On the use of inhibitors of 4-hydroxyphenylpyruvate dioxygenase as a vector-selective insecticide in the control of mosquitoes.

BACKGROUND: Blood-sucking insects incorporate many times their body weight of blood in a single meal. Because proteins are the major component of vertebrate blood, its digestion in the gut generates extremely high concentrations of free amino acids. Previous reports showed that the tyrosine degradation pathway plays an essential role in adapting these animals to blood feeding. Inhibition of 4-hydroxyphenylpyruvate dioxygenase (HPPD), the rate-limiting step of tyrosine degradation, results in the death of insects after a blood meal. Therefore, it has been suggested that compounds that block the

Effect of LongRange™ eprinomectin on Anopheles arabiensis by feeding on calves treated with the drug.

BACKGROUND: Misuse of long-lasting insecticidal nets together with resistance of vectors to most of the insecticides for indoor residual spraying and impregnated nets threaten malaria vector control interventions, requiring search for alternative control methods. Reports have shown that Anopheles mosquitoes die when they feed on endectocidal drugs used to treat humans and animals. A study was designed to investigate the efficacy of LongRange™ (eprinomectin 5%) on laboratory reared Anopheles arabiensis fed on treated calves. METHODS: Anopheles arabiensis from insectary colony was fed on three

Pharmacological characterization of a tyramine receptor from the southern cattle tick, Rhipicephalus (Boophilus) microplus.

The southern cattle tick (Rhipicephalus (Boophilus) microplus) is a hematophagous external parasite that vectors the causative agents of bovine babesiosis or cattle tick fever, Babesia bovis and B. bigemina, and anaplasmosis, Anaplasma marginale. The southern cattle tick is a threat to the livestock industry in many locations throughout the world. Control methods include the use of chemical acaricides including amitraz, a formamidine insecticide, which is proposed to activate octopamine receptors. Previous studies have identified a putative octopamine receptor from the southern cattle tick in

Scabies: molecular perspectives and therapeutic implications in the face of emerging drug resistance.

Limited effective treatments, coupled with recent observations of emerging drug resistance to oral ivermectin and 5% permethrin, raise concerns regarding the future control of scabies, especially in severe cases and in endemic areas where repeated community treatment programs are in place. There is consequently an urgent need to define molecular mechanisms of drug resistance in scabies mites and to develop and assess alternative therapeutic options, such as tea tree oil, in the event of increasing treatment failure. Molecular studies on scabies mites have, until recently, been restricted; howe

Acaricide resistance in northern fowl mite (Ornithonyssus sylviarum) populations on caged layer operations in Southern California.

Southern California caged layer operations were visited over 3 yr. Northern fowl mites from 26 field populations were tested for acaricide resistance using a capillary pipette and glass dish bioassay. One was a susceptible field population with no pesticide exposure for over 30 yr (reference site for resistance ratio calculation). Technical and commercial formulations of malathion, carbaryl (Sevin), permethrin, and a commercial formulation of tetrachlorvinphos/dichlorvos (Ravap) were tested. Malathion did not have high activity for mites relative to other materials, but resistance to both tech

Agrochemicals against malaria, sleeping sickness, leishmaniasis and Chagas disease.

In tropical regions, protozoan parasites can cause severe diseases with malaria, leishmaniasis, sleeping sickness, and Chagas disease standing in the forefront. Many of the drugs currently being used to treat these diseases have been developed more than 50 years ago and can cause severe adverse effects. Above all, resistance to existing drugs is widespread and has become a serious problem threatening the success of control measures. In order to identify new antiprotozoal agents, more than 600 commercial agrochemicals have been tested on the pathogens causing the above mentioned diseases. For a

Endocrine strategies for the control of ectoparasites and insect pests.

The increasing knowledge about endocrine mechanisms in arthropods facilitates the biorational search for drugs against insect pests and parasites that interfere with arthropod hormone action. Juvenile hormone mimics have been successfully applied for about 20 years; however, resistance to juvenile hormone analogues has developed. The introduction of moulting hormone agonists, which compete for binding to the ecdysteroid receptor, is expected in the near future. Despite the considerable progress that has been achieved in peptide hormone research during the last few years, no successful insectic

The black death past and present. 1. Plague in the 1980s.

