Human B cell activation: selective sensitivity of the early stages to calcium channel-blocking drugs.

The importance of Ca2+ in the early events of lymphocyte activation has been suggested by several studies. We examined the effect of calcium channel-blocking drugs (verapamil and nitrendipine) on the progression of human B cells through their activation cycle. Our results show that these drugs suppress the anti-mu-induced human B cell proliferation and interfere with the early events of the B cell activation in a dose-dependent fashion. This suppression correlates with a marked decrease in anti-mu-induced 45Ca2+ uptake. Calcium channel-blocking drugs inhibit the anti-mu-induced uridine incorpo

Beneficial effects of calcium channel blockers and calmodulin binding drugs on in vitro renal cell anoxia.

Proximal tubules (PSTs) of the S1, S2 and S3 segments and cortical collecting tubules (CCTs) were microdissected individually from rabbit kidneys and cultured for 7 days in hormonally defined media. Anoxia was induced by incubation of cultures in normal medium for 45 min at 25 degrees C in an atmosphere of nitrogen and cell death was measured by nigrosine dye uptake. After 45 min of anoxia and a 4- to 6-hr incubation in normal Ca++-containing media, cells from all segments were dead. Addition of calcium channel blockers verapamil and nifedipine (5 X 10(-7) and 10(-6) M, respectively) for the f

Modulation of fibroblast-induced clot retraction by calcium channel blocking drugs and the monoclonal antibody ALB6.

Suspensions of living human fibroblast induce fibrin clot retractile activity (FCR). The efficiency is dependent on the growth phase; it is maximal during active growth and reduced in post-confluent cultures. In contrast human osteosarcoma cells constantly exhibit very low FCR efficiency. Two different calcium channel-blocking drugs Diltiazem and Verapamil inhibit, depending on the concentrations employed, FCR, and spreading within the clots of the normal cells. Intermediate FCR levels are associated with intermediate degrees of spreading. A similar dose dependent inhibition is also obtained b

Research and optimization of screening strategy for calcium-activated chloride channel modulators guided by electrophysiological characteristics.

Calcium-activated chloride channels (CaCCs) are essential for epithelial secretion, neuronal transmission, and smooth muscle function. Among the Anoctamin family, Anoctamin 1 (ANO1) and Anoctamin 2 (ANO2) are classical CaCCs proteins. ANO1 has been identified as a potential therapeutic target due to its involvement in diseases such as cancer and cystic fibrosis. However, current high-throughput screening (HTS) systems face limitations in achieving subtype-specific detection and optimizing screening strategies. Stable HTS cell models expressing ANO1 or ANO2 were constructed via lentiviral trans

Calcium Channel Blockers Inhibit Pancreatic Neuroendocrine Neoplasms Progression via Cav1.2-Epigenetic Circuit.

Pancreatic neuroendocrine neoplasms (pNEN) are rarely encountered, accounting for about 2% of all pancreatic neoplasms. Disease progression is frequently observed as recurrence or distal metastasis. Mechanisms underlying pNEN progression are still poorly investigated, and treatments against pNEN are challenging due to the pronounced neoplastic heterogeneity. Here, by performing clinicomolecular analysis, we report a novel mechanism of positive regulatory circuit between Cav1.2-mediated calcium signaling and epigenetic control by H3K27 acetylation (H3K27ac). Tumor-cell-specific expression of Ca

Pseudolymphoma associated with calcium channel blockers: a FAERS disproportionality analysis comparing dihydropyridines and non-dihydropyridines.

Calcium channel blockers (CCBs), particularly amlodipine, are among the most widely prescribed antihypertensive agents globally. However, no comprehensive disproportionality analysis has quantified this association. This pharmacovigilance study utilized the FDA Adverse Event Reporting System (FAERS) to evaluate the reporting association between CCBs and pseudolymphoma. The FAERS database was parsed through its 14 104 743 adverse event reports. Five CCBs were evaluated: three dihydropyridines (amlodipine, nifedipine, and felodipine) and two non-dihydropyridines (diltiazem, verapamil). Dispropor

Calcium Channel Blockers are Safe and Beneficial in Patients with Heart Failure and Advanced Chronic Kidney Disease.

BACKGROUND: Heart failure and advanced chronic kidney disease frequently coexist and are associated with high mortality and limited therapeutic options. Calcium channel blockers (CCBs) are commonly used for blood pressure control in this population. However, their effect on survival in patients with heart failure and advanced chronic kidney disease is uncertain. We evaluated the association between CCBs use and all-cause mortality in patients with heart failure and advanced chronic kidney disease. METHODS: This retrospective cohort study included 1,835 consecutive patients with heart failure

Cervical dystonia as the presenting symptom of novel calcium voltage-gated channel subunit alpha1A mutation.

