FGF2-induced STAT3 activation regulates pathologic neovascularization.

Cell-autonomous endothelial cell (EC) fibroblast growth factor receptor (FGFR) signaling through FGFR1/2 is essential for injury-induced wound vascularization and pathologic neovascularization as in blinding eye diseases such as age-related macular degeneration. Which FGF ligand(s) is critical in regulating angiogenesis is unknown. Utilizing ex vivo models of choroidal endothelial sprouting and in vivo models of choroidal neovascularization (CNV), we demonstrate here that only FGF2 is the essential ligand. Though FGF-FGFR signaling can activate multiple intracellular signaling pathways, we sho

Stearoyl-CoA desaturase 1 deficiency protects mice from immune-mediated liver injury.

Immunity and metabolism are closely linked. The liver is an important metabolic organ in the body. However, the interactions between hepatocytes and the immune system are poorly understood. In mice developing concanavalin A (ConA)-induced hepatitis (CIH), we found extensive lipid accumulation in hepatocytes. Critical enzyme involved in fat synthesis such as stearoyl-CoA desaturase 1 (SCD1) was upregulated. When we injected ConA to SCD1-deficient mice, we found these mice to be highly resistant to CIH. The mechanisms of the protective effect of SCD1 deficiency might be attributed to the reduced

[Research progress on miRNA-regulated macrophage polarization and autophagy in immune escape by Mycobacterium tuberculosis].

Latent tuberculosis infection (LTBI), characterized by the host immune system's ability to contain Mycobacterium tuberculosis (MTB) proliferation while failing to fully eliminate the pathogen, is a critical stage in tuberculosis prevention and control. Macrophages and alveolar epithelial cells together constitute the first line of defense. Following infection, MTB triggers a series of responses in which miRNAs, in concert with other biomolecules, coordinately regulate macrophage polarization, autophagic activity, the key metabolic pathways of autophagy, and the inflammatory response of host ma

How hyperglycemia promotes ocular diseases: An overview of molecular mechanisms.

Ocular diseases result from a complex interaction of environmental and metabolic factors. The increasingly prevalent sedentary lifestyle and dietary patterns characterized by high sugar and ultra-processed food intake substantially elevate the risk of hyperglycemia. Hyperglycemia induces dysregulation of core metabolic pathways, such as overactivation of the polyol and hexosamine pathways, aberrant activation of protein kinase C, and accumulation of advanced glycation end products. Hyperglycemia further triggers tissue-specific molecular and cellular events in various ocular structures. These

The impact of volatile anesthetics and propofol on phosphatidylinositol 4,5-bisphosphate signaling.

Phosphatidylinositol 4,5-bisphosphate (PIP2), as well as other anionic phospholipids, play a pivotal role in various cellular processes, including ion channel regulation, receptor trafficking, and intracellular signaling pathways. The binding of volatile anesthetics and propofol to PIP2 leads to alterations in PIP2-mediated signaling causing modulation of ion channels such as ɣ-aminobutyric acid type A (GABAA) receptors, voltage-gated calcium channels, and potassium channels through various mechanisms. Additionally, the interaction between anionic phospholipids and G protein-coupled receptors

Anatomy and Embryology of the Middle Ear, Labyrinth, and Intracranial Vestibular Pathways.

The intricate anatomy and embryology of the middle ear, labyrinth, and vertigo-related intracranial pathways involve complex developmental processes and contributions from multiple germ layers. The middle ear, comprised of the tympanic cavity, ossicles, and Eustachian tube, develops from the first and second branchial arches and clefts. In contrast, the inner ear originates from the otic vesicle, forming the bony and membranous labyrinths. The embryological timeline spans from the 40th day of gestation to the 24th week. The vestibulocochlear nerve (VIII cranial nerve) emerges with the inner ea

REV7-p53 interaction inhibits ATM-mediated DNA damage signaling.

REV7 is an abundant, multifunctional protein that is a known factor in cell cycle regulation and in several key DNA repair pathways including Trans-Lesion Synthesis (TLS), the Fanconi Anemia (FA) pathway, and DNA Double-Strand Break (DSB) repair pathway choice. Thus far, no direct role has been studied for REV7 in the DNA damage response (DDR) signaling pathway. Here we describe a novel function for REV7 in DSB-induced p53 signaling. We show that REV7 binds directly to p53 to block ATM-dependent p53 Ser15 phosphorylation. We also report that REV7 is involved in the destabilization of p53. Thes

Müller Cell Metabolic Signatures: Evolutionary Conservation and Disruption in Disease.

Müller cells are glia that play important regulatory roles in retinal metabolism. These roles have been evolutionarily conserved across at least 300 million years. Müller cells have a tightly locked metabolic signature in the healthy retina, which rapidly degrades in response to insult and disease. This variation in metabolic signature occurs in a chaotic fashion, involving some central metabolic pathways. The cause of this divergence of Müller cells, from a single class with a unique metabolic signature to numerous separable metabolic classes, is currently unknown and illuminates potential al

The Yin and Yang of ACE/ACE2 Pathways: The Rationale for the Use of Renin-Angiotensin System Inhibitors in COVID-19 Patients.

The article describes the rationale for inhibition of the renin-angiotensin system (RAS) pathways as specific targets in patients infected by SARS-CoV-2 in order to prevent positive feedback-loop mechanisms. Based purely on experimental studies in which RAS pathway inhibitors were administered in vivo to humans/rodents, a reasonable hypothesis of using inhibitors that block both ACE and ACE2 zinc metalloproteases and their downstream pathways in COVID-19 patients will be proposed. In particular, metal (zinc) chelators and renin inhibitors may work alone or in combination to inhibit the positiv

The genomic landscape of cutaneous melanoma.

