Autophagy Suppresses CCL2 to Preserve Appetite and Prevent Lethal Cachexia.

Macroautophagy (autophagy hereafter) captures intracellular components and delivers them to lysosomes for degradation and recycling1. In adult mice, autophagy sustains metabolism to prevent wasting by cachexia and to survive fasting, and also suppresses inflammation, liver steatosis, neurodegeneration, and lethality2,3. Defects in autophagy contribute to metabolic, inflammatory and degenerative diseases, however, the specific mechanisms involved were unclear 4. Here we profiled metabolism and inflammation in adult mice with conditional, whole-body deficiency in an essential autophagy gene and

Micro-nano-bio acoustic system for the detection of foodborne pathogens in real samples.

The fast and efficient detection of foodborne pathogens is a societal priority, given the large number of food-poisoning outbreaks, and a scientific and technological challenge, given the need to detect as little as 1 viable cell in 25 gr of food. Here, we present the first approach that achieves the above goal, thanks to the use of a micro/nano-technology and the detection capability of acoustic wave sensors. Starting from 1 Salmonella cell in 25 ml of milk, we employ immuno-magnetic beads to capture cells after only 3 h of pre-enrichment and subsequently demonstrate efficient DNA amplificati

Comprehensive assessment of the global and regional vascular responses to food ingestion in humans using novel rapid MRI.

Ingestion of food is known to increase mesenteric blood flow. It is not clear whether this increased flow demand is compensated by a rise in cardiac output (CO) alone or by redistribution of blood flow from other organs. We used a new comprehensive imaging method to assess the human cardiovascular response to food ingestion. Following a 12-h fast, blood flow in segments of the aorta and in organ-specific arteries, and ventricular volumes were assessed in 20 healthy adults using MRI at rest and following ingestion of a high-energy liquid meal. Systemic vascular resistance (SVR) fell substantial

Non food-related risk factors of campylobacteriosis in Canada: a matched case-control study.

BACKGROUND: Campylobacteriosis is a prominent bacterial gastrointestinal infection worldwide with several transmission pathways. Its non-foodborne routes have been less documented and quantified. The study aimed to quantitatively explore the role of potential risk factors not directly associated with food for sporadic cases of C. jejuni infection in Canada. METHODS: This retrospective matched case-control study was built on an enhanced campylobacteriosis surveillance system and on a survey of healthy people and their behaviour with regards to potential risk factors for gastrointestinal infect

Effect of anorexinergic peptides, cholecystokinin (CCK) and cocaine and amphetamine regulated transcript (CART) peptide, on the activity of neurons in hypothalamic structures of C57Bl/6 mice involved in the food intake regulation.

The hypothalamus plays an important role in food consumption, receiving information about energy balance via hormonal, metabolic, and neural inputs. Its neurons produce neuropeptides influencing energy balance. Especially important to regulation of food consumption are certain hypothalamic structures, including the arcuate (ARC) and ventromedial (VMN) nuclei and the lateral hypothalamic area (LHA). We determined the impact of cholecystokinin (CCK) and cocaine and amphetamine regulated transcript (CART) peptide, on activity of ARC and VMN neurons and hypocretin (Hcrt) synthesizing neurons in LH

Exploring the impact of Ramadan fasting on women's reproductive health: A narrative review.

Ramadan is the ninth month of the Islamic lunar calendar in which Muslims abstain from consumption of food and drink, smoking, and engaging in sexual activity during daylight hours for the full lunar month, approximately 29-30 days. Beyond its spiritual significance, fasting during this month represents a distinct physiologic and cultural practice that may influence women's reproductive health. This narrative review integrates findings from published studies to summarize current evidence on the impact of Ramadan fasting (RF) on the menstrual cycle, fertility, polycystic ovary syndrome, pregnan

Transcriptomic Analysis of the Liver Redox Response During Food-Anticipatory Activity Under a Time-Restricted Feeding Protocol in Rats.

Daytime-restricted feeding (TRF) exerts outstanding effects on circadian physiology, nutrient utilization, and energy metabolism. Limiting feeding access to two hours during the daytime (12:00-14:00 h) for three weeks promotes food-anticipatory activity (FAA). FAA encompasses not only behaviors related to meal expectations but also includes diurnal fluctuations in liver metabolic responses, including distinct redox handling. Hepatic microarray profiles of genes associated with redox response processes were analyzed at three crucial time points: at the beginning of the light period or before FA

Ghrelin and feedback systems.

