Chylomicrons Regulate Lacteal Permeability and Intestinal Lipid Absorption.

BACKGROUND: Lymphatic vessels are responsible for tissue drainage, and their malfunction is associated with chronic diseases. Lymph uptake o … 1 (Rac family small GTPase 1), thereby restricting RhoA (Ras homolog family member A)/ROCK-mediated cytoskeleton contraction. CONCLUSIONS: Our results reveal that antagonistic inputs into ROCK-dependent cytoskeleton contractions regulate the interconversion of lymphatic junctions in the intestine and in other tissues, providing a tunable mechanism to control …

[N-WASP regulates cortical neuron migration through its polyPro and VCA domains].

Cortical neuron migration in the developing mouse forebrain is a complex process, which contains several steps related to cytoskeleton dynamics and remodeling. Neural Wiskott-Aldrich syndrome protein (N-WASP), a member of the WASP-WAVE family, regulates actin cytoskeleton reorganization through the binding of its VCA domain to the Arp2/3 complex. Here we report expression patterns of N-WASP gene in the mouse developing embryonic cortex (E12.5~ E18.5) and find its expression levels are decreased during embryonic development. By using in utero electroporation (IUE) method, we find that either N-…

Iron-modulated virulence factors of Entamoeba histolytica.

Entamoeba histolytica is a human parasite that causes amoebiasis, a disease that affects the colon and liver and is prevalent worldwide. This protozoan requires a high concentration of iron to survive and reproduce. Iron modulates the expression of parasite virulence factors, including hemoglobinases, hemoglobin-binding proteins and cysteine proteases, as well as proteins related to the amoebic cytoskeleton. This review summarizes the virulence factors that are affected by iron, resulting in upregulation or downregulation of E. histolytica genes. This review also discusses the functionality of…

Molecular Control of Actin Dynamics In Vivo: Insights from Drosophila.

The actin cytoskeleton provides mechanical support for cells and generates forces to drive cell shape changes and cell migration in morphogenesis. Molecular understanding of actin dynamics requires a genetically traceable model system that allows interdisciplinary experimental approaches to elucidate the regulatory network of cytoskeletal proteins in vivo. Here, we will discuss some examples of how advances in Drosophila genetics and high-resolution imaging techniques contribute to the discovery of new actin functions, signaling pathways, and mechanisms of actin regulation in vivo.

Plectin gene defects lead to various forms of epidermolysis bullosa simplex.

Plectin is an important organizer of the keratin filament cytoskeleton in basal keratinocytes. It is essential for anchoring these filaments to the extracellular matrix via hemidesmosomal integrins. Loss of plectin or incorrect function of the protein due to mutations in its gene can lead to various forms of the skin blistering disease, epidermolysis bullosa simplex. Severity and subtype of the disease is dependent on the specific mutation and can be associated with (late-onset) muscular dystrophy or pyloric atresia. Mouse models mimicking the human phenotypes allow detailed study of plectin f…

Wiskott-Aldrich Syndrome: Immunodeficiency resulting from defective cell migration and impaired immunostimulatory activation.

Regulation of the actin cytoskeleton is crucial for many aspects of correct and cooperative functioning of immune cells, such as migration, antigen uptake and cell activation. The Wiskott-Aldrich Syndrome protein (WASp) is an important regulator of actin cytoskeletal rearrangements and lack of this protein results in impaired immune function. This review discusses recent new insights of the role of WASp at molecular and cellular level and evaluates how WASp deficiency affects important immunological features and how defective immune cell function contributes to compromised host defence.

Genetics of nephrotic syndrome: connecting molecular genetics to podocyte physiology.

Urinary losses of macromolecules in nephrotic syndrome (NS) reflect a dysfunction of the highly permselective glomerular filtration barrier. Genetic studies of hereditary forms of NS have led to the identification of proteins playing a crucial role in slit-diaphragm signalling, regulation of actin cytoskeleton dynamics, maintenance of podocyte integrity and cell-matrix interactions. This review will focus on recent molecular and clinical findings in the field of genetics of NS, thereby providing a better understanding of the complex glomerular filtration barrier physiology.

[Effects of microfilaments on the expression of collagenases and tissue inhibitor of metalloproteinases-1 in fibroblasts].

OBJECTIVE: To investigate the roles of microfilaments in the expression of collagenase and TIMP-1 in normal and hypertrophic scar. METHODS: Cell culture and Northern blot hybridization methods were used in this study. RESULTS: After the disruption of microfilaments with cytochalasin B, mRNA contents of collagenase and TIMP-1 both increased significantly in normal and hypertrophic scar fibroblasts. CONCLUSION: The result suggest that the microfilament cytoskeleton may regulate the synthesis and degradation of ECM, which may be a mechanism of gene regulation during wound healing.