Somatic mutation analysis of melanoma has been performed at the single gene level extensively over the past several decades. This has provided considerable insight into the critical pathways controlling melanoma initiation and progression. During the last 5 yr, next-generation sequencing (NGS) has enabled even more comprehensive mutational screening at the level of multigene panels, exomes and genomes. These studies have uncovered many new and unexpected players in melanoma development. The recent landmark study from The Cancer Genome Atlas (TCGA) consortium describing the genomic architecture…

Pathways leading to interleukin-12 production and protective immunity in cutaneous leishmaniasis.

Leishmaniasis affects millions of people worldwide and continues to pose public health problem. There is extensive evidence supporting the critical role for IL-12 in initiating and maintaining protective immune response to Leishmania infection. Although gene deletion studies show that CD40-CD40L interaction is an important pathway for IL-12 production by antigen-presenting cells and subsequent development of protective immunity in cutaneous leishmaniasis, several studies have uncovered other pathways that could also lead to IL-12 production and immunity in the absence of intact CD40-CD40L sign

Therapeutic targets in head and neck squamous cell carcinoma: identification, evaluation, and clinical translation.

Head and neck squamous cell carcinoma (HNSCC) encompasses a diverse group of malignancies originating in the oral cavity, oropharynx, larynx and hypopharynx. Although treatment modalities have improved, carefully designed biomarker-driven clinical trials will yield the best opportunities to enhance HNSCC therapy options in the future. Due to the heterogeneous nature of HNSCC, discovering a "silver bullet" for the treatment of HNSCC is unlikely. Consequently, impactful HNSCC clinical trials will require multiple assay platforms and expanded technical expertise. In this review, we will outline p

Innate and adaptive immune responses in hepatitis B virus infection.

There is urgent need for novel immunotherapeutic strategies which, combined with current anti-viral therapy, may facilitate viral control and remove the need for long-term suppressive therapy in patients who have chronic hepatitis B virus (HBV) infection. To achieve this, it is critical to better understand mechanisms of innate and adaptive immune dysfunction and pathways of liver pathology. Here, we summarise recent advances pertaining to early immune events during acute HBV infection in an unusual cohort of patients sampled in the pre-clinical phase, and describe mechanisms of CD8 T cell att

Finite-element simulation of in-plane tear propagation in the dissected aorta: Implications for the propagation mechanism.

Computer modeling and numerical simulation are essential for understanding the progression of aortic dissection. However, tear propagation has not been properly modeled and simulated. The in-plane dissection propagation between concentrically distributed elastic lamellae is modeled using the cohesive zone method with a bilinear traction-separation law. The parameters of cohesive elements are calibrated for the three modes of interfacial damage in the media, enabling quantitative predictions of in-plane tear propagation. An idealized cylindrical tube-shaped bilayer thick-wall model of the aorta

Tissue-stiffness-driven platelet activation emerges as a potential target for breast cancer prevention and treatment.

BACKGROUND: High mammographic breast density is a strong independent risk factor for sporadic breast cancer, yet involved mechanisms remain poorly defined. The extracellular compartment plays a critical role in the intercellular communication during tumor initiation and progression. Although several mechanosensitive pathways have been described, the role of platelets (PLTs) in stiffness-driven signaling in the breast is unknown. METHODS: Extracellular soluble proteins were sampled in situ from live breast tissue using microdialysis. A total of 108 postmenopausal women were included: women wit

Hepatocyte Rho-associated kinase signaling is required for mice to survive experimental porphyria-associated liver injury.

BACKGROUND: Rho-associated kinases 1 and 2 (ROCK1 and ROCK2) regulate critical cell functions, including actomyosin contractility, apoptosis, and proliferation. Some studies suggest that ROCK inhibition may serve as a treatment for liver fibrosis. More investigation is needed to understand the role of hepatocyte ROCK signaling in vivo, especially in the context of profibrotic liver injury. METHODS: Rock1fl/fl, Rock2fl/fl, and Rock1fl/fl; Rock2fl/fl mice were given adeno-associated virus serotype 8 (AAV8)-thyroid hormone-binding globulin (TBG)-Cre to produce targeted gene deletion in hepatocyt

Revisiting the Telovelar Approach for Lesions of the Mesial Part of the Cerebellar Peduncles by Using the Cerebellar Functional Networks to Guide Surgical Practice.

BACKGROUND AND OBJECTIVES: Lesions of the cerebellar peduncles (CPs), particularly the mesial CP, pose a surgical challenge because of their deep location and the risk of disrupting critical neural pathways. Given their low incidence, the safest and most anatomically sound surgical approach for this region remains to be determined. This study analyzes the surgical anatomy and technique of the telovelar approach with particular focus on the white matter tract anatomy of the middle (MCP), inferior (ICP), and superior CPs. By integrating cadaveric studies, tractographic anatomy and surgical cases

Predictive modeling of ARDS mortality integrating biomarker/cytokine, clinical and metabolomic data.

Acute Respiratory Distress Syndrome (ARDS), characterized by the rapid onset of respiratory failure and mortality rates of ∼40%, remains a significant challenge in critical care medicine. Despite advances in supportive care, accurate prediction of ARDS mortality remains challenging, resulting in delayed delivery of targeted interventions and effective disease management. Traditional critical illness severity scores lack specificity for ARDS, underscoring the need for more precise prognostic tools for ARDS mortality. To address this crucial gap, we employed a multimodal approach to predict ARDS
