horses. Species involvement and pathogenesis of this parasitosis are poorly understood, and data on foals and broodmares are notably lacking. METHODS: In a longitudinal study undertaken in 2022 in Germany, individual faecal samples (n = 497) and metadata were collected for naturally infected foals and broodmares (n = 48) kept under conventional husbandry conditions. Nematode infections were detected coproscopically via the Mini-FLOTAC method. I

Genetic and pharmacological targeting of HINT2 promotes OXPHOS to alleviate inflammatory responses and cell necrosis in acute pancreatitis.

The necrosis of pancreatic acinar cells is a key molecular event in the progression of acute pancreatitis (AP), with disturbances in mitochondrial energy metabolism considered to be a direct causative factor of acinar cell necrosis. Histidine triad nucleotide-binding protein 2 (HINT2) has been implicated in the development of various diseases, whereas its involvement in the progression of AP remains unclear. This study aims to investigate the role of HINT2 in AP. HINT2 expression in pancreatic tissues was significantly downregulated after AP. The results of glutathione-S-transferase (GST) pull

Metacercariae of Diplostomum spp. from eyes of the Bullhead Cottus koshewnikowi Gratzianov, 1907 from the northern European watershed.

Metacercariae of Diplostomum spp. are widespread fish parasites. In this study we obtained the first data on infection of Bullhead Cottus koshewnikowi with these larvae in five rivers of northern Europe (Finland and Russia) using molecular and morphological description. Three Diplostomum spp. were revealed in the eyes of bullheads. Diplostomum spathaceum and D. mergi Lineage 3 sensu Georgieva et al. (2013) were found in the lens, while Diplostomum sp. Lineage 6 sensu Blasco-Costa et al. (2014) was found in the retina. We obtained molecular data on these three species and provided morphological

Seek, and you will find: Cryptic diversity of the cardiopulmonary nematode Angiostrongylus vasorum in the Americas.

Angiostrongylus vasorum is a metastrongylid parasite infecting wild canids and domestic dogs. Its patchy distribution, high pathogenicity and taxonomical classification makes the evolutionary history of A. vasorum intriguing and important to study. First larval stages of A. vasorum were recovered from feces of two grey foxes, Urocyon cinereoargenteus, from Costa Rica. Sequencing and phylogenetic and haplotypic analyses of the ITS2, 18S and cytochrome oxidase subunit 1 (cox1) fragments were performed. Then p- and Nei´s genetic distance, nucleotide substitution rates and species delimitation ana

Hallmarks of oxidative stress in the livers of aged mice with mild glycogen branching enzyme deficiency.

Glycogen branching enzyme (GBE1) introduces branching points in the glycogen molecule during its synthesis. Pathogenic GBE1 gene mutations lead to glycogen storage disease type IV (GSD IV), which is characterized by excessive intracellular accumulation of abnormal, poorly branched glycogen in affected tissues and organs, mostly in the liver. Using heterozygous Gbe1 knock-out mice (Gbe1+/-), we analyzed the effects of moderate GBE1 deficiency on oxidative stress in the liver. The livers of aged Gbe1+/- mice (22 months old) had decreased GBE1 protein levels, which caused a mild decrease in the d

Genetic diversity of Fasciola hepatica in Austria.

The worldwide occurring common liver fluke Fasciola hepatica can infect humans and animals and leads to considerable illness and economic loss annually. The aim of this study was to determine the genetic diversity of F. hepatica in Austria. In total, 31 adult flukes isolated from cattle from various regions in Austria were investigated for their cytochrome oxidase subunit 1 (cox1) and nicotinamide dehydrogenase subunit 1 (nad1) gene sequences. It was shown that Austrian isolates of F. hepatica reveal extensive genetic diversity. To the best of our knowledge, these are the first data on the div

Diversity of Taenia and Hydatigera (Cestoda: Taeniidae) in domestic dogs in Kenya.

Taenia species of domestic dogs can cause cysticercosis and coenurosis in a wide range of intermediate hosts including humans. Most taeniids of dogs are globally distributed, but some wildlife-transmitted species can be specific for certain regions. Generally, little information exists on the species composition and frequency in most regions of the world, which impairs risk assessment and control strategies. This study determined the range of taeniid species in dogs in four widely spaced areas of Kenya by genetic identification of eggs in faeces collected from the environment. Individual taeni

Genetic characterization of three mitochondrial gene sequences of goat/sheep-derived coenurus cerebralis and cysticercus tenuicollis isolates in Inner Mongolia, China.

