Hypothalamic hamartomata are benign malformations of the brain consisting of heterotopic nervous tissue, and are often associated with precocious puberty and gelastic seizures in early childhood. We report for the first time the melatonin plasma values of a girl with central precocious puberty and gelastic seizures due to a hypothalamic hamartoma. The melatonin plasma levels were low for the chronological age but appropriate for the pubertal status, making a causal relationship between lowered melatonin plasma levels and precocious puberty possible.

We studied the ultrastructural changes in gilts' pineal gland after the oral administration of melatonin in dose 3 mg once daily (in ethanol vehicle) for 46 consecutive days. Appearance of the pinealocytes containing numerous secretory granules and an enlarged Golgi apparatus was the most visible effect of the melatonin administration. The melatonin treatment caused also some differences in the structure of pinealocytes with cytoplasmic dense bodies. The present results suggest that the orally administered melatonin had some effects on pigs' pineal gland.

The pineal melatonin patterns were measured in rats kept 1, 2, 3, 4, 6, 8, 16, or 28 days under the lighting conditions with a dark period of 90 min only (from 18:00 to 19:30 h). The samples were collected at 30-min intervals, and melatonin was measured by radioimmunoassay. During the first and second dark pulse the melatonin levels were low, but thereafter they were significantly higher than the usual daytime levels. The increase of the synthesis started within 15 min after the light offset. The results suggest a stable adjustment of the melatonin rhythm by the dark pulses.

We show that the pineal melatonin synthesis of rats can be uncoupled from the circadian regulation by exposing the animals to abnormally long light periods. Male rats were kept 7 days under 22.5/1.5-h light/dark conditions and then exposed to darkness at different times of the day. The present study describes the development of a new technique to measure melatonin contents in the pineal gland of freely moving rats, by means of on-line microdialysis. The transcerebral cannula was modified, and a sensitive assay of melatonin, using HPLC with fluorimetric detection, was set up. Estrogen effects on pineal secretion of melatonin are controversial. Some feel that estrogen inhibits melatonin output in vitro but not in vivo. Melatonin levels vary with the age in chickens where circulating estrogen levels also vary. Laying hens have minimal melatonin levels and maximal serum iron concentrations. Thus, we reasoned that iron released by estrogen may inhibit melatonin secretion from the chick pineal gland. The present study shows that perifusion of estrogen-treated chick pineal glands with several concentrations of iron greatly inhibited melatonin secretion.

Binding of 2-[125I]melatonin to the anteroventral and anterodorsal nuclei of the rat limbic thalamus was analyzed with in vitro autoradiography. In both nuclei the binding was of high affinity (Kd values 24-41 pM) and was competed with by nanomolar concentrations of 6-hydroxymelatonin. Kd values were in the same range as those reported for other high-affinity binding sites in the brain. Bmax values were 0.68-2.99 fmol/mg protein. The results suggest that some effects of melatonin are mediated via the limbic thalamus.
