Tool Use Modulates Somatosensory Cortical Processing in Humans.

Tool use leads to plastic changes in sensorimotor body representations underlying tactile perception. The neural correlates of this tool-induced plasticity in humans have not been adequately characterized. This study used ERPs to investigate the stage of sensory processing modulated by tool use. Somatosensory evoked potentials, elicited by median nerve stimulation, were recorded before and after two forms of object interaction: tool use and hand use. Compared with baseline, tool use-but not use of the hand alone-modulated the amplitude of the P100. The P100 is a mid-latency component that inde

Relating experimentally-induced fear to pre-existing phobic fear in the human brain.

While prior work has demonstrated that fear-conditioning changes the neural representation of previously neutral stimuli, it remains unknown to what extent this new representation abstracts away from specific fears and which brain areas are involved therein. To investigate this question, we sought commonalities between experimentally-induced fear via electric shocks and pre-existing phobia. Using functional MRI, we tested the effect of fear-conditioning pictures of dogs in 21 spider-fearful participants across three phases: baseline, post-conditioning, and extinction. Considering phobic stimul

Cytisine inhibits the protective activity of various classical and novel antiepileptic drugs against 6 Hz-induced psychomotor seizures in mice.

BACKGROUND: Cytisine (CYT) is a partial agonist of brain α4β2 nicotinic acetylcholine receptors widely used in Central/Eastern Europe for smoking cessation. OBJECTIVES: This study evaluated the effect of CYT on the ability of classical and novel antiepileptic drugs to prevent seizures evoked by the 6-Hz test, a model of psychomotor seizures in mice thought as a model of drug-resistant seizures. RESULTS: CYT administered intraperitoneally (i.p.) in a dose of 2 mg kg-1 significantly inhibited the anticonvulsant activity of lacosamide, levetiracetam, and pregabalin, increasing their median effe

Peripheral and central CB1 cannabinoid receptors control stress-induced impairment of memory consolidation.

Stressful events can generate emotional memories linked to the traumatic incident, but they also can impair the formation of nonemotional memories. Although the impact of stress on emotional memories is well studied, much less is known about the influence of the emotional state on the formation of nonemotional memories. We used the novel object-recognition task as a model of nonemotional memory in mice to investigate the underlying mechanism of the deleterious effect of stress on memory consolidation. Systemic, hippocampal, and peripheral blockade of cannabinoid type-1 (CB1) receptors abolishe

Cue-induced resumption of heroin and cocaine seeking in rats using a conflict model of abstinence and relapse.

RATIONALE AND OBJECTIVES: Most animal research on drug relapse involves the reinstatement model where abstinence is a result of drug removal (extinction). However, abstinence in humans often results from the aversive consequences that accompany drug seeking (conflict situation). This study was aimed at using a conflict-based animal model of abstinence/relapse in rats self-administering heroin or cocaine. METHODS: Rats were trained to self-administer heroin (0.05 mg kg(-1) injection(-1)) or cocaine (0.5 mg kg(-1) injection(-1)) with each injection paired with a light cue. After stable respondi

Exposure to the context and removing the unpredictability of the US: two methods to reduce contextual anxiety compared.

Chronic anxiety may differ from cued fear and hence require other treatment strategies. In a human fear conditioning paradigm, chronic anxiety to the experimental context was experimentally induced by presenting unpredictable shocks. Two methods to reduce chronic anxiety were tested and compared. First, in parallel with the standard extinction procedure, participants were exposed to the anxiety-eliciting context in the absence of shocks (context-exposure group). Second, an alternative procedure was tested in which the previously unpredictable shocks were now signaled by a specific cue (signale

Levetiracetam in submaximal subcutaneous pentylentetrazol-induced seizures in rats.

Despite anticonvulsant efficacy in animal models of generalized epilepsy, levetiracetam was not effective in the maximal subcutaneous PTZ model in mice and rats. Aim of this study was to assess the efficacy of levetiracetam (LEV) against submaximal, s.c. MET test (PTZ at the dose of 70 mg/kg) acute seizures in Wistar rats, in comparison to valproic acid (VPA). Thirty male Wistar rats (P42) were divided in three drug-treatment groups (10 rats in each group) as follows: valproic acid, levetiracetam, and controls. All animals were tested for seizure threshold at age P50. VPA (110 mg/kg) and LEV (

Neuromuscular effects of stun device discharges.

BACKGROUND: Stun guns or electromuscular incapacitation devices (EMIs) generate between 25,000 and 250,000 V and can be discharged continuously for as long as 5 to 10 min. In the United States, over 200,000 individuals have been exposed to discharges from the most common type of device used. EMI devices are being used increasingly despite a lack of objective laboratory data describing the physiological effects and safety of these devices. An increasing amount of morbidity, and even death, is associated with EMI device use. To examine this type of electrical injury, we hypothesized that EMI dis

Effect of antihormones in amygdala-kindled seizures in rats.