Taenia multiceps and Taenia hydatigena are widely distributed tapeworms of canids. Due to a lack of genetic information on these two parasites in China, in this study we analyzed six coenurus cerebralis and two cysticercus tenuicollis cysts from goats or sheep in Inner Mongolia, northern China by amplifying three mitochondrial genes (cox1, nad4, and cytb). Two haplotypes were obtained at each locus for either of the two Taenia cestode species, with ten nucleotide sequences being novel. The degrees of genetic variations were 1.18%, 0.61% and 0.52% for coenurus cerebralis, and 0.24%, 0.46% and 0

Utility of the cytochrome c oxidase subunit I gene for the diagnosis of toxoplasmosis using PCR.

Toxoplasmosis is caused by the protozoan parasite Toxoplasma gondii, which belongs to the phylum Apicomplexa. Since this parasite causes severe clinical symptoms in immunocompromised patients, early diagnosis of toxoplasmosis is essential. PCR is currently used for early diagnosis, but there is no consensus regarding the most effective method for amplifying Toxoplasma DNA. In this study, we considered the utility of the cytochrome c subunit I (cox1) gene, which is encoded in the mitochondrial DNA of this parasite, as a novel target of PCR for the diagnosis of toxoplasmosis. To do this, we comp

Description of a new species, Trichuris ursinus n. sp. (Nematoda: Trichuridae) from Papio ursinus Keer, 1792 from South Africa.

In the present work, we carried out a morphological, biometrical and molecular study of whipworms Trichuris Roederer, 1761 (Nematoda: Trichuridae) parasitizing Papio ursinus Keer 1792 (Chacma baboon). Biometrical and molecular data suggest a new species of Trichuris parasitizing baboons. In addition of main morphological features (spicule, spicule sheath, spicule tube, proximal cloacal tube, distal cloacal tube, vulva, vagina), the mean values of individual variables between Trichuris colobae, Trichuris suis, Trichuris trichiura examined by Student's t tests suggest that T. ursinus n. sp. cons

Prevalence of Hymenolepis nana and H. diminuta from Brown Rats (Rattus norvegicus) in Heilongjiang Province, China.

Hymenolepis nana and Hymenolepis diminuta are globally widespread zoonotic cestodes. Rodents are the main reservoir host of these cestodes. Brown rats (Rattus norvegicus) are the best known and most common rats, and usually live wherever humans live, especially in less than desirable hygiene conditions. Due to the little information of the 2 hymenolepidid species in brown rats in China, the aim of this study was to understand the prevalence and genetic characterization of H. nana and H. diminuta in brown rats in Heilongjiang Province, China. Total 114 fecal samples were collected from brown ra

First insights into the genetic diversity of Echinococcus granulosus sensu stricto (s.s.) in Serbia.

Cystic echinococcosis (CE) caused by Echinococcus granulosus sensu stricto (s.s.) is a cosmopolitan zoonotic infection which is endemic in Serbia where it is subject to mandatory reporting. However, information on the incidence of the disease in humans and prevalence of hydatid infection in livestock remains limited. We used sequenced data of the cytochrome c oxidase subunit 1 (cox 1) mitochondrial gene to examine the genetic diversity and population structure of E. granulosus (s.s.) from intermediate hosts from Serbia. We also compared our generated nucleotide sequences with those reported fo

Decreased copper concentrations but increased lysyl oxidase activity in ischemic hearts of rhesus monkeys.

Myocardial ischemia leads to a decrease in copper (Cu) concentrations, along with collagen deposition in which Cu-dependent lysyl oxidase (LOX) catalyzes the cross-linking of collagens leading to tissue stiffness. The present study was undertaken to determine the relationship between decreased Cu concentrations and LOX activities in ischemic hearts of monkeys. Rhesus monkeys were subjected to coronary artery ligation, leading to ischemic infarction. At 8 weeks after the surgery, Cu concentrations and Cu-dependent cytochrome c oxidase (CCO) activities in the infarct area were significantly decr
