Novel de novo SPAST mutation in a Han Chinese SPG4 patient: a case report.

Spastic paraplegia type 4 (SPG4), the predominant form of Autosomal Dominant Hereditary spastic paraplegia (AD-HSP), is characterized by variants in the SPAST gene. This study reports a unique case of a late-onset SPG4 in a Han Chinese male, manifesting primarily as gait disturbances from lower extremity spasticity. Uncovered through whole-genome sequencing, a previously undocumented frameshift variant, c.1545dupA in exon 14 of the SPAST gene, was identified. Notably, this variant was absent in asymptomatic parents with confirmed paternity and maternity status, suggesting a de novo variant occ…

[Pseudopseudohypoparathyroidism and genomic imprinting].

Pseudopseudohypoparathyroidism (PPHP) is caused by the paternally-derived mutation in the coding region of GNAS gene. The phenotype of PPHP is produced by the sum of both decreased Gsalpha protein in biallelically expressed tissues and other proteins or non-coding RNAs by mutated paternal-allele specific expression. It has been believed that the haploinsufficiency of Gsalpha in non-imprinted tissues is responsible for the Albright hereditary dystrophy (AHO) phenotype. Recently it was reported that obesity is a more prominent feature in pseudohypoparathyroidism type I a than in PPHP. This resul

Skin-to-Skin Care and Spontaneous Touch by Fathers in Full-Term Infants: A Systematic Review.

A series of studies have shown that mothers' early tactile behaviors have positive effects, both on full-term and preterm infants, and on mothers alike. Regarding fathers, research has focused mostly on paternal skin-to-skin care with preterm infants and has overlooked the tactile behavior effects with full-term newborns on infants' outcomes and on fathers themselves. The current systematic review considered the evidence regarding paternal tactile behaviors with full-term infants, including skin-to-skin care (SSC) and spontaneous touch (ST), during parent-infant interactions, and differentiate

A novel p.Gly187Arg TP53 variant appears to result in Li-Fraumeni syndrome.

Li-Fraumeni syndrome is an autosomal dominant cancer syndrome characterized by pathogenic variants in the TP53 gene on chromosome 17. The most common cancers in Li-Fraumeni kindreds include sarcomas, breast cancer, brain tumors, and adrenocortical carcinoma. We report a 9-month-old male who was diagnosed with an adrenocortical tumor and later found to harbor a novel TP53 c.559 G > C germline variant, resulting in p.Gly187Arg. Family history included early-onset breast cancer in his paternal grandmother and paternal great-grandfather, as well as colon cancer at age 31 in a paternal cousin. The

Characterizing the association between parenting and adolescent social phobia.

OBJECTIVES: For characterizing the association between parenting and offspring social phobia (SP), contrasting maternal vs. paternal contributions, putative predictors of unfavorable parenting behaviors and its specificity for SP are warranted to delineate targeted prevention and intervention strategies. METHODS: A population-based sample of 1053 adolescents was followed-up using the M-CIDI. Parenting was assessed via questionnaire in offspring passing the high risk period for SP-onset. Natal complications and childhood serious health problems as assessed by maternal reports were hypothesized

Monogamy in a new species of wild guinea pigs (Galea sp.).

A new species of wild guinea pigs from Bolivia was recently discovered: Galeasp. So far no data are available concerning its social and mating systems. We therefore investigated behavioural compatibility, sexual dimorphism, paternity in a male-competing situation, testicular and epididymal size, and sperm morphology and motility. The results showed a complete intrasexual and moderate intersexual behavioural incompatibility between unfamiliar animals, a reversed sexual dimorphism with females larger than males, and litters always fathered by single males. Furthermore, both the reproductive orga…

Frequent de novo mutations of the ANK1 gene mimic a recessive mode of transmission in hereditary spherocytosis: three new ANK1 variants: ankyrins Bari, Napoli II and Anzio.

A subset of spherocytosis cases associated with mutations of the ANK1 gene present an apparently recessive inheritance pattern on a clinical … ations exhibited by their children. We confirmed the de novo character of the HS mutations based on paternity testing. Recessive HS associated with the ANK1 gene is probably rarer than initially thought, and spherocytosis may often be due to de novo mutations.

Gonadal mosaicism for incontinentia pigmenti in a healthy male.

Incontinentia pigmenti (IP) is a genodermatosis that segregates as an X linked dominant trait with male lethality. The disease has been linked to Xq28 in a number of studies. A few affected males have been documented, most of whom have a 47,XXY karyotype. We report a family with two paternally related half sisters, each affected with IP. The father is healthy, clinically normal, and has a 46,XY normal male karyotype. Linkage analysis of 12 polymorphic markers (two X linked and 10 autosomal) confirms paternity. X inactivation studies with the human androgen receptor (HUMARA) indicate that the p…

Meiotic recombination between two polymorphic restriction sites within the beta globin gene cluster.

Analysis of beta globin gene haplotypes for prenatal diagnosis of beta thalassaemia has revealed a recombination event within the beta globin gene cluster. Both a change in the AvaII polymorphic site within the beta globin gene and a change in the phenotype of the beta globin gene were observed. Paternity was established by the pedigree analysis of hypervariable 'minisatellite' DNA polymorphisms and the most probable explanation of the recombination event is a crossover between the psi beta globin gene and the beta globin gene. The data provide direct evidence in support of a DNA region 3' to …

[Cryptorchidism and fertility].

