PSRC1 overexpression attenuates atherosclerosis progression in apoE-/- mice by modulating cholesterol transportation and inflammation.

AIMS: Human genome-wide association studies (GWAS) have found that proline/serine-rich coiled-coil 1 (PSRC1) encodes a protein that is associated with serum lipid levels and coronary artery disease. In addition, our previous study showed that the cholesterol efflux capacity is decreased in macrophages following a treatment silencing Psrc1, indicating that PSRC1 has anti-atherosclerotic effects. However, the role of PSRC1 in the development of atherosclerosis is unknown. This study aims to explore the effect of PSRC1 on atherosclerosis and its underlying mechanisms. METHOD AND RESULTS: A recom

A biocompatible tissue scaffold produced by supercritical fluid processing for cartilage tissue engineering.

Supercritical fluids are used in various industrial fields, such as the food and medical industries, because they have beneficial physical and chemical properties and are also nonflammable and inexpensive. In particular, supercritical carbon dioxide (ScCO(2)) is attractive due to its mild critical temperature, pressure values, and nontoxicity. Poly(L-lactide-co-ɛ-caprolactone) (PLCL), which is a biocompatible, biodegradable, and very elastic polymer, has been used in cartilage tissue engineering. However, organic solvents, such as chloroform or dichloromethane, are usually used for the fabrica

FTO-Catalysed Demethylation of LUR1 mRNA Suppresses Macrophage Lipid Accumulation and Aortic Atherosclerosis.

Lipid deposition and foam cell formation drive atherogenesis. Lipid uptake regulator 1 (LUR1) and N6-methyladenosine (m6A) modification play key roles in systemic lipid homeostasis. Bioinformatic analyses revealed a negative correlation between fat mass and obesity-associated protein (FTO) and LUR1 expression and abundant m6A sites within the LUR1 mRNA strand. We therefore hypothesized that FTO-catalysed demethylation modulates LUR1 expression and lipid accumulation in macrophages, thereby influencing aortic plaque development. Acetylated low-density lipoprotein-loaded THP-1 macrophages were t

Lysophosphatidylcholine as a mechanistic and therapeutic nexus in atherosclerotic cardiovascular disease.

Atherosclerotic cardiovascular disease (ASCVD) remains the leading cause of morbidity and mortality worldwide despite intensive lipid-lowering therapy. Residual cardiovascular risk persists even in patients achieving optimal lipoprotein-cholesterol (LDL-C) levels, indicating that additional lipid mediators contribute to disease progression. Lysophosphatidylcholine (LPC), a bioactive lysophospholipid generated from phosphatidylcholine by cytosolic or lipoprotein-associated phospholipase A₂ (PLA₂), has emerged as a critical mediator linking dyslipidemia, inflammation, and vascular injury. LPC is

Probing Terra Incognita of Ni-P Catalysts: Operando Explorations during Hydrogen Evolution Reaction.

We have developed two Ni phosphide preparation methods allowing operando XAS surface-sensitive studies of well-defined bulk systems. For Ni K-edge XAS, a Ni2P phase-pure powder was sintered into a high-density pellet and polished for grazing incidence XAS. Ni sites were mildly affected by the acidic electrolyte prior to the HER, while the applied cathodic potential caused the reduction of Ni surface states beyond the states of as-prepared Ni2P. The computed fully H-covered Ni2P [0001] model describes the difference in the operando Ni K-edge GIXAS spectrum well. Upon turning the applied bias of

Sex-dependent acute oral toxicity and in vivo distribution of 6:2 chlorinated polyfluorinated ether sulfonate (6:2 Cl-PFESA) in rats.

6:2 Chlorinated polyfluorinated ether sulfonate (6:2 Cl-PFESA), a widely used substitute for perfluorooctane sulfonic acid (PFOS), has raised increasing concern due to its environmental persistence and potential health risks. However, its acute toxicity profile and sex-dependent effects remain insufficiently characterized. This study conducted a 14-day acute oral toxicity study for single exposure (46.4-464 mg/kg·BW) in male and female Sprague-Dawley (SD) rats to quantify lethality and evaluate sex-dependent differences. The median lethal dose (LD50) was estimated to be 147 mg/kg·BW for both s

Performance of a novel negative pressure wound therapy filler in wound contraction.

