Differential hepatoprotective mechanisms of rutin and quercetin in CCl(4)-intoxicated BALB/cN mice.

AIM: To investigate the mechanisms underlying the protective effects of quercetin-rutinoside (rutin) and its aglycone quercetin against CCl(4)-induced liver damage in mice. METHODS: BALB/cN mice were intraperitoneally administered rutin (10, 50, and 150 mg/kg) or quercetin (50 mg/kg) once daily for 5 consecutive days, followed by the intraperitoneal injection of CCl(4) in olive oil (2 mL/kg, 10% v/v). The animals were sacrificed 24 h later. Blood was collected for measuring the activities of ALT and AST, and the liver was excised for assessing Cu/Zn superoxide dismutase (SOD) activity, GSH an

Reactions of the NO redox forms NO+, *NO and HNO (protonated NO-) with the melatonin metabolite N1-acetyl-5-methoxykynuramine.

The different NO redox forms, NO+, *NO and HNO (=protonated NO-), were compared for their capabilities of interacting with the melatonin metabolite N1-acetyl-5-methoxykynuramine (AMK), using NO+SbF6-, PAPA-NONOate and Angeli's salt as donors of the respective NO species. Particular attention was paid to stability and possible interconversions of the redox forms. *NO formation was followed by measuring the decolorization of 2-(trimethylammonio-phenyl)-4,4,5,5-tetramethyl-imidazoline-1-oxyl-3-oxide (TMA-PTIO), at different pH values, at which NO+ is, in aqueous solution, either highly unstable (

Oxidative chemistry of nitric oxide: the roles of superoxide, peroxynitrite, and carbon dioxide.

The roles of superoxide (O2.-), peroxynitrite, and carbon dioxide in the oxidative chemistry of nitric oxide (.NO) are reviewed. The formation of peroxynitrite from .NO and O2.- is controlled by superoxide dismutase (SOD), which can lower the concentration of superoxide ions. The concentration of CO2 in vivo is high (ca. 1 mM), and the rate constant for reaction of CO2 with -OONO is large (pH-independent k = 5.8 x 10(4) M(-l)s(-1)). Consequently, the rate of reaction of peroxynitrite with CO2 is so fast that most commonly used scavengers would need to be present at very high, near toxic levels

Epigenetic regulation of memory by acetylation and methylation of chromatin: implications in neurological disorders, aging, and addiction.

Synaptic plasticity is one of the most fundamental properties of neurons that underlie the formation of the memory in brain. In recent years, epigenetic modification of both DNA and histones such as DNA methylation and histone acetylation and methylation emerges as a potential regulatory mechanism that governs the transcription of several genes responsible for memory formation and behavior. Furthermore, the recent identification of nitrosylation of proteins has shown to either activate or repress gene transcription by modulating histone methylation or acetylation status in mature neuron. Nitrosatable drug exposure during the first trimester of pregnancy and selected congenital malformations.

BACKGROUND: Nitrosatable drugs can react with nitrite in the stomach to form N-nitroso compounds, and results from animal studies suggest that N-nitroso compounds are teratogens. With data from the National Birth Defects Prevention Study, the relation between prenatal exposure to nitrosatable drugs and limb deficiencies, oral cleft, and heart malformations in offspring was examined. METHODS: Maternal reports of drugs taken during the first trimester of pregnancy were classified with respect to nitrosatability for mothers of 741 babies with limb deficiencies, 2774 with oral cleft malformations

S-Nitrosylation of Akt by organic nitrate delays revascularization and the recovery of cardiac function in mice following myocardial infarction.

The effects of long-term nitrate therapy are compromised due to protein S-Nitrosylation, which is mediated by nitric oxide (NO). This study is to determine the role of Akt S-Nitrosylation in the recovery of heart functions after ischaemia. In recombinant Akt protein and in HEK293 cells, NO donor decreased Akt activity and induced Akt S-Nitrosylation, but was abolished if Akt protein was mutated by replacing cysteine 296/344 with alanine (Akt-C296/344A). In endothelial cells, NO induced Akt S-Nitrosylation, reduced Akt activity and damaged multiple cellular functions including proliferation, mi

Study of the effect of thiols on the vasodilatory potency of S-nitrosothiols by using a modified aortic ring assay.

