Thyroxine 5'-deiodination in brown adipose tissue and pineal gland: implications for thermogenic regulation and role of melatonin.

T4 type II 5'-deiodinase (5'-D II) activity was studied in wild-captured Richardson's ground squirrels. As previously reported for other species, 5'-D II activity was detected in frontal cortex, cerebellum, pineal gland, and brown adipose tissue (BAT); in the median eminence the levels of 5'-D II activity were undetectable with our methodology. When pineal gland, frontal cortex, and cerebellum nyctohemeral profiles were studied, none of them showed variations. Cold exposure for 4 h led to an increase in the enzymatic activity 10-fold above the basal values for BAT, while in the pineal gland th

[Dynamics of interleukin 2 production in patients with diffuse toxic goiter under the influence of treatment].

The paper is devoted to the results of a study of IL-2 production by T-helpers in the course of treatment of patients with diffuse toxic goiter (DTG); 30 patients and 30 healthy controls were investigated. The diagnosis of DTG was confirmed by modern methods including radioimmunoassays. A study of the blood level (relative and absolute) of T-lymphocytes in the patients before therapy, during 2 and 12 mos. of thimazol therapy and during remission of disease showed its significant decrease as compared to the controls. IL-2 production in the blood lymphocyte culture supernatant of the DTG patient

Pattern of Agreement Among Medications Used During Pregnancy as Recorded in Self-Report and Administrative Claims From California.

BACKGROUND: Medication usage prior to conception and over the duration of pregnancy is common. Two frequent sources that record such medications are self-report and administrative databases. Our goal was to examine the pattern of agreement among medications reported in each source. METHODS: We linked all interviewed subjects recruited for the Birth Defects Study To Evaluate Pregnancy exposureS (BD-STEPS) in California to their Medi-Cal insurance claims. BD-STEPS is a population-based case-control study including livebirths, stillbirths, and terminations with a major birth defect and livebirth

Association between maternal thyroid dysfunction and neurodevelopmental disorders in offspring: a population-based cohort study.

Attention-deficit/hyperactivity disorder (ADHD) and autism spectrum disorder (ASD) are both common neurodevelopmental disorders (NDDs), but their association between maternal thyroid dysfunction and NDDs in offspring remains equivocal. This study aimed to evaluate the association between maternal thyroid dysfunction, related medications, and neurodevelopmental disorders in offspring. This population-based retrospective birth cohort study included children born between 2004 and 2020, with surveillance continuing through 2021. Maternal thyroid dysfunction during pregnancy was identified by any o

Remission of Graves' Disease Through Lifestyle Interventions.

Graves' disease, caused by autoimmune thyrotropin receptor antibody-mediated activation of the thyroid, is characterized by hyperthyroidism, orbitopathy, dermopathy, and acropachy. Graves' disease is treated by anti-thyroid drug therapy, radioactive iodine ablation, or total thyroidectomy. We report the case of a 39-year-old female patient with hyperthyroidism secondary to Graves' disease that was managed through lifestyle interventions only. On presentation, she reported intermittent headaches and had an undetectable thyroid-stimulating hormone (TSH) level. Two weeks later, repeat testing sho

Autoimmune Implications in a Patient with Graves' Hyperthyroidism, Pre-eclampsia with Severe Features, and Primary Aldosteronism.

Background and Objectives: Graves' disease (GD) and primary aldosteronism (PA) are two pathologies that can cause significant morbidity and mortality. GD is mediated by autoantibodies, and recent studies have shown autoantibody involvement in the pathophysiology behind both PA and pre-eclampsia. The coexistence of GD and PA, however, is reportedly rare. This report describes a unique case of Graves' hyperthyroidism and concomitant PA in a patient with a history of pre-eclampsia with severe features. Case Presentation: The patient presented at 17 weeks pregnancy with mild hyperthyroidism, negat

Thyroid storm triggered by thyroid nodule fine needle aspiration biopsy: a case report.

BACKGROUND: Thyroid storm is a potentially fatal thyrotoxicosis triggered by an event, such as manipulation of the thyroid gland, acute iodine load, trauma, or infection. Prior to deciding on fine needle aspiration (FNA) biopsy, patients who have been diagnosed with hyperthyroidism or low thyroid stimulating hormone and multinodular goiter (MNG) should be imaged via radionuclide thyroid scan. CASE DESCRIPTION: We present a case of a 62-year-old female patient with history of MNG, who had thyrotoxicosis on presentation due to medication noncompliance and was found to have Graves' disease. Carbimazole-Resistant Grave's Thyrotoxicosis is a Diagnostic and Therapeutic Dilemma, Case Report with Literature Review.

BACKGROUND: Carbimazole (CBZ) (or methimazole) is the most used drug inducing and maintaining remission in thyrotoxicosis, especially Grave's disease (GD). Rarely, situations arise when patients do not respond to recommended or even supratherapeutic doses of CBZ. It poses a challenge to diagnose drug resistance and ultimately manage hyperthyroidism, which can otherwise be fatal if left untreated. Propylthiouracil (PTU) has been used as an alternative in such patients amid increased side effect risks. Additionally, definitive therapy has been recommended with ablation or surgery. However, the b

CYP450-mediated mitochondrial ROS production involved in arecoline N-oxide-induced oxidative damage in liver cell lines.

BACKGROUND: IARC has classified the betel nut as a human environmental carcinogen. Previous studies have found that arecoline (AR) is the major alkaloid present in the saliva of betel quid chewers. Saliva contains a large content of AR which has been further shown to cause mutation of oral mucosa cells, resulting in oral cancer. Whereas, to date, there are only few studies reported the hepatotoxicity associated with arecoline and betel nut chewing. Therefore, the main purpose of this study was to determine the toxic effects of AR and its oxidative metabolite, arecoline N-oxide (ARNO), in norma

Antithyroid Drugs and Congenital Malformations: A Nationwide Korean Cohort Study.

BACKGROUND: Untreated or insufficiently treated Graves disease in pregnancy may pose risks to both mother and fetus. Antithyroid drugs (ATDs) are the treatment mainstay, but the potential teratogenic effect of these drugs has prompted clinicians to question the safe management of this vulnerable population. OBJECTIVE: To examine the association between maternal prescriptions for ATDs and congenital malformations in live births. DESIGN: Nationwide cohort study. SETTING: Korean National Health Insurance database. PARTICIPANTS: A cohort of 2 886 970 completed pregnancies linked to live-born i

PREDICTION OF RELAPSE FROM HYPERTHYROIDISM FOLLOWING ANTITHYROID MEDICATION WITHDRAWAL USING TECHNETIUM THYROID UPTAKE SCANNING.

OBJECTIVE: Technetium thyroid uptake (TTU) is not inhibited by antithyroid drugs (ATD) and reflects the degree of thyroid stimulation. We intended to predict the relapse rate from hyperthyroidism based on TTU measurement. METHODS: Out of 44 initially enrolled subjects, 38 patients aged 41.6 ± 14.6 with Graves disease (duration: 84 ± 78 months) completed the study. TTU was performed with 40-second imaging of the neck and mediastinum 20 minutes after injection of 1 mCi technetium-99m pertechnetate. TTU was measured as the percentage of the count of activity accumulated in the thyroidal region m
