Effects of G-protein activator fluorides, protein kinase C activator phorbol ester and protein kinase inhibitor on insulin binding and hormonal imprinting of Tetrahymena.

AlCl3, BeCl2 and NaF do not influence the insulin binding of Tetrahymena immediately after treatment, but 24 h later insulin binding is decreased or increased by NaF in a dose-dependent manner, AlCl3 barely influences the binding, and BeCl2 increases it. The effect of all the three fluorides is dose-dependent. While NaF and AlF4 decrease binding at low doses and increase the binding at higher doses, BeF3 increases the insulin binding enormously. NaF does not permit insulin imprinting to be developed, AlF4 inhibits or amplifies the imprinting in a dose-dependent manner, while BeF3 allows imprin

Identification of novel putative-binding proteins for cellular prion protein and a specific interaction with the STIP1 homology and U-Box-containing protein 1.

Prion diseases involve the conversion of the endogenous cellular prion protein, PrP(C), into a misfolded infectious isoform, PrP(Sc). Several functions have been attributed to PrP(C), and its role has also been investigated in the olfactory system. PrP(C) is expressed in both the olfactory bulb (OB) and olfactory epithelium (OE) and the nasal cavity is an important route of transmission of diseases caused by prions. Moreover, Prnp(-/-) mice showed impaired behavior in olfactory tests. Given the high PrP(C) expression in OE and its putative role in olfaction, we screened a mouse OE cDNA library

Carbohydrate responsive element binding protein promotes colorectal carcinogenesis via Wnt/β-catenin pathway.

Despite significant advances in colorectal cancer (CRC) diagnosis and treatment, drug therapy of CRC patients is still confronted with considerable challenges. Carbohydrate response element-binding protein (ChREBP), a glucose-responsive transcription factor regulating glycolysis and de novo lipogenesis, shows elevated expression in human CRC tissues and correlates with poor disease-free survival and overall survival. However, the in vivo role and mechanism of ChREBP in colorectal carcinogenesis remain unclear. We used ChREBP knockout mice, which were intraperitoneally injected with azoxymethan

RLBindDeep: A ResNet-LSTM based novel framework for protein-ligand binding affinity prediction.

The prediction of the binding affinity of proteins and ligands in computational drug discovery with high accuracy is critical when evaluating the effectiveness of potential therapeutic compounds. This research work introduces RLBindDeep, a novel deep learning architecture based on the amalgamation of the ResNet and LSTM architectures, for improved accuracy in predicting protein-ligand binding affinities. Most traditional methodologies utilizing conventional molecular docking techniques suffer from poor accuracy owing to semi-flexible modeling approaches and limited considerations of complex in

Cardiac myosin-binding protein C in community-based patients with suspected heart failure.

INTRODUCTION: Myocardial injury is a hallmark of heart failure (HF). It is unknown whether established (e.g. troponin) or novel biomarkers of myocardial injury [e.g. cardiac myosin-binding protein C (cMyBP-C)] provide additive diagnostic value beyond natriuretic peptides in community patients with suspected HF. METHODS: Community-based patients with suspected HF and elevated NT-proBNP levels were recruited into a multicentre, prospective, observational study at five sites (NCT04724200). Venous blood sampling was performed at the time of echocardiography. HF was classified according to left ve

Insulin-like growth factor-binding protein 7 is a soluble cell adhesion molecule.

Insulin-like growth factor-binding protein 7 (IGFBP7) is a secreted protein with diverse roles in angiogenesis, cell differentiation, tissue remodeling, and regulating cell signaling and is linked to numerous human diseases. The molecular basis of the multifunctionality of IGFBP7 remains unclear. Using cryo-electron microscopy, we show that IGFBP7 assembles into a barrel-shaped dodecamer in the presence of heparin. The carboxyl-terminal IgC2 domain forms the central core of the barrel, which is capped by the amino-terminal heparin-binding IB domain at both ends. This homo-oligomer can simultan

RNA-Binding Protein RBMS3 Inhibits Cervical Cancer Progression by Enhancing HSPA6 mRNA Stability.

BACKGROUND: Cervical cancer (CC) is a prevalent malignancy in women. RNA-binding motif single-stranded interacting protein 3 (RBMS3) acts as a tumor suppressor in many cancer types, but its role and underlying regulatory mechanisms in CC remain unclear. METHODS: The expression levels of RBMS3 and heat shock protein family A member 6 (HSPA6) were evaluated in clinical CC tissues. Gain- and loss-of-function assays, transcriptome sequencing, and rescue experiments were performed in CC cell lines and nude mouse xenograft models. RESULTS: RBMS3 expression was notably lower in CC tissues (n = 306)

A Campomelic Dysplasia A76E Mutation in Sox9 Destabilizes Protein and DNA Binding Dynamics.

