Evaluation of Aib and PEG-polymer insect kinin analogs on mosquito and tick GPCRs identifies potent new pest management tools with potentially enhanced biostability and bioavailability.

Insect kinins modulate aspects of diuresis, digestion, development, and sugar taste perception in tarsi and labellar sensilla in mosquitoes. They are, however, subject to rapid biological degradation by endogenous invertebrate peptidases. A series of α-aminoisobutyric (Aib) acid-containing insect kinin analogs incorporating sequences native to the Aedes aegypti mosquito aedeskinins were evaluated on two recombinant kinin invertebrate receptors stably expressed in cell lines, discovering a number of highly potent and biostable insect kinin mimics. On the Ae. aegypti mosquito kinin receptor, thr

Functional and antigenic maturation of Brugia malayi microfilariae.

Brugia malayi microfilariae of specified ages were obtained from gerbils implanted with fertile adult worms. Such microfilariae were tested for their capacity to infect mosquitoes. A strong age dependence was found for the microfilariae's capacity to: penetrate the mosquito midgut, exsheath in response to 20 mM calcium, and develop to third stage larvae in the mosquito. In addition, differences were found between 2-day-old microfilariae and controls (from larva-infected gerbils) in their reactivities with a series of monoclonal antibodies. Thus, defined immunochemical changes occur in microfil

Residual efficacy of standard and new-generation long-lasting insecticidal nets 3 years after mass distribution in Côte d'Ivoire.

BACKGROUND: Long-lasting insecticidal nets (LLINs) are essential for malaria control, but their effectiveness is threatened by increasing resistance of Anopheles to pyrethroids. In 2021, Côte d'Ivoire distributed both standard and new-generation LLINs during its mass distribution campaign. This study assessed the physical integrity and residual efficacy of these nets 3 y after their distribution. METHODS: A cross-sectional survey was conducted in 10 health districts chosen for ecological diversity. A total of 570 LLINs were withdrawn from randomly selected households and evaluated through WHO

Integrating multiplexing into confineable gene drives effectively overrides resistance in Anopheles stephensi.

Anopheles stephensi is a major malaria vector mainly present in southern Asia and the Arabian Peninsula. Since 2012 it has invaded several countries of eastern Africa, stimulating urgent efforts to develop more efficient strategies for vector control such as CRISPR/Cas9-based homing gene drives. Target site resistance due to end-joining repair is a significant challenge to the deployment of these systems. The use of multiple sgRNAs has the potential to solve this issue. Here we perform experimental crosses to assess the homing and cutting efficiency of both classical (e.g. four adjacent sgRNAs

Impact of insecticide resistance evolution on malaria vector control.

BACKGROUND: Insecticide-treated nets (ITNs) are the staple of malaria vector control. Given their importance, insecticide resistance (IR) is extremely concerning. Understanding how ITNs simultaneously impact the evolution of IR and malaria control is of critical importance, especially with next-generation mixture ITNs (NGM-ITNs) becoming available. METHOD: We developed a mosquito population dynamics and genetics model to explore the simultaneous impact of ITNs on both malaria transmission (measured as vectorial capacity, an entomological measure of transmission) and IR evolution (resistance g

Community participation and operational lessons from implementing a protective coating as a novel vector control tool in dengue-endemic neighborhoods of Cúcuta and Los Patios, Colombia.

BACKGROUND: The control of Aedes-borne diseases (dengue, Zika, chikungunya, yellow fever) is a high public health priority in tropical and subtropical countries. The adoption of new vector control tools, such as protective coating (PC), requires understanding the social dynamics that influence their acceptance and feasibility. This study investigated the experiences and perspectives of diverse stakeholders involved in a vector control project in urban Colombia, with the aim of assessing community acceptance and operational feasibility. Insights gained are important for optimizing implementatio

Do monetary incentives encourage local communities to collect and upload mosquito sound data using smartphones? A case study in the Democratic Republic of the Congo.