Ghrelin is produced primarily in the stomach in response to hunger, and circulates in the blood. Plasma ghrelin levels increase during fasting and decrease after ingesting glucose and lipid, but not protein. The efferent vagus nerve contributes to the fasting-induced increase in ghrelin secretion. Ghrelin secreted by the stomach stimulates the afferent vagus nerve and promotes food intake. Ghrelin also stimulates pituitary gland secretion of growth hormone (GH) via the afferent vagus nerve. GH inhibits stomach ghrelin secretion. These findings indicate that the vagal circuit between the centra

Glucoprivation induces anestrus and lipoprivation may induce hibernation in Syrian hamsters.

Previous results supported the notion that estrous cycles in Syrian hamsters are responsive to the general availability of metabolic fuels, rather than to either fatty acid or glucose availability per se. To test this idea, we monitored estrous cycles in hamsters that were fed ad libitum and treated with a range of doses of 2-deoxy-D-glucose (2-DG), an inhibitor of glucose utilization. Hamsters treated with 2-DG at doses ranging from 750-1,250 mg/kg showed normal estrous cycles, but higher doses (1,750 or 2,000 mg/kg) induced anestrus. While it is clear from these data that estrous cycles are

Generation of Lactose- and Protease-Positive Probiotic Lacticaseibacillus rhamnosus GG by Conjugation with Lactococcus lactis NCDO 712.

Lacticaseibacillus rhamnosus GG (LGG) is the most studied probiotic bacterium in the world. It is used as a probiotic supplement in many foods, including various dairy products. However, LGG grows poorly in milk, as it neither metabolizes the main milk carbohydrate lactose nor degrades the major milk protein casein effectively. In this study, we made L. rhamnosus GG lactose and protease positive by conjugation with the dairy Lactococcus lactis strain NCDO 712 carrying the lactose-protease plasmid pLP712. A lactose-hydrolyzing transconjugant colony was obtained on agar containing lactose as the

Timing of food intake impacts cardiovascular and neuroendocrine physiology and risk of disease.

Despite many advances in the last 75 years in the fight against cardiovascular disease mortality, it remains the leading cause of death worldwide. Despite improved therapeutic approaches, cardiovascular disease remains the leading cause of death worldwide. Mounting evidence suggests that restricting food intake to a limited period in the day or extending periods of fasting may be a lifestyle intervention that reduces progression of cardiovascular disease. Various protocols for time-restricted feeding paradigms suggest significant benefits throughout the cardiovascular system. Despite some limi

A novel fluorescent probe for sensitive detection copper in food samples and in vivo.

Copper (Cu2+) play crucial roles in both environmental and biological system, yet excessive levels can disrupt self-purification and accumulate in organisms, posing health risks. Herein, we developed a new fluorescent probe, HCY-Cl-Cu, for selective and sensitive detecting Cu2+. HCY-Cl-Cu exhibits a strong fluorescent response to copper ions, achieves a linear correlation range of 0-4 μM, low detection limit (LOD) 66 nM, fast reaction time of 10 min, alongside excellent selectivity. Furthermore, HCY-Cl-Cu displayed low cytotoxicity and successfully used to monitor Cu2+ within HepG2 cells and z

Near-Infrared Fluorescent Probe with a Large Stokes Shift for Detection of Hydrogen Sulfide in Food Spoilage, Living Cells, and Zebrafish.

Hydrogen sulfide (H2S) is a significant component of various physiological processes, and it can also cause a negative effect on foodstuffs. In this work, we designed and synthesized an NIR fluorescent turn-on responding probe (DDM-H2S) with a large Stokes shift (190 nm) for the detection of H2S. DDM-H2S exhibited high selectivity and sensitivity, obvious color changes, and a fast response time for tracing H2S. When DDM-H2S reacted with H2S, the PET process was eliminated, and the recovered ICT process and NIR fluorescence were observed. Moreover, DDM-H2S could image endogenous and exogenous H
