BACKGROUND: Current experimental models of pancreatic cancer either fail to reproduce the ductal phenotype or cause simultaneous cancers in other organs also. To develop an animal of pancreatic cancer that accurately mimics the human condition, we restricted carcinogenic exposure to the pancreas and specifically targeted ductal epithelial cells. Three different carcinogens were either implanted directly into the pancreas or infused into the pancreatic duct, with or without near-total pancreatectomy (as a means of inducing pancreatic ductal cell proliferation).

The coincidental radiological diagnosis following minor trauma or a few months' history of mild pain involving joints is characteristic. The disease process has a sudden onset. Nine patients with metastatic breast cancer were treated with a minimum of 6 X 10(6) U/day of beta-interferon (IFN-beta) for at least 6 weeks. In patients whose disease did not progress during this period treatment was continued to a maximum of 13 weeks, while in other patients doses were escalated. With daily treatments over 3 weeks the maximum tolerated dose was found to be around 60 X 10(6) U/day. Fever occurred regularly. From June 1978 to June 1990, 48 patients (34 patients with skull base tumors and 14 with congenital hyperterolism) were treated by combined craniofacial surgery. Cranial approaches were performed through the anterior fossa in 7 patients, middle fossa in 19, posterior fossa in 1, both anterior and middle fossa in 6, middle and posterior fossae in 1 and bilateral anterior fossae in 14.