Malaria is one of the deadliest vector borne diseases affecting sub-Saharan Africa. A suite of systems are being used to monitor and manage malaria risk and disease incidence, with an increasing focus on technological interventions that allow private citizens to remotely record and upload data. However, data collected by citizen scientists must be standardised and consistent if it is to be used for scientific analysis. Studies that aim to improve data collection quality and quantity have often included incentivisation, providing citizen scientists with monetary or other benefits for their part

The experimental hut efficacy of next-generation insecticide-treated nets against pyrethroid-resistant malaria vectors after 12, 24 and 36 months of household use in Benin.

BACKGROUND: Cluster-randomized controlled trials (cluster-RCTs) have demonstrated variation in the epidemiological efficacy of different next-generation insecticide-treated net (ITN) types, with some providing shorter-lived impact than others. Further studies are needed to assess changes in the insecticidal durability of these ITNs over time to complement cluster-RCT results. METHODS: A series of experimental hut trials were performed to evaluate the bioefficacy of new and field-aged next-generation ITNs (PermaNet® 3.0, Royal Guard®, Interceptor® G2) compared to a pyrethroid-only net (Interce

Assessing Wolbachia-mediated sterility for dengue control: emulation of a cluster-randomized target trial in Singapore.

BACKGROUND: Matings between male Aedes aegypti mosquitoes infected with wAlbB strain of Wolbachia and wildtype females yield non-viable eggs. We evaluated the efficacy of releasing wAlbB-infected Ae. aegypti male mosquitoes to suppress dengue. METHODS: We specified the protocol of a two-arm cluster-randomized test-negative controlled trial (cRCT) and emulated it using a nationally representative test-negative/positive database of individuals reporting for febrile illness to any public hospital, general practitioner or polyclinic. We retrospectively built a cohort of individuals who reside in

Sub-lethal exposure to chlorfenapyr reduces the probability of developing Plasmodium falciparum parasites in surviving Anopheles mosquitoes.

BACKGROUND: Pyrethroid resistance in the key malaria vectors threatens the success of pyrethroid-treated nets. To overcome pyrethroid resistance, Interceptor® G2 (IG2), a 'first-in-class' dual insecticidal net that combines alpha-cypermethrin with chlorfenapyr, was developed. Chlorfenapyr is a pro-insecticide, requiring bio-activation by oxidative metabolism within the insect's mitochondria, constituting a mode of action preventing cross-resistance to pyrethroids. Recent epidemiological trials conducted in Benin and Tanzania confirm IG2's public health value in areas with pyrethroid-resistant

Blood-feeding patterns of Anopheles vectors of human malaria in Malawi: implications for malaria transmission and effectiveness of LLIN interventions.

BACKGROUND: Access to human hosts by Anopheles mosquitoes is a key determinant of vectorial capacity for malaria, but it can be limited by use of long-lasting insecticidal nets (LLINs). In Malawi, pyrethroid-treated LLINs with and without the synergist piperonyl butoxide (PBO) were distributed to control malaria. This study investigated the blood-feeding patterns of malaria vectors and whether LLINs containing pyrethroid and PBO led to a reduction of human blood feeding than those containing only pyrethroids. METHODS: Mosquitoes were sampled inside houses from May 2019 through April 2020 by a

Observing the distribution of mosquito bites on humans to inform personal protection measures against malaria and dengue vectors.

BACKGROUND: Understanding mosquito biting behaviours is important for designing and evaluating protection methods against nuisance biting and mosquito-borne diseases (e.g. dengue, malaria and zika). We investigated the preferred biting sites by Aedes aegypti and Anopheles arabiensis on adult volunteers in standing or sleeping positions; and estimated the theoretical protection limits affordable from protective clothing or repellent-treated footwear. METHODS: Adult volunteers dressed in shorts and t-shirts were exposed to infection-free laboratory-reared mosquitoes inside screened chambers fro

Investigating the blood-host plasticity and dispersal of Anopheles coluzzii using a novel field-based methodology.