Tamoxifen (TXF; an antiestrogen), cyproterone acetate (CYP; an antiandrogen) and mifepristone (MIF; an antigestagen) did not affect kindling parameters (afterdischarge threshold, seizure severity, seizure duration and afterdischarge duration) in fully-kindled rats. TXF (50 mg/kg) and CYP (50 mg/kg), when combined with carbamazepine, or phenobarbital, both antiepileptics administered at their highest subprotective doses of 15 mg/kg, resulted in significant reduction of the seizure and afterdischarge durations, both in male and female rats. Additionally, the combination of carbamazepine and cypr

Acquisition deficit and time-dependent retrograde amnesia for contextual fear conditioning in agmatine-treated rats.

Cumulative evidence indicates that the hippocampus plays a time-limited role in contextual learning paradigms. Pharmacological studies have indicated that acquisition of background contextual cues during Pavlovian fear conditioning is dependent upon hippocampal function, whereas early inactivation of the hippocampus after training produces retrograde amnesia. When administered prior to contextual fear conditioning, agmatine (5 and 10 mg/kg, i.p.), an endogenous polyamine and N-methyl-D-aspartate (NMDA) receptor ligand found at excitatory synapses in the hippocampus, impaired the acquisition of

Effects of C-type natriuretic peptide on pituitary-adrenal activation in rats.

The effects of C-type natriuretic peptide (CNP) on the hypothalamo-pituitary-adrenal system response to different stressors was studied. Various doses of CNP (0.2, 2, 4 microg) were injected into the lateral cerebral ventricle of freely moving rats 30 min before stress and activation of the adrenal was measured by plasma corticosterone. CNP did not affect basal corticosterone secretion in the doses applied, but inhibited in a dose-dependent manner the increase in plasma corticosterone induced by ether stress, electric shock and restraint. CNP exerted a more profound inhibitory effect on the re

Active and passive avoidance learning in rats neonatally treated with intraventricular 6-hydroxydopamine.

To clarify the behavioral characteristics of rats neonatally treated with 6-hydroxydopamine (6-OHDA), their performance on 4 aversive learning tasks, 3 active (shuttle, one-way, and rearing) avoidance tasks and one passive (step-through) avoidance task, was examined. On days 2 and 4 after birth, each rat of F344/Du strain received bilateral intraventricular injections of 6-OHDA (35 micrograms x 2) or vehicle solution following desmethylimipramine (20 mg/kg, s.c.) pretreatment. From day 90, each rat was trained in one of the 4 avoidance tasks. 6-OHDA-treated rats showed significantly less avoid

Isoniazid-induced inhibition of GABAergic transmission enhances neurosteroid content in the rat brain.

Isoniazid (375 mg/kg, s.c.), a drug that decreases GABAA receptor-mediated transmission, elicited a time-dependent increase of neuroactive steroid (pregnenolone, progesterone and allotetrahydrodeoxycorticosterone) concentrations in rat brain and plasma. This treatment also time-dependently increased the plasma concentration of corticosterone. Brain and plasma neuroactive steroid levels peaked between 40 and 120 min after isoniazid administration, respectively, and returned to control values by 5 hr. Acute foot shock stress mimicked the effect of isoniazid by increasing in a time-dependent mann

Effect of prenatal ethanol and stress on levels of beta-endorphin in different brain regions of the rat.

The combination of prenatal ethanol exposure and footshock stress was investigated for its effects on brain beta-endorphin levels. Subjects were offspring of rats that received 1 of 3 prenatal dietary treatments: an ethanol-containing liquid diet, a identical liquid diet with ethanol substituted isocalorically with maltose-dextrin (pair-fed group), and standard laboratory rat chow (chow-fed group). Two different stress paradigms were used: a short (30-sec) footshock stress paradigm and a prolonged (180-sec) footshock stress paradigm. Levels of beta-endorphin were measured with radioimmunoassay

A selective suppression of human pain sensitivity by carbon dioxide: central mechanisms implicated.

The effect of breathing CO2 on somatic sensitivity was studied in human subjects. Healthy humans breathed room air, 100% O2, or CO2 (5% or 8% in O2). Thresholds to heat pain, mechanical pain, electrically evoked pain, and electrically evoked perception were measured using psychophysical techniques. Also, the effects on sensory and affective components of experimental ischaemic pain, alertness and on cardiovascular parameters were observed. In addition, the effect of CO2 on heat pain threshold was determined in the ischaemic limb to exclude the possibility that the threshold elevation was due t

The effect of electroconvulsive shock and nifedipine on spatial learning and memory in rats.

Several traumatic events including brain contusion, electroconvulsive shock therapy, epileptic seizures and others, may cause short-term retrograde amnesia. In spite of much recent attention, pharmacological treatment of memory impairment has not been fully successful. In the present paper we report on the possible antiamnesic action of the L-type calcium channel blocker, nifedipine. Rats trained in the spatial memory task showed gradual improvement in the escape latency to find the submerged platform. After completion of the learning, they also showed a strong spatial bias toward the place th

The parabrachial region as a possible region modulating simultaneously pain and tonic immobility.

Unilateral microinjection of carbachol (1 microgram) into the dorsal parabrachial region (PBR) of conscious guinea pigs produced a 100% increase in the duration of restraint-induced tonic immobility (TI) episodes. In another group of animals with a subcutaneous electrode introduced into the thigh region, microinjections of equivalent doses of the same drug in similar sites also significantly reduced the motor defense and vocalization responses elicited by the application of a noxious electrical stimulus to the skin. Both effects were blocked by pretreatment with atropine. The possible simultan