This paper considers firstly the epidemiology of plague in the 1980s. The largest number of cases occurred in Tanzania. Most cases were in children and young adults; in the USA the male:female ratio was about 2:1. Plague had a seasonal distribution. Almost all cases arose from bites of infected rodent fleas, and Rattus spp. were the most important reservoir hosts. Virulence is linked with the presence of a 45 MDa plasmid. The predominant clinical form of plague is bubonic, followed by septicaemic, meningitic and pneumonic. For treatment, streptomycin is the antibiotic of choice, with tetracycl

Discovery of antiplasmodial pyridine carboxamides and thiocarboxamides.

Malaria continues to be a significant burden, particularly in Africa, which accounts for 95% of malaria deaths worldwide. Despite advances in malaria treatments, malaria eradication is hampered by insecticide and antimalarial drug resistance. Consequently, the need to discover new antimalarial lead compounds remains urgent. To help address this need, we evaluated the antiplasmodial activity of twenty-two amides and thioamides with pyridine cores and their non-pyridine analogues. Twelve of these compounds showed in vitro anti-proliferative activity against the intraerythrocytic stage of Plasmod

CRISPR-based gene drives generate super-Mendelian inheritance in the disease vector Culex quinquefasciatus.

Culex mosquitoes pose a significant public health threat as vectors for a variety of diseases including West Nile virus and lymphatic filariasis, and transmit pathogens threatening livestock, companion animals, and endangered birds. Rampant insecticide resistance makes controlling these mosquitoes challenging and necessitates the development of new control strategies. Gene drive technologies have made significant progress in other mosquito species, although similar advances have been lagging in Culex. Here we test a CRISPR-based homing gene drive for Culex quinquefasciatus, and show that the i…

Perspective on Biology and Management of Bed Bugs: Introduction.

Bed bugs are an important group of medical and urban insect pests. They are obligate blood-feeders. Their bites may cause skin irritation and allergic reactions and, under some circumstances, may lead to mental and other health issues. Despite numerous discoveries on the biology of these obnoxious pests and progress in control strategies over the last two decades, bed bugs continue to preferentially plague those from low socioeconomic communities because the poor generally could not afford effective control options. As a result, such infestations in poorer communities serve as a reservoir for

Saponarin, a Diglycosyl Flavone from the Ancestral Wheat Triticum monococcum, Is an Allelochemical Reducing the Survival of Cereal Aphids.

The English grain aphid (Sitobion avenae) threatens wheat production through direct feeding and indirectly as a virus vector (e.g., barley yellow dwarf virus - BYDV). Aphid management still relies predominantly on insecticides, an increasingly unsustainable practice; however, host-plant resistance is a promising solution to manage aphids and the viruses that they transmit. Resistance has been recently identified in the ancestor wheat Triticum monococcum MDR045 and MDR049 against S. avenae and Rhopalosiphum padi. In this study, we investigated whether foliar chemistry plays a role in this resis

Expression and regulation of CYP6D3 in the house fly, Musca domestica (L.).

Recently, a new cytochrome P450 gene, CYP6D3, was identified from house fly. CYP6D3 was found upstream of a related gene (CYP6D1) on autosome 1. CYP6D3 cDNA sequences were obtained and compared from insecticide resistant (LPR) and susceptible (CS and Edinburgh) strains. Although each strain had a different CYP6D3 allele, the deduced amino acid sequences revealed no consistent differences between the susceptible and resistant strains. There was approximately 12-fold more CYP6D3 mRNA detected in adult LPR flies compared to CS, and the elevated level of expression in LPR was not due to gene ampli

Breeding for disease resistance in Merino sheep in Australia.

Breeding for disease resistance in Merino sheep in Australia has attracted considerable research and development attention. Increased labour costs, the reduced efficacy of common anthelmintics and insecticides, consumer demand for products which are free of chemicals and the poor prognosis of alternative long-term control strategies are all forcing sheep breeders to contemplate the best animal health options available, including selective breeding for resistance. The three major diseases which affect sheep production include gastrointestinal nematode parasites, flystrike (cutaneous myiasis) an

Comprehensive Structural and Functional Characterization of a Seed γ-thionin as a Potent Bioactive Molecule Against Fungal Pathogens and Insect Pests.

BACKGROUND: Fungi and insect pests ruin stored crop grain, which results in millions of dollars of damage, presenting an ongoing challenge for farmers in addition to diminishing the safety of stored food. A wide-range defensive system against pathogens is needed to reduce or even eliminate the dependence of the crop yield upon the use of pesticides. Plant defensins (γ-thionins) are antimicrobial peptides (AMPs) that are a component of the host defense system. They are known to interact with cell membranes to exhibit antifungal and insecticidal activity. They exhibit a broad range of activities
