Proansamycin B, the formerly postulated intermediate of rifamycin B biosynthesis, was isolated from cultures of the Amycolatopsis mediterranei mutant F1/24. The structure was determined using UV, IR, NMR and MS techniques. Biotransformation studies demonstrate that proansamycin B is an intermediate of a shunt pathway, a 8-deoxy variant, of rifamycin B biosynthesis leading to 8-deoxy-rifamycin B as the final product. Chromium(VI) reduction has been carried out by a variety of molecules of cellular and soil importance, such as thiol containing ones, ascorbic acid, saccharides and their derivatives, and nucleotides and their components using absorption spectroscopy. A direct dye-binding technique has been developed to measure total glycosaminoglycans in urine. Fifty or 100 microliters of urine was mixed with a solution of Azure A (10 mg/l) and Azure B (10 mg/l) and the resulting decrease in absorbance at 610 nm was measured. The standard curve with chondroitin sulfate C was linear from 1 to 20 micrograms/assay. The normal value for total urinary glycosaminoglycans in adults was 1.8 +/- 0.6 g/mol creatinine. Spectrophotometric measurement of the absorption characteristics of the lens capsule was carried out on 20 cases of so-called complicated or senile cataract operated through intracapsular cataract extraction (ICCE). The lens capsule almost completely transmitted visible radiation (between 400 and 750 nm) and near-ultraviolet (UV) radiation (between 300 and 400 nm). In the UV portion below 300 nm, the lens capsule showed one absorption peak at 280 nm and rapid increase of absorbance below 240 nm.