BACKGROUND: The biting behaviour and dispersal of insect vectors in the field underlies the transmission of many diseases. Here, a novel collection methodology coupled with the molecular analysis of blood-meal sources and digestion rates is introduced with the aim of aiding the understanding of two critical and relatively understudied mosquito behaviours: plasticity in blood-host choice and vector dispersal. RESULTS: A collection strategy utilising a transect of mosquito traps placed at 50 m intervals allowed the collection of blood-fed Anopheles coluzzii from a malaria-endemic village of sou

Host Decoy Trap (HDT) with cattle odour is highly effective for collection of exophagic malaria vectors.

BACKGROUND: As currently implemented, malaria vector surveillance in sub-Saharan Africa targets endophagic and endophilic mosquitoes, leaving exophagic (outdoor blood-feeding) mosquitoes underrepresented. We evaluated the recently developed host decoy trap (HDT) and compared it to the gold standard, human landing catch (HLC), in a 3 × 3 Latin square study design outdoors in western Kenya. HLCs are considered to represent the natural range of Anopheles biting-behaviour compared to other sampling tools, and therefore, in principle, provide the most reliable profile of the biting population trans

Assessment of Resistance to Organophosphates and Pyrethroids in Aedes aegypti (Diptera: Culicidae): Do Synergists Affect Mortality?

Aedes aegypti (L.) is the primary vector of Zika, dengue, yellow fever, and chikungunya viruses. Insecticides used in mosquito control can help prevent the spread of vector-borne diseases. However, it is essential to determine insecticide resistance (IR) status before control measures are undertaken. Only the most effective insecticides should be used to avoid ineffective control and/or promotion of IR. Pyrethroids and organophosphates are the most commonly used insecticides for mosquito control. Here, the efficacy of two active ingredients (AIs; permethrin [pyrethroid], chlorpyrifos [organoph…

Whole-mount in situ hybridization for analysis of gene expression during Aedes aegypti development.

Blood-feeding mosquitoes, including the dengue and yellow fever vector Aedes aegypti, transmit many of the world's deadliest diseases. Such diseases have resurged in developing countries and pose clear threats for epidemic outbreaks in developed countries. Recent mosquito genome projects have stimulated interest in the potential for arthropod-borne disease control by genetic manipulation of vector insects, and genes that regulate development are of particular interest. This protocol for whole-mount in situ hybridization can be used to analyze gene expression in Ae. aegypti embryos and larvae,

Knockdown of the Sodium/Potassium ATPase Subunit Beta 2 Reduces Egg Production in the Dengue Vector, Aedes aegypti.

The Na+/K+ ATPase (NKA) is present in the cellular membrane of most eukaryotic cells. It utilizes energy released by ATP hydrolysis to pump sodium ions out of the cell and potassium ions into the cell, which establishes and controls ion gradients. Functional NKA pumps consist of three subunits, alpha, beta, and FXYD. The alpha subunit serves as the catalytic subunit while the beta and FXYD subunits regulate the proper folding and localization, and ion affinity of the alpha subunit, respectively. Here we demonstrate that knockdown of NKA beta subunit 2 mRNA (nkaβ2) reduces fecundity in female A

Bioactive Molecules Derived from Plants in Managing Dengue Vector Aedes aegypti (Linn.).

Mosquitoes are the potential vectors of several viral diseases such as filariasis, malaria, dengue, yellow fever, Zika fever and encephalitis in humans as well as other species. Dengue, the most common mosquito-borne disease in humans caused by the dengue virus is transmitted by the vector Ae. aegypti. Fever, chills, nausea and neurological disorders are the frequent symptoms of Zika and dengue. Thanks to various anthropogenic activities such as deforestation, industrialized farming and poor drainage facilities there has been a significant rise in mosquitoes and vector-borne diseases.
