Phytotherapeutic Frontiers: A Comprehensive Review of Medicinal Plants as Adjuncts in COVID-19 Management.

Unexpected findings emerge when nature encounters pandemic challenges. From mountain slopes, remedies arise shaped by harsh climates. Such conditions foster strong chemical defenses in plants. One such defense disrupts a key virus enzyme, similar to lab-made drugs. Instead of only attacking germs, these substances also calm body reactions gone awry. They can block the spike protein's interaction with human cells while reducing excessive immune signals associated with severe illness. Structural features within these compounds may explain why some are more effective than others. Tiny carriers de

The transcription factor BnaWRKY75 enhances resistance to the necrotrophic pathogen Sclerotinia sclerotiorum by promoting salicylic acid biosynthesis in oilseed rape.

Sclerotinia sclerotiorum, a devastating necrotrophic fungus causing stem rot in oilseed rape, results in significant global yield losses. Deciphering plant-S. sclerotiorum interactions is crucial for disease control. Here, we report that the transcription factor BnaWRKY75 positively regulates resistance to this pathogen. BnaWRKY75 was the most highly induced WRKY gene upon S. sclerotiorum infection. Overexpressing BnaA10.WRKY75 in both Arabidopsis and oilseed rape enhanced resistance to S. sclerotiorum, whereas BnaWRKY75 knockout plants exhibited reduced resistance. RNA sequencing and DNA affi

Selected Indian Medicinal Plants Exhibit Anti-Inflammatory Activity by Modulating Pro-Inflammatory Cytokines In Vitro and in Carrageenan-Induced Rat Paw Edema.

Inflammation is a vital immune response to injury, infection, or harmful stimuli, and prolonged inflammation leads to various disorders. Traditional medicinal systems such as Ayurveda offer a rich repository of plant-based remedies for inflammatory conditions. In this study, 18 medicinal plants were screened for anti-inflammatory potential using in vitro and in vivo assays. Methanolic extracts and solvent-partitioned fractions were evaluated for cytotoxicity, nitric oxide (NO) inhibition, pro-inflammatory cytokines (IL-1β, IL-6, tumor necrosis factor α [TNF-α]), nuclear factor κB (NF-κB), and

Melatonin-Nitric Oxide Crosstalk in Plants and the Prospects of NOMela as a Nitric Oxide Donor.

Melatonin regulates vital physiological processes in animals, such as the circadian cycle, sleep, locomotion, body temperature, food intake, and sexual and immune responses. In plants, melatonin modulates seed germination, longevity, circadian cycle, photoperiodicity, flowering, leaf senescence, postharvest fruit storage, and resistance against biotic and abiotic stresses. In plants, the effect of melatonin is mediated by various regulatory elements of the redox network, including RNS and ROS. Similarly, the radical gas NO mediates various physiological processes, like seed germination, flower

CRISPR/Cas9-mediated simultaneous mutation of three salicylic acid 5-hydroxylase (OsS5H) genes confers broad-spectrum disease resistance in rice.

Salicylic acid (SA) is an essential plant hormone that plays critical roles in basal defence and amplification of local immune responses and establishes resistance against various pathogens. However, the comprehensive knowledge of the salicylic acid 5-hydroxylase (S5H) in rice-pathogen interaction is still elusive. Here, we reported that three OsS5H homologues displayed salicylic acid 5-hydroxylase activity, converting SA into 2,5-dihydroxybenzoic acid (2,5-DHBA). OsS5H1, OsS5H2, and OsS5H3 were preferentially expressed in rice leaves at heading stage and responded quickly to exogenous SA trea

Plant-derived VLP: a worthy platform to produce vaccine against SARS-CoV-2.

After its emergence in late 2019 SARS-CoV-2 was declared a pandemic by the World Health Organization on 11 March 2020 and has claimed more than 2.8 million lives. There has been a massive global effort to develop vaccines against SARS-CoV-2 and the rapid and low cost production of large quantities of vaccine is urgently needed to ensure adequate supply to both developed and developing countries. Virus-like particles (VLPs) are composed of viral antigens that self-assemble into structures that mimic the structure of native viruses but lack the viral genome. Thus they are not only a safer altern

In vitro antiviral activity of peptide-rich extracts from seven Nigerian plants against three non-polio enterovirus species C serotypes.

BACKGROUND: As frequent viral outbreaks continue to pose threat to public health, the unavailability of antiviral drugs and challenges associated with vaccine development underscore the need for antiviral drugs discovery in emergent moments (endemic or pandemic). Plants in response to microbial and pest attacks are able to produce defence molecules such as antimicrobial peptides as components of their innate immunity, which can be explored for viral therapeutics. METHODS: In this study, partially purified peptide-rich fraction (P-PPf) were obtained from aqueous extracts of seven plants by rev

Biological mechanisms of glycan- and glycosaminoglycan-based nutraceuticals.

A nutraceutical is defined as a standardized pharmaceutical-grade nutrient. Among hundreds of nutraceuticals, polysaccharide or glycan-based products such as those containing chondroitin sulfate glycosaminoglycan isolated from animal cartilage have been on the top nutraceutical selling list for many years. It is expected that the nutraceutical market will reach $250 billion dollars worldwide by 2018. Glycans are most abundant biopolymers on earth those are synthesized by bacteria, fungi, plants, insects, and animals. Glycans that are synthesized by animals or from all marine sources can be mod

Ecological effects, transport, and fate of mercury: a general review.