The Wiskott-Aldrich syndrome: disordered actin dynamics in haematopoietic cells.

The Wiskott-Aldrich syndrome protein (WASp) is a member of a unique family whose members share similar domain structures and are responsible for the transduction of signals from the cell membrane to the actin cytoskeleton. For WASp, the interactions with Rho family GTPases and the cytoskeletal organising complex Arp2/3 are critical to these functions, which when disturbed translate into abnormalities of haematopoietic cell signaling, polarisation, migration and phagocytosis. This review discusses the evidence for regulation of highly dynamic cytoskeletal structures by WASp and the consequences…

[Intrahepatic cholestasis in chronic liver disease].

Cholestasis syndromes are characterized by a reflux of compounds usually excreted with bile. ATP dependent carriers and cytoskeleton proteins guarantee physiological bile flux. There are several clinical conditions in which this system is affected. Intrahepatic cholestasis is characterized by damage to hepatocytes or intrahepatic bile ducts. Primary biliary cirrhosis and primary sclerosing cholangitis represent examples of cholestatic chronic liver disease. The pathogenesis of these two conditions seems to be mediated by immunological reactions. Moreover, hepatitis viruses are able to induce c…

A transmission electron microscope study of fibroblast changes in human deciduous tooth pulp.

The coronal pulps of four primary canines, free from radicular resorption, and of three primary teeth, with roots entirely resorbed, were prepared for electron microscopy. Mature fibroblasts, similar to those reported in the pulps of permanent teeth, were present in the pulps of the clinically mature teeth. Regression of fibroblasts, evident in the pulps of deciduous teeth at the root resorption stage, was characterized by cytoplasmic changes, modifications in the cytoskeleton and dissociation of cells and fibrils. Deciduous teeth might provide a model for studying pulp fibroblast ageing.

A distinct interphase microtubule state marks host cell permissiveness to Chlamydia pneumoniae entry.

Entry of intracellular pathogenic bacteria is widely considered an actin-driven process, potentially overlooking contributions of the microtubule cytoskeleton. Interphase microtubules exist in distinct states, but whether this heterogeneity influences bacterial entry is unknown. Here, using the obligate intracellular bacterium Chlamydia pneumoniae, we show that microtubules not only contribute to bacterial entry but that their pre-existing state influences host-cell permissiveness. Cells enriched in acetylated microtubules are preferentially infected, whereas detyrosinated microtubules show no…

Loss of Cotl1 disrupts mitochondrial fission and impairs learning and memory in mice.

Mitochondrial morphology dynamics are essential for neuronal function and are regulated by fission and fusion machinery, in which the actin cytoskeleton plays a crucial role. Coactosin-like F-actin-binding protein 1 (Cotl1), a key component of the actin cytoskeleton, is gaining attention for its profound pathophysiological implications in human neurodegenerative disorders. However, its role in neuronal function remains unclear. Therefore, we aimed to explore the role of Cotl1 in mitochondrial morphology and neuronal dysfunction. We generated Cotl1-knockdown and Cotl1-K75E mutants to examine mi…

The impact of Talin2, a signaling protein regulating the focal adhesion complex, on asthma.

Talin protein as a mechanosensitive cytoskeleton protein connects the extracellular matrix to the cytoskeleton by linking to integrins and actin, thereby mediating the conversion of mechanical signals into biochemical signals and influencing disease progression. The biological significance of Talin2 in asthma is not well understood. The aim of this study was to elucidate the role of Talin2 in asthma. Mice sensitized and challenged with ovalbumin (OVA) or saline and MRC-5 cells were used to investigate the role of Talin2 in the pathogenesis of bronchial asthma. In addition, Talin2 levels were m…

Effects of actin remodeling inhibitors on cellular energy metabolism of a model marine bivalve, the Pacific oyster.

Actin, the most abundant cellular protein, is essential for maintaining structural organization, mechanical stability and cellular motility. The actin cytoskeleton undergoes continuous ATP-dependent reorganization, incurring significant energy costs through treadmilling. However, experimental quantifications of these energy expenditures, especially in ectotherms, remain scarce. In this study, we assessed the energy costs of actin remodeling in the Pacific oyster Crassostrea [also Magallana] gigas, a marine bivalve, by measuring oxygen consumption in the presence of inhibitors of actin treadmil…
