Centesimal composition and meat yield of Hoplias malabaricus: association with intestinal parasites.

Hoplias malabaricus is a non-migratory fish commonly found in the Mogi Guaçu River basin, mainly feeding on fish, small crustaceans and insects. It forms part of the diet for humans, birds and some mammals. This fish has great nutritional value, with both good quality and good quantities of essential vitamins and amino acids. Regarding parasitic fauna, this fish can host different species of helminths in its gastrointestinal tract. The aim of the present study was to investigate the possible interference of parasitism in the meat yield from H. malabaricus and the centesimal composition. For th

Gastrointestinal parasites in captive and free-living wild birds in Goiania Zoo.

Helminths are endoparasites that infect a variety of bird species. Endoparasite infections can cause severe diseases, including kill captive avian hosts and represents a problem to maintenance and conservation. The eggs and larval forms of these parasites are usually eliminated in the host feces. The main interest of this study is to report the occurrence of eggs and oocysts in feces from captive wild birds in Goiânia Zoo and free-living birds in its surroundings. The fecal samples were subjected to parasitological examination to identify the presence of helminths and to classify their eggs ba…

Diagnosis of bovine gastrointestinal parasites: comparison of different techniques and different solutions.

Cattle are important livestock species for protein and income supply. But disease agents, including gastrointestinal (GI) parasites, hinder their productivity. For effective GI parasites control, there is need for rapid, highly sensitive and specific diagnostic techniques and tools, lack of which poses problem to clinician, diagnostic staff and livestock owners. One hundred cattle faeces were analyzed through Simple Faecal Flotation (SFF), Modified Centrifugal Faecal Flotation (MCFF) and Mini-FLOTAC techniques (MFT) using sodium chloride, brine, sugar, salt/sugar and zinc sulphate at specific …

Stable isotopes unravel the feeding mode-trophic position relationship in trematode parasites.

Stable isotopes have been sporadically used over the last two decades to characterise host-parasite trophic relationships. The main reason for this scarcity is the lack of an obvious pattern in the ratio of nitrogen stable isotope values (δ15 N) of parasites in comparison to their host tissues, which would be key to understand any host-parasite system dynamics. To circumvent this, we focused on a single snail host, Zeacumantus subcarinatus, and three of its trematode parasites. We used stable isotopes to investigate each host-trematode trophic relationship and shed light on the mechanisms util…

Novel molecule combinations and corresponding hybrids targeting artemisinin-resistant Plasmodium falciparum parasites.

Malaria is still considered as the major parasitic disease and the development of artemisinin resistance does not improve this alarming situation. Based on the recent identification of relevant malaria targets in the artemisinin resistance context, novel drug combinations were evaluated against artemisinin-sensitive and artemisinin-resistant Plasmodium falciparum parasites. Corresponding hybrid molecules were also synthesized and evaluated for comparison with combinations and individual pharmacophores (e.g. atovaquone, mefloquine or triclosan). Combinations and hybrids showed remarkable antima…

Geographical spread and structural basis of sulfadoxine-pyrimethamine drug-resistant malaria parasites.

The global spread of sulfadoxine (Sdx, S) and pyrimethamine (Pyr, P) resistance is attributed to increasing number of mutations in DHPS and DHFR enzymes encoded by malaria parasites. The association between drug resistance mutations and SP efficacy is complex. Here we provide an overview of the geographical spread of SP resistance mutations in Plasmodium falciparum (Pf) and Plasmodium vivax (Pv) encoded dhps and dhfr genes. In addition, we have collated the mutation data and mapped it on to the three-dimensional structures of DHPS and DHFR which have become available. Data from genomic databas…

Predators in northern Germany are reservoirs for parasites of One Health concern.

Urbanisation and invasion of wildlife into urban areas as well as human leisure activities create diverse wildlife-domestic animal-human interfaces, increasing the risk of (zoonotic) parasite spillover from sylvatic to domestic and synanthropic cycles. This study investigated the endo- and ectoparasite fauna, emphasising on parasites of One Health Concern, of the most common predators in northern Germany between November 2013 and January 2016. Eighty red foxes (Vulpes vulpes), 18 stone martens (Martes foina) and nine raccoon dogs (Nyctereutes procyonoides) were available for the study. Overall…

How Many Is Enough? - Challenges of Multinucleated Cell Division in Malaria Parasites.

Regulating the number of progeny generated by replicative cell cycles is critical for any organism to best adapt to its environment. Classically, the decision whether to divide further is made after cell division is completed by cytokinesis and can be triggered by intrinsic or extrinsic factors. Contrarily, cell cycles of some species, such as the malaria-causing parasites, go through multinucleated cell stages. Hence, their number of progeny is determined prior to the completion of cell division. This should fundamentally affect how the process is regulated and raises questions about advantag…

Anti-PfGARP activates programmed cell death of parasites and reduces severe malaria.

