A 2-year-old male domestic shorthair cat was presented for a progressive history of abnormal posture, behavior, and mentation. Menace response was absent bilaterally, and generalized tremors were identified on neurological examination. A neuroanatomical diagnosis of diffuse brain dysfunction was made. A neurodegenerative disorder was suspected. Magnetic resonance imaging findings further supported the clinical suspicion. Whole-genome sequencing of the affected cat with filtering of variants against a database of unaffected cats was performed. Candidate variants were confirmed by Sanger sequenc

In the United States, the most commonly diagnosed arboviral disease is West Nile virus (WNV) infection. Diagnosis is made by detecting WNV IgG or viral genomic sequences in serum or cerebrospinal fluid. To determine frequency of this testing in WNV-endemic areas, we examined the proportion of tests ordered for patients with meningitis and encephalitis at 9 hospitals in Houston, Texas, USA. We identified 751 patients (567 adults, 184 children), among whom 390 (52%) experienced illness onset during WNV season (June-October). WNV testing was ordered for 281 (37%) of the 751; results indicated acu

Deafblindness is part of several genetic disorders. We investigated a consanguineous Egyptian family with two siblings affected by congenital hearing loss and retinal degeneration, initially diagnosed as Usher syndrome type 1. At teenage, severe enamel dysplasia, developmental delay, and microcephaly became apparent. Genome-wide homozygosity mapping and whole-exome sequencing detected a homozygous missense mutation, c.1238G>T (p.Gly413Val), affecting a highly conserved residue of peroxisomal biogenesis factor 6, PEX6. Biochemical profiling of the siblings revealed abnormal and borderline plasm

Cells exposed to densely ionizing radiation (high-LET) experience more severe biological damage than do cells exposed to sparsely ionizing radiation (low-LET). The prevailing hypothesis is that high-LET radiations induce DNA double strand-breaks (DSB) that are more complex and clustered, and are thereby more challenging to repair. Here, we present experimental data obtained by atomic force microscopy imaging, DNA-dependent protein kinase (DNA-PK) activity determination, DNA ligation assays, and genomic studies to suggest that short DNA fragments are important products of radiation-induced DNA

Several studies suggest a correlation between genome architecture and gene function. To elucidate mechanisms of gene positioning during cell differentiation and malignant transformation we investigated the nuclear positions of the BCL2 alleles and chromosome 18 territories in different layers of nonneoplastic cervical squamous epithelium and cervical squamous carcinomas in relation to gene expression. Fluorescence in situ hybridization and three-dimensional (3D) image analysis using tissue sections revealed that one BCL2 allele was located more peripherally than the other one in nuclei of the

We report a sporadic case of congenital muscular dystrophy (CMD) in a 13-year-old girl with early manifestation of muscle weakness and hypotonia, severe contractures, bulbar syndrome, progressive external ophtalmoplegia, and white matter changes on magnetic resonance imaging (MRI) of the brain, but no mental defect. Serum creatine kinase (CK) level was normal. Muscle biopsy revealed a dystrophic picture with a prominent inflammatory infiltrate mimicking inflammatory myopathy-typical histological findings in CMD. Immunostaining showed normal expression of merosin, alpha and beta-dystroglycans.

A 3-month-old girl who developed severe measles encephalitis after neonatal measles is reported. Her mother had measles when she was ten days old and she was admitted to our hospital with low grade fever, Koplik spot, and mild exanthema seventeen days after birth, and she recovered in 7 days without any complication. At three months of age, she was readmitted because of intractable seizures. The levels of IgM and IgG antibodies against measles in the cerebrospinal fluid were elevated. The measles virus genome, amplifying the region encoding the nucleocapsid protein, was detected from the brain

BACKGROUND AND OBJECTIVES: Cerebral small vessel disease (cSVD), characterized by pathologic changes in the structure and function of small brain vessels, is detectable on brain MRI in the absence of clinical symptoms. However, imaging cerebral small vessels themselves in vivo remains costly and challenging. There is growing interest in investigating whether retinal microvascular imaging features could be proxies for changes in the brain microvasculature. Using a multipronged approach, we explored the relation of retinal microvascular characteristics with MRI markers of cSVD (MRI-cSVD).

INTRODUCTION: Primary breast lymphoma (PBL) is rare, and most cases occur in female patients, with few reported cases in male patients. The clinical presentation is similar to that of breast cancer, but the condition needs to be well understood, as treatment options and clinical course vary. Hence, we provide a relatively rare case of primary breast diffuse large B cell lymphoma (PB-DLBCL) in a male, including its complete clinicopathological features, radiological findings, genomic mutational profiles, and clinical course.