A woman in her mid-50s presented with an uncomfortable pulling sensation in the right neck that worsened with movement. This was diagnosed as cervical dystonia and treated symptomatically for several years through trials of botulinum toxin. As the years progressed, her symptoms worsened and evolved with the patient eventually experiencing debilitating ataxia requiring use of a wheelchair for mobility, severe dysphagia requiring the use of a feeding tube for nutrition and dysarthria requiring the need of a family member for translation. Through genetic testing, the patient was diagnosed with sp

Blockade of calcium-activated chloride channel ANO1 ameliorates ionizing radiation-induced intestinal injury.

INTRODUCTION: Radiation enteritis is one of the most frequent clinical complications of radiotherapy (RT), yet few effective strategies currently exist to protect against that. Anoctamin 1 (ANO1) functions both as a chloride channel and a signal transduction protein, influencing numerous pathophysiological processes. OBJECTIVES: This study aimed to investigate whether targeting ANO1 could mitigate radiation-induced enteritis while enhancing tumor radiosensitivity. METHODS: Quantitative PCR (qPCR) and Western blot (WB) were used to assess ANO1 expression and its changes after irradiation. Sur

Calcium channel blocker use and outcomes following transcatheter aortic valve intervention for aortic stenosis.

Calcium channel blockers (CCBs) are commonly used to treat coronary artery disease (CAD). The effects of the use of CCBs on the prognosis of patients with aortic stenosis (AS) after transcatheter aortic valve intervention (TAVI) has not been explored. This study elucidated the effects of the use of CCBs on clinical outcomes of patients who underwent TAVI for severe AS. This retrospective observational study included 993 consecutive patients who underwent TAVI for severe AS between January 2017 and July 2023. All patients were followed up for all-cause mortality and hospitalisation for heart fa

Management of hypotension in dihydropyridine calcium channel blocker overdose: The role of high-dose insulin therapy.

AIMS: Amlodipine poisoning is a leading cause of cardiovascular medication-related deaths, commonly managed with high-dose insulin (HDI) therapy. However, HDI is a vasodilator that is counterproductive in managing vasoplegia. We aim to study HDI therapy in patients with hypotension following dihydropyridine calcium channel antagonist (CCA) overdose. METHODS: This retrospective study includes adult patients (≥15 years) with deliberate dihydropyridine CCA overdose and hypotension (mean arterial pressure <65 mmHg or systolic blood pressure <90 mmHg) managed by two Poisons Information Centres and

L-type calcium channel blockade attenuates cue-induced cocaine-seeking in female rats.

Periods of cocaine abstinence are associated with a high risk of relapse, often triggered by exposure to drug-associated cues. Previous research has implicated L-type calcium channels (LTCCs) in drug-seeking behaviors, yet their role in cue-induced relapse, particularly in females, remains underexplored. This study investigated the effects of LTCC inhibition on cue-induced cocaine-seeking behavior during abstinence in female Sprague-Dawley rats. Following a 10-day cocaine self-administration and a 14-day forced abstinence period, the rats were tested for cue-induced cocaine-seeking after recei…

Development of nifedipine isosteres: an integrated approach to the design, synthesis, and biological assessment of calcium channel blockers.

This study reports the synthesis of a series of calcium channel blockers via Biginelli's reaction. The core dihydropyridine (DHP) scaffold, an isostere of nifedipine, was synthesized using three aldehydes incorporated with trifluoromethyl (-CF3) substitutions at the ortho, meta, and para positions. The resulting series (4a-c to 9a-c) was evaluated for antihypertensive and calcium channel-blocking activities in male and female rats, administered intraperitoneally. Among the synthesized compounds, the ortho-substituted derivatives (4a, 7a, 8a, and 9a) demonstrated the highest antihypertensive ac

High-Dose Insulin Euglycemic Therapy in Concomitant Beta-Blocker and Calcium Channel Blocker Overdose.

Beta-blockers and calcium channel blockers are among the most commonly used drugs in the adult population, and the National Poison Data System annual reports note 11 075 beta-blocker overdoses and 6256 calcium-channel blocker overdoses in 2022. Poisoning with these medications presents clinically with profound bradycardia, depression of myocyte contractility, and vasodilation resulting in hemodynamic instability often refractory to initial vasopressor treatment. Herein we present a case of concomitant beta-blocker and calcium-channel blocker overdose with hypotension refractory to vasopressors