Mercury at low concentrations represents a major hazard to microorganisms. Inorganic mercury has been reported to produce harmful effects at 5 microg/l in a culture medium. Organomercury compounds can exert the same effect at concentrations 10 times lower than this. The organic forms of mercury are generally more toxic to aquatic organisms and birds than the inorganic forms. Aquatic plants are affected by mercury in water at concentrations of 1 mg/l for inorganic mercury and at much lower concentrations of organic mercury. Aquatic invertebrates widely vary in their susceptibility to mercury. I

Biogenic Caralluma sinaica-derived silver nanoparticles as a synergistic antibacterial and osteoinductive nanoplatform for osteomyelitis management.

INTRODUCTION: Osteomyelitis is an inflammatory bone disease caused by infectious microorganisms. Challenges in treating chronic osteomyelitis include antibiotic resistance, inadequate immune response, and the need for surgical debridement. As an alternative treatment strategy, we developed a formulation of green phytosynthesized silver nanoparticles (AgNPs) and plant extract, as a novel multifunctional drug to achieve antibacterial and osteogenic activities in osteomyelitis. METHODS: Green silver nanoparticles (CS-AgNPs) were successfully phytosynthesized using Caralluma sinaica methanolic le

Bioactive compounds in the modulation of oxidative stress in monocytes and macrophages.

Human immunity involves both innate and adaptive defence mechanisms, with inflammation playing a central role in responding to cellular injury, pathogenic infections, and allergic stimuli. Reactive oxygen species (ROS) are closely associated with the onset and progression of inflammation. While moderate ROS levels function as crucial signalling molecules, excessive ROS can damage cellular components. This study aimed to evaluate the anti-inflammatory and antioxidant potential of plant-derived bioactive compounds including chlorogenic acid, oleuropein, tomatine, and tyrosol using human monocyti

Health effects of mixed fruit and vegetable concentrates: a systematic review of the clinical interventions.

Diets rich in fruits and vegetables (FV) have been associated with a reduced risk of chronic disease, including cardiovascular disease. Unfortunately, public health campaigns to increase FV intake have had limited success. A number of mixed concentrated FV products have been studied, which may help certain individuals improve nutrient status. However, the possible health benefits of FV supplements have not been systematically reviewed. We, therefore, undertook a systematic search of MEDLINE and EMBASE to identify clinical interventions that examined the effect of commercially available concent

Tripartite Interaction Between Penicillium pinophilum-Host Plants and CMV-Y: A Model for Endophyte-Mediated Viral Biocontrol.

Plant-fungus-virus tripartite interactions represent complex ecological systems in which mutualistic endophytes can influence host physiology, yet the molecular basis of endophyte-mediated defence remains poorly understood. Here, we demonstrate that the endophytic fungus Penicillium pinophilum EU0013 suppresses the yellow strain of cucumber mosaic virus (CMV-Y) in Solanum lycopersicum and Nicotiana benthamiana. CMV-Y infection induced pronounced oxidative and hormonal perturbations, which were mitigated by P. pinophilum colonisation. Endophyte-associated plants exhibited early accumulation of

AraVirusPPI: A protein language model-powered method for predicting Arabidopsis thaliana-virus protein-protein interactions.

Plant-virus protein-protein interactions (PPIs) play crucial roles in viral infection and host immune responses. However, large-scale experimental characterization of plant-virus PPIs is costly and labor-intensive, suggesting the need to develop computational methods. Recent advances in biological sequence language models are offering unprecedented opportunities to develop machine learning-based plant-virus PPI predictors. In this study, we collected publicly available PPI data between Arabidopsis thaliana (A. thaliana) and viruses, establishing a robust dataset for method development. Based o

Prevention and Treatment of Staphylococcus aureus Biofilms Using Promising Agr-QS-Targeting Anti-Biofilm Agents.

Staphylococcus aureus (S. aureus), a leading cause of nosocomial infections, contributes significantly to increased morbidity and mortality, especially when it forms biofilms on medical devices. This pathogen, specifically methicillin-resistant S. aureus (MRSA), remains a challenge to treat due to its ability to form biofilms and rapidly develop resistance against antibiotics. Biofilm formation allows bacteria to adhere to biotic and abiotic surfaces, creating a protective matrix that shields them from immune responses and antibiotic therapies. The widespread prevalence of multidrug-resistant

Activation of the ChvG-ChvI pathway promotes survival during cell wall stress in Agrobacterium tumefaciens.

Agrobacterium tumefaciens shifts from a free-living soil bacterium to a plant-invading state upon encountering the plant root microenvironment. The acid-induced two-component sensor system ChvG-ChvI drives this shift and triggers a complex transcriptional program that promotes host invasion and survival against host immune defenses. Remarkably, ChvG-ChvI is also activated under cell wall stress conditions, suggesting that the transcriptional response may have a broader function. Here, we find that blocking cell wall synthesis either genetically or chemically leads to ChvG-ChvI activation. Muta

Multiple Strategies for the Application of Medicinal Plant-Derived Bioactive Compounds in Controlling Microbial Biofilm and Virulence Properties.

Biofilms are complex microbial communities encased within a self-produced extracellular matrix, which plays a critical role in chronic infections and antimicrobial resistance. These enhance pathogen survival and virulence by protecting against host immune defenses and conventional antimicrobial treatments, posing substantial challenges in clinical contexts such as device-associated infections and chronic wounds. Secondary metabolites derived from medicinal plants, such as alkaloids, tannins, flavonoids, phenolic acids, and essential oils, have gained attention as promising agents against biofi