BACKGROUND: Negative pressure wound therapy promotes wound healing through fluid removal, edema reduction, and mechanical contraction. A novel thermoplastic elastomer (TPE) dressing has been developed to improve exudate evacuation and reduce tissue ingrowth, but its effect on wound contraction compared with reticulated open cell foam (ROCF) remains unclear. OBJECTIVE: To compare the degree of wound contraction with the TPE dressing, ROCF, or no filler at different wound depths and negative pressure settings in a porcine explant model. MATERIALS AND METHODS: The effect of the materials or lac

[Pathological mechanisms and traditional Chinese medicine treatment of atherosclerosis from perspective of "deficiency, stasis, and toxicity" spatiotemporal evolution].

Atherosclerosis(AS) is a major cause of cardiovascular and cerebrovascular diseases, with plaque formation and rupture commonly resulting in severe acute cardiovascular events. This study elucidated the spatiotemporal correlations between modern pathological mechanisms of AS and the evolution of traditional Chinese medicine(TCM) pathogenesis from the perspective of the "deficiency, stasis, and toxicity" theory. The temporal progression revealed that AS developed in three dynamic stages, including "deficiency damage" "stasis damage" and "toxicity damage".

Comparison of Novel Chest Seal Designs to Commercially Available Chest Seals at Relevant Physiological Pressures.

BACKGROUND: Tension pneumothorax is a leading cause of preventable death in combat scenarios. When treating a chest wound with the potential for open hemopneumothorax using a chest seal, it is important that it efficiently drain fluid from the chest cavity. We tested the ability of commercial and novel chest seal designs to drain fluid from a simulated chest wound. METHODS: Eight novel laminar chest seal designs were created and compared to six commercially available chest seals. Closed-cell foam with a hole was used to simulate a chest wound. Fluid pressures of 10, 30, and 100cmH2O were test

Current Knowledge of the Impact of Vitamin D in Coronary Artery Disease.

Coronary artery disease and vitamin D deficiency are both widespread conditions with a high incidence worldwide. Coronary artery disease is a complex illness with variable manifestation and pathogenesis. It often involves the development of atherosclerosis, and it frequently has serious or even fatal consequences for the patient. Vitamin D receptor expression is found in many tissues throughout the body, which results in a broad effect of the vitamin. Studies have found correlations between vitamin D deficiency and the development of coronary artery disease as well as other cardiovascular dise

Targeting LDL aggregation decreases atherosclerotic lipid burden in a humanized mouse model of familial hypercholesterolemia: Crucial role of ApoB100 conformational stabilization.

BACKGROUND AND AIMS: Low-density lipoprotein (LDL) aggregation is nowadays considered a therapeutic target in atherosclerosis. DP3, the retro-enantio version of the sequence Gly1127-Cys1140 of LRP1, efficiently inhibits LDL aggregation and foam cell in vitro formation. Here, we investigate whether DP3 modulates atherosclerosis in a humanized ApoB100, LDL receptor (LDLR) knockout mice (Ldlr-/-hApoB100 Tg) and determine the potential LDL-related underlying mechanisms. METHODS: Tg mice were fed an HFD for 21 days to induce atherosclerosis and then randomized into three groups that received a dai

Giant Arachnoid Granulations: Diagnostic Workup and Characterization in Three Symptomatic Adults.

Giant arachnoid granulations (GAGs) are poorly investigated. Here, we document clinical findings associated with five new GAGs and illustrate the anatomical composition of these structures as well as diagnostic considerations in three symptomatic adults. The GAGs ranged from 1.1 to 3.6 cm (mean, 2.2 cm) in maximum dimension and manifested in middle-aged individuals who presented with long-standing brain mass and/or chronic headache. On imaging examinations, the tissues appeared as irregular parasagittal and/or perisinus structures that demonstrated heterogeneous internal elements. The GAGs abu

Vimentin is an endogenous ligand for the pattern recognition receptor Dectin-1.

AIMS: Atherosclerosis is a chronic inflammatory disorder of cholesterol deposition in monocyte-derived macrophages (MDM) within the arterial wall leading to impingement on the lumen of the vessel. In atherosclerotic lesions, MDM are the primary source of NADPH oxidase-derived superoxide anion (O₂⁻) inducing low-density lipoprotein (LDL) oxidation leading to their unregulated uptake of oxidized LDL and foam cell formation. We recently discovered that zymosan potently activates monocyte NADPH oxidase via the non-toll pattern recognition receptor (PRR), Dectin-1. Other PRRs bind endogenous human

Experimental diet-induced atherosclerosis in Quaker parrots (Myiopsitta monachus).

