Prefrontal cortical projections to the rat dorsal raphe nucleus: ultrastructural features and associations with serotonin and gamma-aminobutyric acid neurons.

Studies of human brain indicate that both the ventromedial prefrontal cortex (PFC) and the dorsal raphe nucleus (DRN) may be dysfunctional in major depressive illness, making it important to understand the functional interactions between these brain regions. Anatomical studies have shown that the PFC projects to the DRN, although the synaptic targets of this excitatory pathway have not yet been identified. Electrophysiological investigations in the rat DRN report that most serotonin neurons are inhibited by electrical stimulation of the PFC, suggesting that this pathway is more likely to synap

Testosterone regulation of the regenerative properties of injured rat sciatic motor neurons.

We have previously demonstrated that systemic administration of testosterone differentially regulates the regenerative properties of injured hamster facial motor neurons, which are androgen receptor-containing cranial motor neurons. In this investigation, the hypothesis that testosterone alters the regenerative properties of rat sciatic motor neurons, which are androgen receptor-containing spinal motor neurons, was tested using fast axonal transport of radioactively labeled proteins to assess sciatic nerve regeneration. Adult castrated male rats were subjected to crush axotomy of the sciatic n…

Pathological conformations involving the amino terminus of tau occur early in Alzheimer's disease and are differentially detected by monoclonal antibodies.

Conformational changes involving the amino terminus of the tau protein are among the earliest alterations associated with tau pathology in Alzheimer's disease and other tauopathies. This region of tau contains a phosphatase-activating domain (PAD) that is aberrantly exposed in pathological forms of the protein, an event that is associated with disruptions in anterograde fast axonal transport. We utilized four antibodies that recognize the amino terminus of tau, TNT1, TNT2 (a novel antibody), Tau12, and Tau13, to further study this important region. Using scanning alanine mutations in recombina…

Retrograde neuronal tracing with a deletion-mutant rabies virus.

We have constructed a deletion-mutant rabies virus encoding EGFP and find it to be an excellent tool for studying detailed morphology and physiology of neurons projecting to injection sites within the mammalian brain. The virus cannot spread beyond initially infected cells yet, unlike other viral vectors, replicates its core within them. The cells therefore fluoresce intensely, revealing fine dendritic and axonal structure with no background from partially or faintly labeled cells.

Telencephalic connections of the visual system of the chicken: tracing the interrelation of the efferents of the visual Wulst and the hyperstriatum ventrale.

In the present study the telencephalic connections between the centres of the tectofugal and thalamofugal pathways were investigated with Phaseolus vulgaris leucoagglutinin and biotinylated dextran-amine anterograde tracers in chicken using light and electron microscopy. No direct connection was found between the visual Wulst and the ectostriatum or the telencephalic centres of the tectofugal and thalamofugal pathways. Besides other projections, the visual Wulst emitted fibres also to the middle and lateral parts of the hyperstriatum ventrale. Further experiments revealed that the middle part

Study of the topographical distribution of different populations of motoneurons within rat's nucleus ambiguus, by means of four different fluorochromes.

The topographical neuronal distribution within the rat nucleus ambiguus has been studied with the simultaneous retrograde labeling technique by means of four different fluorochromes injected within the various muscles and/or nerves of the oro-pharyngeal region. This technique has permitted the identification of several types of neurons along the same coronal plane. Most were motoneurons innervating the various muscles of the upper airway, including pharyngeal constrictor, stylopharyngeal, intrinsic laryngeal and the upper portion of the esophagus. Some neurons may have been preganglionic paras

Pacinian corpuscles in rats with carbon disulphide neuropathy.

Adult Wistar rats were exposed to carbon disulphide vapours at a concentration of 2.4 mg/l of air for 5 days a week (6 h a day), and the ultrastructure of Pacinian corpuscles and their nerve supply was investigated after 6 months of exposure. Both degenerative and regenerative changes were observed in sensory axons and the corpuscles. In a sample of corpuscles examined, 30% were denervated and about 60% showed clear signs of reinnervation. Some of the reinnervated corpuscles were supplied by unmyelinated axons. In others, one to three myelinated axons were already found at the nerve entry. The

Collaterals of corticospinal and pyramidal fibres to the pontine grey demonstrated by a new application of the fluorescent fibre labelling technique.

