Plant resistance to necrotrophic fungi is regulated by a complex set of signaling pathways that includes those mediated by the hormones salicylic acid (SA), ethylene (ET), jasmonic acid (JA), and abscisic acid (ABA). The role of ABA in plant resistance remains controversial, as positive and negative regulatory functions have been described depending on the plant-pathogen interaction analyzed. Here, we show that ABA signaling negatively regulates Arabidopsis (Arabidopsis thaliana) resistance to the necrotrophic fungus Plectosphaerella cucumerina. Arabidopsis plants impaired in ABA biosynthesis,

Atropa belladonna L. (Solanaceae) is a significant source of the pharmacologically important tropane alkaloids (TAs), particularly hyoscyamine and scopolamine. Although rolB and rolC from Rhizobium rhizogenes are known to enhance plant secondary metabolism, their functional specificities and combined actions in TAs biosynthesis remain unclear. Here, we generated transgenic A. belladonna overexpressing 35S::rolB, 35S::rolC, and 35S::rolB + rolC to systematically investigate their effects on TAs accumulation, plant morphology, and drought tolerance. Co-expression of rolB and rolC exhibited metab

Boreal and temperate perennial woody plants coordinate annual growth cycle with seasonal environmental changes with age-dependent phenological differences. However, the underlying molecular mechanisms remain limited. In this study, we demonstrated that the MADS-box transcription factor DAL1 in Larix kaempferi (Japanese larch) directly regulates the expression of PEBP family genes LkFT1 and LkMFT to govern the annual growth cycle. LkDAL1 binds to the LkFT1 promoter to repress its expression, and to the LkMFT promoter to activate its expression. LkFT1 is highly expressed in the dormant stage, wi

Weeds, pests, and pathogens are among the pre-harvest constraints in rice farming across rice-growing countries. For weed management, manual weeding and herbicides are widely practiced. Among the herbicides, glyphosate [N-(phosphonomethyl) glycine] is a broad-spectrum systemic chemical extensively used in agriculture. Being a competitive structural analog to phosphoenolpyruvate, it selectively inhibits the conserved 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) enzyme required for the biosynthesis of aromatic amino acids and essential metabolites in eukaryotes and prokaryotes. The SUPERMAN (SUP) proteins, which belong to the single C2H2 zinc finger proteins (ZFP) subclass, participate in various aspects of gene regulation in plant morphogenesis and stress response, but their role in melon (Cucumis melo) is still largely unknown. We identified a total of 28 CmSUP genes in the melon genome, all containing QALGGH conserved domain. Collinearity analysis showed that melon had several homologous gene pairs with Arabidopsis and tomato, indicating the gene duplication events during the evolution. Expression analyses in RT-qPCR and transcriptomic data showed that CmSUPs can

We previously isolated, by differential display and 5' RACE (rapid amplification of cDNA ends), cDNAs corresponding to genes activated following cryptogein treatment of tobacco cell suspensions, among them tcI 7 (tcI for tobacco cryptogein Induced), a gene encoding a beta-subunit of proteasome. Here, we report that tcl 7 was up-regulated in tobacco plants treated with elicitins (cryptogein and parasiticein) that have been shown to induce a systemic acquired resistance (SAR). Moreover, subsequent inoculation of tobacco with the pathogen Phytophthora parasitica var. nicotianae (Ppn) was shown to

Phytophthora cinnamomi Rands (Pc) is a hemibiotrophic oomycete and the causal agent of Phytophthora root rot (PRR) of the commercially important fruit crop avocado (Persea americana Mill.). Plant defense against pathogens is modulated by phytohormone signaling pathways such as salicylic acid (SA), jasmonic acid (JA), ethylene (ET), auxin and abscisic acid. The role of specific signaling pathways induced and regulated during hemibiotroph-plant interactions has been widely debated. Some studies report SA mediated defense while others hypothesize that JA responses restrict the spread of pathogens

Living organisms have developed finely regulated homeostatic networks to mitigate the effects of environmental fluctuations in transition metal micronutrients, including iron, zinc, and copper. In Saccharomyces cerevisiae, the tandem zinc-finger protein Cth2 post-transcriptionally regulates gene expression under conditions of iron deficiency by controlling the levels of mRNAs that code for non-essential ferroproteins. The molecular mechanism involves Cth2 binding to AU-rich elements present in the 3' untranslated region of target mRNAs, negatively affecting their stability and translation. Ara

Cutin matrices in the cuticle cover plant epidermis, facilitating plant adaptation to stressful environments. Although cutin biosynthesis is extensively explored in the model plant Arabidopsis thaliana, the molecular mechanism governing cutin biosynthesis in the agriculturally important crop bread wheat (Triticum aestivum L.) remains largely unknown. The aim of the study is the characterization of the function and transcriptional regulation of a wheat gene involved in cutin biosynthesis. Long-chain acyl-CoA synthetase TaLACS2 was identified as an essential component of the wheat cutin biosynth

A proof-of-concept reversible genetic logic circuit in tobacco plants is presented. The genetic circuit implements a Boolean NOR function using a Cas6-based translational repression system, with exposure to estradiol and ethanol as inputs, and expression of GFP as the output. Expressed in the presence of the inducers, two Cas6 proteins are used to selectively prevent the translation of GFP. The circuit yields a 40-90% reduction in GFP expression in the presence of the inducers. A mathematical model of the circuit's mechanism of action is proposed and validated using experimentally acquired dat