Spontaneous atherosclerosis is common in psittaciformes, and clinical signs associated with flow-limiting stenosis are encountered in pet birds. Nevertheless, a psittacine model of atherosclerosis has not been developed for research investigations. Sixteen captive-bred Quaker parrots (Myiopsitta monachus) were used in this study. While 4 control birds were fed a maintenance diet, 12 other birds were fed an atherogenic diet composed of 1% cholesterol controlling for a calorie-to-protein ratio for periods ranging from 2 to 8 months. The birds were euthanized at the end of their respective food t

Host-Microbiota Metabolic Interactions in Atherosclerosis: Oral, gut, and Blood Perspectives.

Atherosclerosis is a chronic inflammatory disease influenced by host-microbiota interactions beyond traditional risk factors. Microbial communities in the oral cavity, gut, and blood contribute to vascular dysfunction through metabolic and immune mechanisms, yet an integrated perspective across these compartments remains lacking. This narrative review synthesizes current evidence on the distinct and interconnected roles of oral, gut, and blood microbiotas in atherosclerosis pathogenesis. We critically evaluate key microbial metabolites, trimethylamine N-oxide (TMAO), short-chain fatty acids (S

The hidden burden of kidney damage in chylomicronemia syndromes.

BACKGROUND: It has been recognized that hypertriglyceridemia (HTG) is associated with kidney damage. Monogenic and polygenic causes of extreme HTG characterized by severe chylomicronemia (TG > 885 mg/dl or 10 mmol/L) constitute unique models to investigate the potential nephrotoxic effects of sustained and severe exposure to HTG. METHODS: This study had two components: (1) kidney biopsies of three patients with severe chylomicronemia and proteinuria were analyzed; (2) a cross-sectional review of the medical files of two independent cohorts of patients with the Familial Chylomicronemia Syndrom

Long term rebound and transport of perfluorinated sulfonic acids (PFSAs) in the vadose zone following flushing.

Increases in perfluorinated sulfonic acid (PFSA) porewater concentrations over a 35 month duration following in situ flushing were monitored at an aqueous film-forming foam (AFFF) site using porous cup suction lysimeters within a highly instrumented test cell. Results provided evidence that perfluorooctane sulfonate (PFOS) slow desorption kinetics contributed to slow contaminant rebound in measured porewater concentrations. PFSAs in the shallow (0.23 m depth) highly PFSA-impacted soils migrated downward during the monitored post-flushing period, with short-chained PFSAs migrating more rapidly

Nanozyme-Based Therapeutic Strategies in Atherosclerosis: Mechanistic Insights, Targeting Approaches and Translational Challenges.

Atherosclerosis is a gradual process that occurs on the artery walls caused by multiple drivers, such as the presence of oxidative stress, chronic inflammation and disorganized lipid metabolism resulting in increased plaque formation and life-threatening cardiovascular disease. Due to their poor bioavailability, rapid clearance from the body and inability to maintain catalytic activity over time, traditional antioxidant drugs have reported minimal clinical success. Conversely, nanozyme-based therapeutics are being investigated as a new approach to treat atherosclerosis. Nanozymes are specially

Chitosan Oligosaccharides Modulate Macrophage Inflammatory Signaling: Molecular Docking and Transcriptomic Evidence.

Oxidative stress-induced macrophage activation and vascular injury are major contributors to atherosclerosis, a chronic inflammatory disease associated with approximately 20 million deaths worldwide. This study investigated the antioxidant and anti-inflammatory activities of structurally characterized low-molecular-weight chitosan oligosaccharides (COS) derived from mud crab (Scylla olivacea) shell waste by hydrochloric acid hydrolysis and explored their molecular interactions with inflammation-related targets. Structural characterization by 13C-NMR and MALDI-TOF confirmed the identity of COS,

Stroke and Vascular Cognitive Impairment: The Role of Intestinal Microbiota Metabolite TMAO.

The gut microbiome interacts with the brain bidirectionally through the microbiome-gutbrain axis, which plays a key role in regulating various nervous system pathophysiological processes. Trimethylamine N-oxide (TMAO) is produced by choline metabolism through intestinal microorganisms, which can cross the blood-brain barrier to act on the central nervous system. Previous studies have shown that elevated plasma TMAO concentrations increase the risk of major adverse cardiovascular events, but there are few studies on TMAO in cerebrovascular disease and vascular cognitive impairment. This review