Selective visualization of collaterals of corticospinal and pyramidal fibres to the pons in cat was obtained by retrograde transport of the fluorescent tracer fast blue (FB) through the stem fibres. Unilateral FB injections in the cervical cord and the pyramidal tract respectively produced soft blue fluorescent labelling of pyramidal fibres and of fibres and structures resembling 'terminals' in the pontine grey: contralateral to the spinal injections and ipsilateral to the pyramidal injections. These labelled elements were concluded to represent collaterals of corticospinal and pyramidal fibre

The origin of the reticulo-olivary projection in the rat: a retrograde horseradish peroxidase study.

Electrophoretic injections of horseradish peroxidase were made in different parts of the rat inferior olivary complex using a ventral approach. Data from these injections provide anatomical evidence for the existence of a projection to the inferior olive which takes origin from reticular nuclei in the brainstem. The majority of reticulo-olivary neurons are located in the nucleus raphe obscurus and nucleus raphe pallidus. Other reticular nuclei which contribute to this projection include the nucleus reticularis ventralis and nucleus reticularis gigantocellularis. Analysis of injections confined

A barbiturate-induced potassium permeability increase in the myelinated nerve membrane.

Effects of the highly lipid soluble barbiturates methohexital and thiopental on potential clamped myelinated nerve fibres were analyzed. Both were found to affect the potassium as well as sodium transport system. The methohexital effect on the potassium system was complex. At low potential values it reduced the permeability, while at high values it increased the permeability as well as slowed down the kinetics. It antagonized the rectification of IK at high potential values caused by increased axoplasmic [Na+]. The effect may be described as a combination of a decreased permeability constant (

Rehabilitation of an electrical burn patient through thermal biofeedback.

A 22-year-old male subject, with high-voltage electrical burns to one wrist, utilized differential relaxation and visual biofeedback to increase skin temperature in the damaged hand. Through 14 thermal biofeedback and passive relaxation sessions, the subject was able to produce temperature increases in his damaged hand of up to 21 degrees F, which considerably diminished the pain. Healing, feeling, and movement control seemed to progress with extreme rapidity, suggesting that axoplasmic transport was greatly enhanced.

Direct transfer of viral and cellular proteins from varicella-zoster virus-infected non-neuronal cells to human axons.

Varicella Zoster Virus (VZV), the alphaherpesvirus that causes varicella upon primary infection and Herpes zoster (shingles) following reactivation in latently infected neurons, is known to be fusogenic. It forms polynuclear syncytia in culture, in varicella skin lesions and in infected fetal human ganglia xenografted to mice. After axonal infection using VZV expressing green fluorescent protein (GFP) in compartmentalized microfluidic cultures there is diffuse filling of axons with GFP as well as punctate fluorescence corresponding to capsids. Use of viruses with fluorescent fusions to VZV pro

Distribution of neurons projecting to the rostral ventrolateral medullary pressor region that are activated by sustained hypotension.

Hypotension produces a reflex increase in the activity of sympathetic vasomotor and cardiac nerves. It is believed that the reflex sympathoexcitation is due largely to disinhibition of sympathoexcitatory neurons in the rostral ventrolateral medulla, but it is possible that it may also be mediated by excitatory inputs from interneurons that are activated by a fall in blood pressure. The aim of this study in conscious rabbits was to identify and map neurons with properties that are characteristic of interneurons conveying excitatory inputs to the rostral ventrolateral medullary pressor region in

Dendritic arbors and central projections of physiologically characterized auditory fibers from the saccule of the toadfish, Opsanus tau.