The extent to which physical properties and intranuclear locations of chromatin can influence transcription output remains unclear and poorly quantified. Because the scale and resolution at which structural parameters can be queried are usually so different from the scale that transcription outputs are measured, the integration of these data is often indirect. To overcome this limitation in quantifying chromatin structural parameters at different locations in the genome, a Chromatin Charting collection with 277 transposon-tagged Arabidopsis lines has been established in order to discover corre

PpAPRR2 promotes chlorophyll accumulation in peach fruit, and its overexpression enhances fruitgreening in tomato. Fruit peel color represents an important quality trait in peach (Prunus persica L.), determined by the dynamic balance between chlorophyll and anthocyanin metabolism. While the regulatory networks controlling anthocyanin regulation have been well documented, the molecular mechanisms governing chlorophyll accumulation and peel greening remain less understood. Here, we identified nine ARR-B family transcription factors in peach and found that PpAPRR2 was predominantly expressed duri

Tree peony (Paeonia suffruticosa) is an important garden plant with high ornamental and economic value. Bud endodormancy release is prerequisite for bud break, which affects its flowering time and quality, and activating gibberellin (GA) signaling is a key factor in this process. The DELLA protein PsRGL1 negatively regulates bud break, but its interacting transcription factors and their roles remain largely unexplored. Here, we identified a direct interacting partner of PsRGL1, PsWRKY71, using yeast two-hybrid, pull-down, and luciferase complementation assays. PsWRKY71 was significantly induce

BACKGROUND: Autophagy plays a central role in maintaining cellular homeostasis, regulating growth and development, and responding to multiple stresses. Autophagy-related genes (ATGs) play critical roles in autophagy, yet their functional diversity in soybean (Glycine max) remains underexplored.
METHODS: Genome-wide identification of GmATG genes was performed using sequence similarity and domain-based searches against the Wm82.gnm4 reference genome, followed by characterization of physicochemical properties, chromosomal distribution, phylogenetic relationships, gene duplication, conserved moti

Drought stress significantly impacts crop productivity and plant physiology. Nitric oxide (NO) signalling is essential for drought tolerance. This study explores the relationship between in vivo NO levels, mediated by NO scavenging phytoglobin (encoded by Pgb, non-symbiosis associated hemoglobin), and drought tolerance in oat (Avena sativa). Real-time in vivo NO measurements suggested increased production under moderate to high water stress in the susceptible cultivar Flega compared to the resistant Patones. This elevated NO correlated with increased senescence in Flega. Conversely, the resist

Plant height is a vital agronomic trait that greatly determines crop yields because of the close relationship between plant height and lodging resistance. Legumes play a unique role in the worldwide agriculture; however, little attention has been given to the molecular basis of their height. Here, we characterized the first dwarf mutant mini plant 1 (mnp1) of the model legume plant Medicago truncatula. Our study found that both cell length and the cell number of internodes were reduced in a mnp1 mutant. Using the forward genetic screening and subsequent whole-genome resequencing approach, we c

Phosphorus, one of the essential elements for plants, is often a limiting nutrient because of its low availability and mobility in soils. Significant changes in plant morphology and biochemical processes are associated with phosphate (Pi) deficiency. However, the molecular bases of these responses to Pi deficiency are not thoroughly elucidated. Therefore, a comprehensive survey of global gene expression in response to Pi deprivation was done by using Arabidopsis thaliana whole genome Affymetrix gene chip (ATH1) to quantify the spatio-temporal variations in transcript abundance of 22,810 genes.

Phosphate (Pi) is a critical nutrient for plants and is often a limiting factor in food production, as many agricultural soils are limited in available Pi. Inositol pyrophosphates (PP-InsPs) are signaling molecules involved in Pi sensing and jasmonic acid (JA)-regulated plant defense. Here, we report that overexpression of 1,3,4-trisphosphate 5/6-kinase 1 (ITPK1) and the kinase domain of the dual-domain diphosphoinositol pentakisphosphate kinase 2 (VIP2KD) in Arabidopsis thaliana results in unique elevations in PP-InsPs, accompanied by altered leaf growth and senescence patterns, as well as de

In a previous study, GmMYB14 overexpressing (GmMYB14-OX) transgenic soybean plants displayed a semi-dwarfism and compact phenotype, which was regulated by the brassinosteroid (BR) pathway. However, the phenotype of GmMYB14-OX plants could be partly rescued after spraying them with exogenous BR. This indicates that other hormones, in addition to BR, also play a role in regulating the architecture of GmMYB14-OX plants. We observed a significant decrease in the content of endogenous indole-3-acetic acid (IAA) in transgenic soybean lines (OX9 and OX12) compared to wild type (WT) plants. The plant

BACKGROUND: Verticillium wilt, causes mainly by the soilborne pathogen Verticillium dahliae, is a devastated vascular disease resulting in huge financial losses in cotton, so research on improving V. dahliae stress tolerance in cotton is the utmost importance. Calcium as the second messenger acts as a crucial role in plant innate immunity. Cytosolic Ca2+during the pathogen infection is a significant increase in plant immune responses. Calcineurin B-like (CBL) proteins are widely known calcium sensors that regulate abiotic stress responses. However, the role of cotton CBLs in response to V. dah