Both low-molecular-mass thiols (LMM-SH) and protein thiols (P-SH) can modulate the biological activity of S-nitrosothiols (RSNO) via S-transnitrosation reactions. It has been difficult to evaluate the entity of this effect in blood circulation by in vitro assays with isolated aorta rings so far, because media rich in proteins cannot be used due to the foaming as a consequence of the needed gas bubbling. We have modified the original apparatus for organ bioassay in order to minimize foaming and to increase analytical performance. By using this modified bioassay we investigated the vasodilatory …

Size-Dependent Zinc Delivery and Nanozyme-Mediated Stress Alleviation by ZnO Nanoparticles in Zinc-Deficient Tomato.

Zinc (Zn) deficiency severely limits plant growth and yield, highlighting the need for strategies to improve Zn nutrition. Here, Zn oxide nanoparticles (nZnOs) of two sizes (∼8 nm and ∼45 nm, 100 mg·L-1) were evaluated as seed priming agents in tomato grown under Zn-deficient conditions. Both nanoparticle sizes were internalized into embryonic tissues and showed distinct intracellular localization. While 45 nm nZnO and bulk Zn disturbed seed nutrient homeostasis, 8 nm nZnO increased seed Zn content more efficiently, with milder effects on other nutrients. Across developmental stages, nZnO prim

Nitric Oxide, Reactive Oxygen Species, and Focal Adhesion Kinase Mediate Anoikis Resistance in A375 and SK-MEL-28 Human Melanoma Cells.

Melanoma is a highly aggressive and invasive form of skin cancer that arises from the uncontrolled growth of melanocytes. It is characterized by early spread through the lymphatic system and metastasis. The success of metastasis is linked to the ability of melanoma and other cancer cells to resist anoikis, a type of cell death that occurs when cells lose their adhesion to the extracellular matrix. Redox signaling plays an essential role in anoikis resistance. The balance between intracellular levels of nitric oxide (NO) and the reactive oxygen species (ROS) O2- and H2O2 stimulate signaling pat

Hesperetin-Loaded PLGA Nanoparticles Ameliorate Cisplatin-Induced Oxidative Stress and Testicular Dysfunction in Rats: Association with NRF2/HO-1, NF-κB, and ACSL4/GPX4/SLC7A11 Signaling Modulation.

BACKGROUND: Cisplatin is a widely used chemotherapeutic agent whose gonadotoxic effects, largely driven by oxidative stress and inflammation, pose a major threat to male reproductive health. This study evaluated whether hesperetin (HES) and hesperetin-loaded poly (lactic-co-glycolic acid) nanoparticles (HES-PLGA-NPs) can protect against cisplatin-induced testicular dysfunction in adult male Sprague Dawley rats. METHODS: Sixty rats were randomly assigned to six groups: control, HES, HES-PLGA-NPs, cisplatin (CIS), CIS + HES, and CIS + HES-PLGA-NPs. CIS (7.5 mg/kg/week, i.p.) was given intraperi

Microbiota metabolite lithocholic acid in cancer: Mechanisms and therapeutic potential.

The human gut microbiome is centrally implicated in host disease and health control via the production of a range of bioactive metabolites. Of these, lithocholic acid (LCA), a derivative of microbial metabolism secondary bile acid, has received attention owing to its multifaceted functions in cancer. LCA, a hydrophobic microbiota-derived bile acid, was implicated in carcinogenicity and hepatotoxicity even in the ancient era. In this sense, studies have found a contrasting role of LCA as a tumor promoter or tumor suppressor based on the molecular environment and type of tumor. Antitumor effects

Determining biocide efficacy for treating established sulfate-reducing biofilms using flow cell systems.

Sulfate-reducing microorganisms (SRM) can contribute to souring and to the corrosion of infrastructure built to support many industrial operations, including in aquatic environments. While chemicals such as biocides can effectively treat planktonic cells, less is known about biocide efficacy for treating established biofilms potentially plaguing infrastructure. We used a biofilm flow cell system to examine the efficacy of sodium nitroprusside (SNP, a nitrosating compound proposed as a "green" biocide) and alkyl dimethyl benzyl ammonium chloride (ADBAC), a membrane-disrupting biocide used acros

Interaction between purinergic signalling and purinergic modulators with skin wound repair effectors: a systematic review of preclinical in vivo models.

