[Pathogenetic interrelations of hypogonadism and depression in men].

The study of the relationship between depression and functional hypogonadism in men is an interesting and difficult task. The purpose of the presented review was to identify the problem of functional male hypogonadism, to show a complex multi-stage neurohumoral mechanism of regulation of its potential to influence the affective sphere and to address the influence of pharmacotherapy on the formation of sexual disorders in men. The primary search for literature data was conducted using PubMed (Medline), PsycINFO and Cochrane CENTRAL, e-library.ru, databases using the keywords «hypogonadism», «de

Beyond the itch: the complex interplay of immune, neurological, and psychological factors in chronic urticaria.

Chronic urticaria (CU) arises from a multifaceted interplay of immunological, neurological, and psychological components. Immune dysregulation, mediated through both immunoglobulin E (IgE)-dependent and IgE-independent pathways, plays a pivotal role in CU pathogenesis, involving key effector cells such as mast cells (MCs), basophils, and eosinophils. This dysregulation culminates in the release of histamine, prostaglandins, and other mediators, which precipitate pruritus. The chronicity of the disease leads to sustained pruritic symptoms, contributing to both central and peripheral sensitizati

Small-molecule CBP/p300 histone acetyltransferase inhibition mobilizes leukocytes from the bone marrow via the endocrine stress response.

Mutations of the CBP/p300 histone acetyltransferase (HAT) domain can be linked to leukemic transformation in humans, suggestive of a checkpoint of leukocyte compartment sizes. Here, we examined the impact of reversible inhibition of this domain by the small-molecule A485. We found that A485 triggered acute and transient mobilization of leukocytes from the bone marrow into the blood. Leukocyte mobilization by A485 was equally potent as, but mechanistically distinct from, granulocyte colony-stimulating factor (G-CSF), which allowed for additive neutrophil mobilization when both compounds were co

Glucocorticoids and cognitive function: a walkthrough in endogenous and exogenous alterations.

PURPOSE: The hypothalamic-pituitary-adrenal (HPA) axis exerts many actions on the central nervous system (CNS) aside from stress regulation. Glucocorticoids (GCs) play an important role in affecting several cognitive functions through the effects on both glucocorticoid (GR) and mineralocorticoid receptors (MR). In this review, we aim to unravel the spectrum of cognitive dysfunction secondary to derangement of circulating levels of endogenous and exogenous glucocorticoids. METHODS: All relevant human prospective and retrospective studies published up to 2022 in PubMed reporting information on

The Hypothalamic-Pituitary-Thyroid Axis Equivalent in Normal and Cancerous Oral Tissues: A Scoping Review.

The hypothalamic-pituitary-thyroid (HPT) axis is crucial in regulating thyroid hormone levels that contribute to the development and homeostasis of the human body. Current literature supports the presence of a local HPT axis equivalent within keratinocytes of the skin, with thyroid hormones playing a potential role in cancer progression. However, this remains to be seen within oral tissue cells. An electronic search of Scopus and PubMed/Medline databases was conducted to identify all original publications that reported data on the production or effects of HPT axis components in normal or malig

Imaging the Hypothalamo-Neurohypophysial System.

The hypothalamo-neurohypophysial system (HNS) is a brain peptidergic neurosecretory apparatus which is composed of arginine vasopressin (AVP) and oxytocin (OXT) magnocellular neurones and their neuronal processes in the posterior pituitary (PP). In response to specific stimuli, AVP and OXT are secreted into the systemic circulation at the neurovascular interface of the PP, where they act as hormones, but they can also behave as neurotransmitters when released at the somatodendritic compartment or by axon collaterals to other brain regions. Because these peptides are crucial for several physiol

Stress and sex determination in fish: from brain to gonads.

Fish present remarkable malleability regarding gonadal sex fate. This phenotypic plasticity enables an organism to adapt to changes in the environment by responding with different phenotypes. The gonad and the brain present this extraordinary plasticity. These organs are involved in the response to environmental stressors to direct gonadal fate, inducing sex change or sex reversal in hermaphroditic and gonochoristic fish, respectively. The presence of such molecular and endocrine plasticity gives this group a large repertoire of possibilities against a continuously changing environment, result

Mechanisms of Central Hypogonadism.

Reproductive function depends upon an operational hypothalamo-pituitary-gonadal (HPG) axis. Due to its role in determining survival versus reproductive strategies, the HPG axis is vulnerable to a diverse plethora of signals that ultimately manifest with Central Hypogonadism (CH) in all its many guises. Acquired CH can result from any pituitary or hypothalamic lesion, including its treatment (such as surgical resection and/or radiotherapy). The HPG axis is particularly sensitive to the suppressive effects of hyperprolactinaemia that can occur for many reasons, including prolactinomas, and as a

Transcriptome analysis revealed the possible regulatory pathways initiating female geese broodiness within the hypothalamic-pituitary-gonadal axis.