Brain metastases (BMs) are the most common intracranial tumors in adults. Their heterogeneity, potential multifocality, and complex biomolecular behavior pose significant diagnostic and therapeutic challenges. Artificial intelligence (AI) has the potential to revolutionize BM diagnosis by facilitating early lesion detection, precise imaging segmentation, and non-invasive molecular characterization. Machine learning (ML) and deep learning (DL) models have shown promising results in differentiating BMs from other intracranial tumors with similar imaging characteristics-such as gliomas and primar

BACKGROUND: Accurate prediction of recurrence-free survival (RFS) is important for the prognosis of cutaneous melanoma patients. The image-based pathological examination remains as the gold standard for diagnosis. It is of clinical interest to account for computer-aided processing of pathology image when performing prognostic analysis.
METHODS: We enrolled in this study a total of 152 patients from TCGA-SKCM (The Cancer Genome Atlas Skin Cutaneous Melanoma project) with complete information in recurrence-related survival time, baseline variables (clinicopathologic variables, mutation status o

Crohn's disease can progress from non-stricturing, non-penetrating inflammation (B1) to stricturing (B2) or penetrating (B3) complications, yet risk is commonly recognized only after structural damage is established. This narrative review examines clinical, endoscopic, genomic, circulating, stool, tissue, imaging, radiomic, and computational biomarkers relevant to behavioral-progression risk and separates direct B1-to-B2 or source-defined B3 evidence from related prognostic and mechanistic evidence, while distinguishing internal penetrating from perianal fistulizing disease when source reporti

Therapy resistance remains a leading cause of treatment failure and mortality in thoracic malignancies, despite major advances in targeted therapies and immunotherapies. Resistance evolves through heterogeneous, patient-specific mechanisms, including genetic alterations, epigenetic reprogramming, lineage plasticity, and tumor-microenvironment interactions, often emerging before radiographic progression or clinical relapse. Conventional tissue biopsies and imaging, while essential for diagnosis and treatment selection, are limited in their ability to capture spatial and temporal tumor heterogen

Appendiceal cancer is a rare and biologically distinct gastrointestinal malignancy characterized by marked molecular heterogeneity and a strong propensity for peritoneal dissemination. Traditional surveillance modalities, including cross-sectional imaging and serum tumor markers, often demonstrate limited sensitivity for detecting residual or recurrent peritoneal disease, creating a need for more effective biomarkers. Circulating tumor DNA (ctDNA) has emerged as a promising noninvasive tool for genomic profiling, prognostication, and disease monitoring across gastrointestinal (GI) cancers. In

BACKGROUND: Recently, a novel clinical picture associated with characteristic, progressive alterations in the ellipsoid zone (EZ) and thinning of the outer retinal layers referred to as Persistent Diffuse Photoreceptor Disorganization (PDPD) was first described.
METHODS: We performed detailed longitudinal clinical assessments in five patients with PDPD, i.e. ophthalmological examinations, including imaging (optical coherence tomography (OCT), fundus autofluorescence, dye angiography) and functional diagnostics (best-corrected visual acuity (BCVA), visual field, electroretinography), comparati

Quantitative detection of Escherichia coli in water is essential for assessing fecal contamination because of its potential adverse effects on human health. Polymerase chain reaction (PCR)-based techniques enable rapid detection, facilitating timely decision-making and public health interventions. However, conventional quantitative real-time PCR (qPCR) assays generally require expensive, non-portable equipment and are often inaccessible in resource-limited settings. Therefore, this study evaluated a simple endpoint PCR combined with SYBR Green I (SGI)-based colorimetric deoxyribonucleic acid (

Ubiquitin-conjugating enzymes (UBCs) serve as core components of the ubiquitin-proteasome pathway and extensively participate in plant development and stress responses. However, the regulatory mechanism of grape (Vitis vinifera L.) UBC family genes underlying berry ripening remains unclear. Genome‑wide screening identified 44 VviUBC members within the grape UBC gene family. Systematic characterization of physicochemical features, conserved motifs, gene structures, chromosomal locations, cis‑regulatory elements and evolutionary expansion uncovered whole‑genome duplication as the major driving f

Polyploid cells, which contain more than two copies of the genome, are widely present across plants and animals, where they are often found in tissues with high biosynthetic and metabolic demands, such as the mammalian liver and placenta. While somatic polyploidy is frequently associated with increased cell growth and biosynthetic capacity, unscheduled polyploidization in cell types that are not normally programmed to become polyploid is often linked to reduced cellular fitness and genome instability. To understand whether these divergent outcomes stem from distinct immediate cellular response

Enveloped viruses enter cells by membrane fusion. The viral membrane fuses with a host membrane, either at the cell surface or within endocytic compartments. For endocytic entry, fusion is typically triggered by low pH and often requires proteolytic priming by compartment-specific host proteases, which together define the site and mechanism of fusion and shape viral tropism. Inhibition of the lipid kinase PIKfyve, which generates PI(5)P and PI(3,5)P2 in late endosomes and lysosomes, swells those compartments and blocks infection by a subset of enveloped viruses, including Ebola virus, Marburg