Purinergic signalling plays a central regulatory role on cell proliferation, migration, differentiation, survival, and inflammation. However, the impact of purinergic modulators-based therapies on tissue repair is still overlooked. By using a systematic review framework, we investigated the therapeutic relevance of purinergic agonists and antagonists in animal models of skin wound healing. Following PRISMA guidelines, PubMed/Medline, Scopus, Web of Science, Embase, and CINAHL databases were searched, identifying 12 in vivo preclinical studies. Risk of bias was assessed using the SYRCLE tool. T

In Vitro Studies on Induction of Ferroptosis Upon Low-level Exposure of Bisphenol A on Rat Erythrocytes.

Bisphenol A (BPA), is a xenoestrogen used in plastic manufacturing companies. Some countries are limiting the use of BPA. The widespread use of BPA has resulted in its accumulation in the natural environment. This compound is extensively utilised in various industrial products and is linked to harmful effects on both wildlife and human health. In recent years, extensive research has focused on ferroptosis-an iron-dependent form of programmed cell death characterised by lipid peroxidation. Also, several studies highlight the involvement of ferroptosis in mediating the toxic effects of environme

Eucalyptol Ameliorates Neuroendocrine Stress-Aggravated Rheumatoid Arthritis Through Anti-inflammatory, Antioxidant, and Chondroprotective Mechanisms Associated with NF-κB and MAPK Signaling Pathways.

BACKGROUND: Rheumatoid arthritis (RA) is a lingering autoimmune disorder typified by synovial inflammation, oxidative imbalance, and advanced articular damage. Psychological and physical stress are known to exacerbate disease progression by enhancing inflammatory signaling and disrupting redox homeostasis. Although methotrexate remains a first-line therapy, its long-term use is accompanied by adverse effects, necessitating safer adjunctive strategies. Eucalyptol (1,8-cineole), a naturally occurring monoterpene, has demonstrated anti-inflammatory and antioxidant potential; however, its role in

Diazinon-induced oxidative stress and organ toxicity in rodent models: a narrative review of mechanistic pathways, biomarkers and antioxidant interventions.

Diazinon (DZN) is a widely used organophosphorus insecticide whose toxicity has traditionally been attributed to acetylcholinesterase inhibition after metabolic activation to diazoxon. However, a growing body of rodent evidence indicates that this mechanism alone does not adequately explain the breadth and persistence of DZN-induced organ injury, particularly under subacute and subchronic exposure conditions. This narrative review synthesizes current evidence on the toxicokinetic, mechanistic and organ-specific basis of DZN-induced oxidative stress in rodent models, with emphasis on biomarker

Effects of Diazepam on Oxidative Stress, Apoptosis, and Vascular Reactivity in Human Umbilical Arteries Under Experimental Hyperhomocysteinemia.

In obstetrics, diazepam is commonly used to treat anxiety, insomnia and as a second-line treatment for seizures in preeclampsia and eclampsia. However, the effects of diazepam on the oxidative balance and vascular function of the umbilical circulation are still insufficiently studied. In this study, we investigated the effects of suprapharmacological concentrations of diazepam in an in vitro model of human umbilical artery (HUA) injury, induced by incubation with 1 mM homocysteine, representing experimental conditions that mimic hyperhomocysteinemia. HUAs were divided into four groups: control

Bolus MPTP Injection in Aged Mice to Mimic Parkinson Disease: Effects of Low-Dose Antioxidant Treatment with Fullerene (C60) and Fullerenol (C60(OH)24).

Background: Parkinson's disease (PD) is a neurodegenerative disorder for which no curative therapies currently exist. Experimental models employing 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) reproduce PD features such as striatal dopaminergic dysfunction and motor deficits. Various MPTP dosing regimens are used to screen drug candidates for PD, but their validity is limited because of the predominant use of young male animals. Sex bias is another issue that is underrepresented in PD research, since females are more susceptible to this pathology. Here, we studied the model of bolus adm