Geese have the strongest tendency toward broodiness among all poultry. The mechanisms initiating broodiness within the goose hypothalamic-pituitary-gonadal axis (HPGA) are still unclear. Here, we reported the transcriptome differences between laying and initial nesting within the HPGA tissues of geese. We constructed a unigene database based on HPGA tissues and identified 128,148 unigenes, 100% of which have been annotated. By using Digital Gene Expression (DGE) sequencing, we screened 19, 110, 289, and 211 differentially expressed genes (DEGs) in the hypothalamus, pituitary gland, stroma ovar

Direct effects of stress on adrenocortical function.

The adrenal gland plays a pivotal role in the stress response since this response involves the hypothalamic-pituitary-adrenal axis (HPAA) and the sympatho-adrenomedullary system (SAMS) as its two principal components. An important relation between the immune system and the other stress response systems is also centered on the adrenal gland. It is well known that the cortex secretes glucocorticoids while the medulla secretes epinephrine, two of the major effects of the stress response. Some other aspects, however, also deserve special consideration: The paracrine effects of the cortical secreti

Stress and cardiovascular disease.

The statistical associations between stress and cardiovascular and other prevalent diseases have not been explained. Perceived stress, resulting in an uncontrollable defeat reaction, has been shown by James Henry (Henry 1993) to be followed by specific endocrine abnormalities, including sensitization of the hypothalamo-pituitary-adrenal (HPA) axis, and inhibited sex steroid and growth hormone secretions. With an elevated waist/hip circumference ratio (WHR)--a simple, surrogate, measurement of intraabdominal, visceral fat masses--combined with insulin resistance, similar endocrine perturbations

The potential role of excessive cortisol induced by HPA hyperfunction in the pathogenesis of depression.

Prolonged hypothalamic-pituitary-adrenocortical (HPA) axis overactivity occurs at all levels of this axis during stress in normals and some depressed patients. This can induce enlargement of the pituitary and adrenals. Various reports showed that cortisol can affect mood and behavior, and disrupt memory and recall. The integrity of the hippocampus is essential for memory function and, via the high density of its cortisol receptors, cortisol induced inhibitory feedback to the HPA axis. Animal data suggest that over time aging and stress can permanently downregulate hippocampal cell receptors, p

Aetiology and pathogenesis of growth hormone deficiency.

Growth hormone deficiency (GHD) represents an aetiologically non-uniform disorder: it can have multiple causes. Three main groups can be differentiated: (1) GHD due to manifest organic alterations of the hypothalamo-hypophyseal system; (2) so-called idiopathic cases; and (3) genetically determined forms. The first group comprises the congenital malformations, affecting the midline structures of the brain more often than the pituitary itself, and the acquired lesions, mainly tumours, which are generally suprasellar rather than intrasellar. The large majority of the so-called idiopathic cases re

Role of prolactin in the regulation of sensitivity of the hypothalamic-pituitary system to steroid feedback.

During sexual maturation, pituitary gonadotropins stimulate the gonads to produce increasing amounts of biologically active steroids and yet gonadotropin release does not become suppressed until concentrations of sex hormones, LH and FSH, in peripheral circulation stabilizes at a higher adult level. There is a substantial amount of evidence that in many mammals, this transition from prepubertal to adult level of activity of the pituitary-gonadal axis is associated with a reduction in the sensitivity of the hypothalamic-adenohypophyseal system to negative feedback of gonadal steroids. In the fe

Aminergic regulation of neuroendocrinological functions: theoretical background and some clinical examples.

Remarkable progress has been made during recent years in the central regulation of the hypothalamic releasing and inhibiting factors and the respective anterior pituitary hormones. There are two nearly universal inhibitory organizations: short tuberoinfundibular dopamine (TIDA) neurons and somatostatinergic system originating from the periventricular hypothalamus and terminating to the median eminence. It is now known that e.g. dopamine, noradrenaline and acetylcholine enhance while 5-hydroxytryptamine and GABA inhibit somatostatin secretion. These transmitters are also involved in the regulat

Hydrocephalus-induced central hypothalamic-pituitary-thyroid axis dysfunction: a conceptual framework linking ventricular distension to behavioral deficit.

BACKGROUND: Despite successful CSF diversion for pediatric obstructive hydrocephalus, up to 40% of children in Sub-Saharan Africa develop persistent neurodevelopmental delay, growth arrest, and executive dysfunction. Models based on axonal injury or ischemia alone do not fully explain this burden. HYPOTHESIS: We propose the Hypothalamic Compression-Endocrine-Glymphatic Dysfunction Theory. Third ventricular expansion compresses and shears periventricular hypothalamic neurons, particularly TRH-secreting cells of the paraventricular nucleus. This disrupts pulsatile TRH release and impairs TSH gl

Adrenal Insufficiency in Adults.

Adrenal insufficiency (AI) is a potentially life-threatening disease characterized by cortisol deficiency. The hypothalamic-pituitary-adrenal axis synthesizes and regulates cortisol secretion. Damage in any of these glands may cause AI. The 2 main types of AI include primary adrenal insufficiency, where the adrenal cortex itself is damaged and central adrenal insufficiency, where the hypothalamus and/or pituitary are damaged. This article reviews the physiology, diagnosis, and management of AI focusing on early disease recognition, improved diagnostic assessments, and stress dosing algorithms.