This article describes a fertility study based on 490 cases of surgically treated undescended testicles. Some of the patients were seen very early, and others later, even after puberty. The results are assessed as a function of the onset of pregnancy, or, failing this, on the basis of spermograms, and as a function of whether one or both testicles were undescended. Among the married patients with a unilateral complaint, the paternity rate was 69 p. cent, failing to 43 per cent for bilateral cases. Their spermograms showed an average count of 33.7 X 10(6)/ml for unilateral cryptorchids, and 3.6…

Stress and child internalizing symptoms: Examining the moderating role of Latino fathers' warmth and support.

Latino immigrants within the United States experience various stressors, which have been linked to the development and exacerbation of internalizing symptoms among Latino youth. Therefore, it is crucial to explore factors that may buffer the impact of stress among immigrant families. Fathers may influence child outcomes through positive parenting behaviors, yet Latino fathers have been underrepresented in research. This study examined how paternal warmth and support may moderate the association between the accumulation of stress and child internalizing symptoms among low-income, Latino immigra

Parental Folate Deficiency Inhibits Proliferation and Increases Apoptosis of Neural Stem Cells in Rat Offspring: Aggravating Telomere Attrition as a Potential Mechanism.

The effect of maternal folate status on the fetal central nervous system (CNS) is well recognized, while evidence is emerging that such an association also exists between fathers and offspring. The biological functions of telomeres and telomerase are also related to neural cell proliferation and apoptosis. The study aimed to investigate the effect of parental folate deficiency on the proliferation and apoptosis of neural stem cells (NSCs) in neonatal offspring and the role of telomeres in this effect. In this study, rats were divided into four groups: maternal folate-deficient and paternal fol

Parental reports of stress and anxiety in their migrant children in China: The mediating role of parental psychological aggression and corporal punishment.

BACKGROUND: Both ecological system theory and developmental contextualism framework have pointed out that family is one of the most direct microsystem environmental factors affecting children adjustment, especially for migrant children. As two major family microsystem factors, parenting stress and harsh discipline may be closely related to migrant children's anxiety. OBJECTIVE: This study examined the predictive effects of both mothers' and fathers' parenting stress on Chinese migrant children's anxiety one year later in the single model and the mediating role of mothers' and fathers' harsh d

Reconsideration of the Role of Regulatory T Cells during Pregnancy: Differential Characteristics of Regulatory T Cells between the Maternal-Fetal Interface and Peripheral Sites and between Early and Late Pregnancy.

Regulatory T (Treg) cells play an important role in implantation of the embryo and maintenance of pregnancy after allogeneic mating. Implantation failure, miscarriage, and preeclampsia are associated with decreased numbers of Treg cells or with dysfunctional Treg cells. Treg cells are classified into naturally occurring Treg (nTreg) cells or thymus-derived Treg (tTreg) cells that differentiate in the thymus and induce tolerance to self-antigens, while induced Treg (iTreg) or peripheral Treg (pTreg) cells differentiate in the periphery and induce transient tolerance to foreign antigens. Interventions to change parental parenting behaviour to reduce unintentional childhood injury: a randomised controlled trial.

OBJECTIVE: The previous study has shown the impact of paternal involvement in childcare on unintentional childhood injury; yet the causality is unknown. The purpose of this study is to investigate whether the intervention of an educational video on paternal involvement in childcare can prevent unintentional injury among young children. METHODS: A randomised controlled trial of parents of children born at two obstetrics wards in Japan (n=451, intervention group: 223, control group: 228) was conducted. Parents in the intervention group watched an educational video that promote paternal involvem

Effect of parental age on treatment response in adolescents with schizophrenia.

BACKGROUND: Advanced paternal age (APA) is associated with increased risk for schizophrenia, but its effect on treatment response has not been longitudinally studied. METHODS: Association of parental ages at the time of the child's birth with age of onset, initial symptom severity and treatment response (to placebo and three different weight-based doses of paliperidone ER) in adolescents with schizophrenia was assessed in a post-hoc analysis using data from a 6-week double-blind study, the primary results of which are published (NCT00518323). RESULTS: The mean (SD) paternal age was 29.2 (6.2

The disparate maternal aunt-uncle ratio in male transsexuals: an explanation invoking genomic imprinting.

A significant skewing in the sex ratio in favour of females has been reported for the families of homosexual men such that there are fewer maternal uncles than aunts. This finding is repeated for a large series of transsexual families in this study. Four hundred and seventeen male-to-female transsexuals and 96 female-to-male transsexuals were assessed. Male-to-female transsexuals have a significant excess of maternal aunts vs. uncles. No differences from the expected parity were found for female-to-male transsexuals or on the paternal side. A posited explanation for these findings invokes X in

The transition to fatherhood beyond early infancy: Hormonal, behavioral and psychological changes.

Previous cross-sectional studies have documented hormonal differences between human fathers and nonfathers. However, the timing and persistence of these changes remain unclear. We previously reported that first-time expecting fathers exhibited lower plasma testosterone (T) and arginine vasopressin (AVP), as well as dynamic changes in oxytocin (OT) across portions of the prenatal and early postpartum period compared with partnered nonfather controls. By four months postpartum, however, these group differences were no longer significant. Here, we extend these findings longitudinally into late in