Neurobiotin was injected iontophoretically into saccular afferents of toadfish (Opsanus tau) after intracellular recording to examine dendritic arbors and central projections with respect to the physiological and directional response properties of the cells. Dendritic arbors of 36 afferents were examined in detail. Maximum diameter of the arbor and the number of terminal points were positively correlated with each other, but neither was predictive of spontaneous activity or sensitivity. Best azimuths were centered around 30 degrees -40 degrees, which corresponds to the angle of the saccule wit

The temporal lobe is a target of output from the basal ganglia.

The basal ganglia are known to receive inputs from widespread regions of the cerebral cortex, such as the frontal, parietal, and temporal lobes. Of these cortical areas, only the frontal lobe is thought to be the target of basal ganglia output. One of the cortical regions that is a source of input to the basal ganglia is area TE, in inferotemporal cortex. This cortical area is thought to be critically involved in the recognition and discrimination of visual objects. Using retrograde transneuronal transport of herpes simplex virus type 1, we have found that one of the output nuclei of the basal

Distribution and morphological characterization of viscerofugal projections from the large intestine to the inferior mesenteric and pelvic ganglia of the male rat.

Viscerofugal neurons are enteric neurons in the myenteric plexus of the stomach and intestine that project to the prevertebral ganglia as the afferent limb of intestino-intestinal reflexes. This study characterizes viscerofugal projections to the inferior mesenteric ganglion and investigates the possibility of similar projections to the major pelvic ganglia in the male rat. The colon and rectum were examined for retrogradely labelled neurons following the injection of retrograde tracer into the inferior mesenteric or major pelvic ganglia, or following the application of tracer to the caudal en

Zinc deficiency and peripheral neuropathy in chicks.

Zinc-deficient chicks develop an arthritic-like neuromuscular disorder. They walk with a stilted gait and tend to remain in a squat position, bearing little weight on the legs. The purpose of this study was to determine the basis of the syndrome by making electrophysiologic measurements of nerve function. Chicks were fed low zinc (6 mg/kg) and zinc-adequate (50 mg/kg) diets, the latter ad libitum and pair-fed. At the end of 3 weeks, sciatic nerve function was determined in vivo by use of an electrodiagnostic system. Motor nerve conduction velocity was significantly lower in chicks fed the low

A retinal projection to the paraventricular nuclei of the hypothalamus in the Syrian hamster (Mesocricetus auratus).

The retino-hypothalamic tract of the Syrian hamster (Mesocricetus auratus) was examined using intraocular injections of horseradish peroxidase conjugated to cholera toxin. In addition to the retinal input to the suprachiasmatic nuclei (SCN), a retinal projection to the caudal paraventricular nuclei was identified. This projection may mediate some effects of light upon pineal gland physiology independently of the SCN and the circadian-rhythm generating system.

A comparative topographical analysis of dorsal column nuclear and cerebral cortical projections to the basilar pontine gray in rats.

The somatotopic distribution of dorsal column nuclear projections within the basilar pontine gray was examined in relation to the massive corticopontine projection system that emanates most heavily from motor and somatosensory cortex. The distribution patterns of these two systems were compared by combining autoradiographic and degeneration axonal tracing methods within individual animals. Stereotaxic injections of tritiated leucine (50 microCi/microliter) and lesions by aspiration were made in animals under ketamine hydrochloride anesthesia. The forelimb cortical injections (0.1-0.3 microlite

Anatomical and behavioral recovery from the effects of spinal cord transection: dependence on metamorphosis in anuran larvae.

This study of spinal cord injury in bullfrog (Rana catesbeiana) tadpoles using the neuroanatomical tracer horseradish peroxidase (HRP) was undertaken to determine (1) whether the same anatomical regions that normally give rise to ascending or descending spinal tracts do so following complete spinal cord transection and (2) whether the course of behavioral recovery could be related to the anatomical results. The results of this study show that (1) spinal cord continuity is readily restored in tadpoles subjected to spinal cord transection, but nerve fibers crossing the site of injury end within