Malaria caused by Plasmodium falciparum remains the leading single-agent cause of mortality in children1, yet the promise of an effective vaccine has not been fulfilled. Here, using our previously described differential screening method to analyse the proteome of blood-stage P. falciparum parasites2, we identify P. falciparum glutamic-acid-rich protein (PfGARP) as a parasite antigen that is recognized by antibodies in the plasma of children who are relatively resistant-but not those who are susceptible-to malaria caused by P. falciparum. PfGARP is a parasite antigen of 80 kDa that is expressed…

cAMP signalling and its role in host cell invasion by malaria parasites.

Cyclic adenosine monophosphate (cAMP) is an important signalling molecule across evolution, but until recently there was little information on its role in malaria parasites. Advances in gene editing - in particular conditional genetic approaches and mass spectrometry have paved the way for characterisation of the key components of the cAMP signalling pathway in malaria parasites. This has revealed that cAMP signalling plays a critical role in invasion of host red blood cells by Plasmodium falciparum merozoites through regulating the phosphorylation of key parasite proteins by the cAMP-dependen…

Linking nutrient sensing and gene expression in Plasmodium falciparum blood-stage parasites.

Malaria is one of the most life-threatening infectious diseases worldwide, caused by infection of humans with parasites of the genus Plasmodium. The complex life cycle of Plasmodium parasites is shared between two hosts, with infection of multiple cell types, and the parasite needs to adapt for survival and transmission through significantly different metabolic environments. Within the blood-stage alone, parasites encounter changing levels of key nutrients, including sugars, amino acids, and lipids, due to differences in host dietary nutrition, cellular tropism, and pathogenesis. In this revie…

Molecular surveillance and temporal monitoring of malaria parasites in focal Vietnamese provinces.

BACKGROUND: While the World Health Organization (WHO) Southeast Asia region has the second highest incidence of malaria worldwide, malaria in Vietnam is focal to few provinces, where delayed parasite clearance to anti-malarial drugs is documented. This study aims to understand Plasmodium species distribution and the genetic diversity of msp1 and msp2 of parasite populations using molecular tools. METHODS: A total of 222 clinical isolates from individuals with uncomplicated malaria were subjected to Plasmodium species identification by nested real-time PCR. 166 isolates positive for Plasmodium

Bloody flukes: a review of aporocotylids as parasites of cultured marine fishes.

Fish blood flukes (Digenea: Aporocotylidae) are important pathogens of fishes in aquaculture. Severe infections have been associated with mass mortality events in cultured marine species of teleosts in Australia, Asia and Europe, leading to significant socio-economic losses. Here we review recent advances towards understanding the biology and ecology of fish blood flukes, and the integral role molecular techniques have played in this development. Techniques include molecular matching of aporocotylid life stages using ITS-2 rDNA, and targeting ITS-2 rDNA to distinguish aporocotylid species usin

Transmission of Artemisinin-Resistant Malaria Parasites to Mosquitoes under Antimalarial Drug Pressure.

Resistance to artemisinin-based combination therapy (ACT) in the Plasmodium falciparum parasite is threatening to reverse recent gains in reducing global deaths from malaria. While resistance manifests as delayed parasite clearance in patients, the phenotype can only spread geographically via the sexual stages and mosquito transmission. In addition to their asexual killing properties, artemisinin and its derivatives sterilize sexual male gametocytes. Whether resistant parasites overcome this sterilizing effect has not, however, been fully tested. Here, we analyzed P. falciparum clinical isolat…

Prevalence of protozoan parasites in small and medium mammals in Texas, USA.

Wildlife interaction with humans increases the risk of potentially infected ticks seeking an opportunistic blood meal and consequently leading to zoonotic transmission. In the United States, human babesiosis is a tick-borne zoonosis most commonly caused by the intraerythrocytic protozoan parasite, Babesia microti. The presence of Babesia microti and other species of Babesia within Texas has not been well characterized, and the molecular prevalence of these pathogens within wildlife species is largely unknown. Small (e.g. rodents) and medium sized mammalian species (e.g. racoons) represent pote

Intracellular protozoan parasites: living probes of the host cell surface molecular repertoire.

Intracellular protozoans co-evolved with their mammalian host cells a range of strategies to cope with the composite and dynamic cell surface features they encounter during migration and infection. Therefore, these single-celled eukaryotic parasites represent a fascinating source of living probes for precisely capturing the dynamic coupling between the membrane and contractile cortex components of the cell surface. Such biomechanical changes drive a constant re-sculpting of the host cell surface, enabling rapid adjustments that contribute to cellular homeostasis. As emphasized in this review, …
