Novel noninvasive biomarkers of prodromal Alzheimer disease: The role of optical coherence tomography and optical coherence tomography-angiography.

BACKGROUND AND PURPOSE: A reduction of retinal thickness and an alteration of retinal perfusion have been found in Alzheimer disease (AD). Nowadays, retinal layers and retinal perfusion can be evaluated by means of noninvasive imaging techniques, namely, optical coherence tomography (OCT) and OCT-angiography (OCT-A). Here, we have compared the retinal thickness and the perfusion index, measured by means of OCT and OCT-A, in patients with mild cognitive impairment due to AD (MCI-AD) and in age- and sex-matched cognitively healthy controls. METHODS: Twenty-four MCI-AD patients and 13 control su

Lower retinal capillary density in minimal cognitive impairment among older Latinx adults.

INTRODUCTION: We investigated the hypothesis that retinal capillary perfusion is a biomarker of early cognitive decline and cerebrovascular perfusion associated with small vessel disease in a pilot data set of Latinx adults at high risk for vascular cognitive impairment and dementia. METHODS: High-resolution optical coherence tomography angiography (OCTA) images were acquired from dilated eyes of Latinx subjects using a 3 × 3 mm2 scan pattern from a commercially available device. A previously validated method was used to quantify the density of perfused retinal capillaries as the retinal vess

Multi-label deep learning for comprehensive optic nerve head segmentation through data of fundus images.

Early diagnosis and continuous monitoring of patients with eye diseases are critical in computer-aided detection (CAD) techniques. Semantic segmentation, a key component in computer vision, enables pixel-level classification and provides detailed information about objects within images. In this study, we present three U-Net models designed for multi-class semantic segmentation, leveraging the U-Net architecture with transfer learning. To generate ground truth for the HRF dataset, we combine two U-Net models, namely MSU-Net and BU-Net, to predict probability maps for the optic disc and cup regi

Response of capillaries and small arterioles to full-field flicker is not dependent on local ganglion cell thickness.

To measure the influence of ganglion cell layer (GCL) thickness on the changes in size and red blood cell (RBC) flow in small retinal vessels evoked by full-field flicker. We used a dual-beam adaptive optics scanning laser ophthalmoscope to image 11 healthy young controls in two retinal areas with significantly different GCL thicknesses. All capillaries and arterioles of the superficial vascular plexus were responsive to the flicker stimulation. Average lumen dilation and RBC flow changes were greater in capillaries than in arterioles (vasodilation: 10.9%, 6.7%; RBC flow: 51%, 38%, respectivel…

Microangiopathy of brain, retina, and inner ear.

Microangiopathy of brain, retina, and inner ear is a rare syndrome manifesting as arteriolar occlusions of the brain, retina, and inner ear, with resultant encephalopathy, visual, and hearing loss. Despite exhaustive laboratory examinations in these patients, no evidence of a systemic disease can be found. We treated and followed an adolescent with this disorder who initially presented with a branch retinal artery occlusion. A unique finding in this case was retinal vessel wall hyperfluorescence noted five days prior to retinal infarction. The patient developed recurrent branch artery occlusio

Phenotypic and genetic characteristics of retinal vascular parameters and their association with diseases.

Fundus images allow for non-invasive assessment of the retinal vasculature whose features provide important information on health. Using a fully automated image processing pipeline, we extract 17 different morphological vascular phenotypes, including median vessels diameter, diameter variability, main temporal angles, vascular density, central retinal equivalents, the number of bifurcations, and tortuosity, from over 130,000 fundus images of close to 72,000 UK Biobank subjects. We perform genome-wide association studies of these phenotypes. From this, we estimate their heritabilities, ranging

Repeatability of binarization thresholding methods for optical coherence tomography angiography image quantification.

Binarization is a critical step in analysis of retinal optical coherence tomography angiography (OCTA) images, but the repeatability of metrics produced from various binarization methods has not been fully assessed. This study set out to examine the repeatability of OCTA quantification metrics produced using different binarization thresholding methods, all of which have been applied in previous studies, across multiple devices and plexuses. Successive 3 × 3 mm foveal OCTA images of 13 healthy eyes were obtained on three different devices. For each image, contrast adjustments, 3 image processin

No such support is provided in the eye, and retinal vessels may rupture. Within the skull there may be drastic effects on brain circulation, but since changes in vascular transmural pressure are minimal vessel distension is not induced. CO2 dependence of retinal arterial and capillary blood velocity.

OBJECTIVE: Blood flow to the brain is extremely sensitive to changes in PCO2. While animal studies show a similar potent PCO2 dependence in retinal and choroidal vessels, the PCO2-retinal blood flow relationship has never been adequately studied in humans. METHODS: Video scanning laser ophthalmoscopy after fluorescein angiography was used to analyze retinal arterial and capillary blood velocity under conditions of mild hypercapnia and hypocapnia. Control conditions (end-tidal PCO2 = 38.3 +/- 0.4 mmHg) were contrasted with hyperventilation-induced hypocapnia (PCO2 = 34.0 +/- 0.4 mmHg) and hype
