Brain functional changes following electroacupuncture in a mouse model of comorbid pain and depression: A resting-state functional magnetic resonance imaging study.

OBJECTIVE: Comorbid pain and depression are common but remain difficult to treat. Electroacupuncture (EA) can effectively improve symptoms of depression and relieve pain, but its neural mechanism remains unclear. Therefore, we used resting-state functional magnetic resonance imaging (rs-fMRI) to detect cerebral changes after initiating a mouse pain model via constriction of the infraorbital nerve (CION) and then treating these animals with EA. METHODS: Forty male C57BL/6J mice were divided into 4 groups: control, CION model, EA, and sham acupuncture (without needle insertion). EA was performe

Sex differences in baseline and drug-induced behavioural responses in classical behavioural tests.

Behavioural pharmacology relies on animal models which are primarily validated using the male laboratory rat. Many researchers solely employ male animals in studies; this is primarily due to concerns about the impact of variations in the female estrous cycle on behavioural responses. The objective of the present study therefore was to examine whether sex has any effect in some commonly employed behavioural pharmacology tests. Male and female Sprague Dawley rats were examined in the following behavioural pharmacology tests: diazepam (DZP) effects on anxiolytic behaviour in the elevated plus maz

Inhibition of the ERK1/2 Phosphorylation by Dextromethorphan Protects against Core Autistic Symptoms in VPA Induced Autistic Rats: In Silico and in Vivo Drug Repurposition Study.

The imbalance between excitatory and inhibitory neurotransmitters is explicitly related to the pathophysiology of autism spectrum disorder (ASD). The role of an NMDA receptor antagonist, dextromethorphan, was studied in ameliorating the ASD-like symptoms by regulating the excitatory and inhibitory imbalance using the valproic acid (VPA) model of ASD. Female Wistar rats were administered VPA [600 mg/kg on embryonic day ED-12.5] through intraperitoneal (ip) injection to induce ASD in pups. Autistic pups were then given dextromethorphan (10, 15, and 30 mg/kg; ip) and risperidone (2.5 mg/kg; ip) f

Diet-induced obesity progressively alters cognition, anxiety-like behavior and lipopolysaccharide-induced depressive-like behavior: focus on brain indoleamine 2,3-dioxygenase activation.

Obesity is associated with a high prevalence of mood symptoms and cognitive dysfunctions that emerges as significant risk factors for important health complications such as cardiovascular diseases and type 2 diabetes. It is therefore important to identify the dynamic of development and the pathophysiological mechanisms underlying these neuropsychiatric symptoms. Obesity is also associated with peripheral low-grade inflammation and increased susceptibility to immune-mediated diseases. Excessive production of proinflammatory cytokines and the resulting activation of the brain tryptophan cataboli

Investigation of the effect of berberine on memory and learning in aged rats undergoing sevoflurane anesthesia.

BACKGROUND: Sevoflurane is a widely used inhalation anesthetic during surgical procedures. Experimental animal models have demonstrated that exposure to sevoflurane leads to cognitive dysfunction. The molecular mechanisms underlying sevoflurane-induced neurotoxicity, as well as the potential neuroprotective mechanisms of berberine, have not yet been fully elucidated. OBJECTIVES: The current investigation sought to evaluate the effects of berberine on memory and learning in aged rats undergoing sevoflurane anesthesia. MATERIAL AND METHODS: Fifty-six male Sprague Dawley rats aged 20-24 weeks w

Antioxidant Effects of Vitamin E and L-Carnitine on Hippocampal Histology, Epigenetics, and Cognition in Morphine-Exposed Mice.

Chronic exposure to morphine induces oxidative stress and alters the hippocampal expression of genes encoding enzymes involved in epigenetic modifications (e.g., DNA methyltransferases; DNMTs and Histonedeacetylases; HDACs) and apoptosis-related signaling (e.g., Bax and Bcl-2). This study examined the effects of vitamin E and L-carnitine supplementation on cognitive function and hippocampal gene expression following morphine administration in adult male NMRI mice. Morphine was administered subcutaneously twice daily at escalating doses: 10 mg/kg for 10 days, 20 mg/kg from day 10 to 20, and 30

The influence of various glutamate receptors antagonists on anxiety-like effect of ethanol withdrawal in a plus-maze test in rats.

The aim of the present study was to determine whether various glutamate receptor antagonists could affect ethanol withdrawal-induced anxiety-like behavior measured in the elevated plus-maze test in rats. In our study, memantine (8 and 12 mg/kg), a noncompetitive N-methyl-d-aspartate (NMDA) receptor antagonist, did not show any effect on ethanol withdrawal anxiety. Acamprosate (NMDA and metabotropic glutamate5 (mGlu5) receptor antagonist), at a dose of 400 mg/kg showed anxiolytic-like effect, thus increasing the percent of time spent in open arms and open arms entries. Effects of pre-gestational exposure to the stressors and perinatal mirtazapine administration on the excitability of hippocampal glutamate and brainstem monoaminergic neurons, hippocampal neuroplasticity, and anxiety-like behavior in rats.

When accompanied by excessive exposure to the stressors, pregnancy may result in prenatal depression, that has in turn negative influence on the offspring's brain. Mirtazapine, among other antidepressants, is commonly used to treat prenatal depression. Even though mirtazapine is generally considered safe for pregnant women, its effect on the offspring brain have not been sufficiently investigated. The present study aimed to examine the effects of chronic unpredictable stress (CUS) in pregestational rats, perinatal mirtazapine treatment, and their combination, on offspring behavior and brain fu

Behavioral Effects of Renin-Angiotensin System Modulators in a Rotenone-Induced Model of Parkinson's Disease in Wistar Albino Rats.

INTRODUCTION: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor and non-motor dysfunction resulting from dopaminergic neuronal loss. Emerging evidence suggests a role of the renin-angiotensin system (RAS) in neurodegeneration, making it a potential therapeutic target. The present study aimed to evaluate the behavioral effects of captopril, perindopril, and losartan on motor, behavioral, and anxiety parameters in a rotenone-induced model of Parkinson's disease in Wistar albino rats. MATERIALS AND METHODS: An experimental in vivo study was conducted usi

Synergistic Effects of Levodopa, Benserazide, and Nortriptyline on Behavioral Impairments and Brain Pathology in an Experimental Rat Model of Parkinson's Disease.

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the degeneration of dopaminergic neurons in the substantia nigra pars compacta (SNpc). In addition to postural instability, rigidity, tremor, and bradykinesia, patients will experience depression and/or anxiety at any time during PD. Nortriptyline, as a dual reuptake inhibitor of norepinephrine and serotonin, inhibits alpha-synuclein aggregation and may play an important role in improving the pathological effects of PD. OBJECTIVE: This study investigated the effects of nortriptyline combined with