Sox9, a pivotal transcription factor belonging to the Sox family, orchestrates critical processes throughout embryonic development, maintenance and differentiation, and exerts a profound influence on organogenesis. Its regulatory versatility stems from precise binding to defined DNA regions, often in collaboration with tissue-specific partners. The dysregulation of Sox9 during chondrogenesis leads to a skeletal malformation termed campomelic dysplasia and has emerged as a significant factor in various other human diseases, including cancer. A point mutation at position 76 (alanine to glutamic

RA-PLA: Retrieval-augmented graph convolutional networks for protein-ligand binding affinity prediction.

Protein-ligand binding affinity (PLA) prediction is essential for computational drug discovery, yet existing deep learning methods typically employ a single globally trained model that fails to adapt to individual query samples. In this paper, we propose a novel end-to-end framework that retrieves hard protein-ligand pairs-samples whose nearest neighbors have conflicting labels under manifold smoothness constraints-and integrates them via a semi-supervised graph convolutional network (GCN). For each query, we automatically construct a graph where nodes represent protein-ligand pairs and edges

RNA-binding protein hnRNPK: a multifunctional regulator of skeletal muscle biology and disease.

Heterogeneous nuclear ribonucleoprotein K (hnRNPK) is a highly conserved, multifunctional DNA/RNA-binding protein that regulates gene expression at both transcriptional and post-transcriptional levels. In skeletal muscle, hnRNPK is essential for development, regeneration, and homeostasis, influencing satellite cell activation, myoblast proliferation and differentiation, and myofiber maturation. Its dysregulation is linked to muscle atrophy, degenerative diseases, and impaired regeneration. This review summarizes current knowledge of hnRNPK's molecular structure, subcellular localization and dy

GTP-binding protein DIRAS2 inhibits ferroptosis in oral leukoplakia via regulating iron metabolism.

BACKGROUND: Oral leukoplakia (OLK) is the most prevalent and extensively studied oral potential malignant disorder (OPMD), with a high risk of progression to malignancy. Ferroptosis has been implicated as a key process in the progression of OLK, and iron metabolism significantly influences the ferroptosis process. DIRAS2, a member of the GTPase family within RAS proteins, is associated with ferroptosis, however, how DIRAS2 affects iron metabolism remains unknown. METHODS: Immunohistochemical staining was used to assess the expression of SLC7A11 and LCN2 in human normal oral mucosa and OLK tis

Retinol-binding protein 3 in ophthalmology: current evidence, research progress, and future perspectives.

Retinol-binding protein 3 (RBP3) is a photoreceptor-derived glyco-lipoprotein that plays a central role in retinoid trafficking between photoreceptors and the retinal pigment epithelium. Beyond its canonical role in the visual cycle, emerging evidence suggests that RBP3 may participate in retinal metabolic regulation, neurovascular protection, inflammatory modulation, and disease-related immune responses. This review critically summarizes current evidence regarding RBP3 in diabetic retinopathy, retinal degeneration, pathological myopia, primary congenital glaucoma, and uveitis. Among these con

Deficiency of the RNA-binding protein RBMS1 improves myocardial fibrosis and heart failure.

BACKGROUND AND AIMS: Previous studies have highlighted the significance of RNA-binding proteins and alternative splicing (AS) in the progression of complex diseases, but the specific involvement of AS in heart failure (HF) remains unclear. This study aimed to elucidate the role of RNA-binding motif single-stranded interacting protein 1 (RBMS1), an RNA-binding protein, in the development of HF by regulating AS and its effect on cardiac fibrosis. METHODS: The level of RBMS1 was investigated in the hearts of both HF patients and mice. Fibroblast-specific knockout RBMS1 mice were generated to inv

Predicted high affinity binding of prion PRPC protein to Human Leukocyte Antigen (HLA).

Misfolding of the cellular prion protein (PRPC) is associated with fatal neurodegenerative prion diseases for which no treatments are currently available. Although the immune system is generally non-responsive (tolerant) to self-proteins such as PRPC, evidence of anti-prion antibodies suggests escape from self-tolerance in some individuals and supports the potential for the human immune system to be leveraged against prion disease. Human leukocyte antigen (HLA) plays a central role in rejecting endogenous non-self proteins (e.g. cancer neoantigens) by activating CD8+ cytolytic T cells via the

A small molecule inhibitor of RNA-binding protein IGF2BP3 shows anti-leukemic activity.

The RNA-binding protein IGF2BP3 is an oncofetal protein over-expressed in B-cell acute lymphoblastic leukemia and is critical for leukemogenesis in experimental models. With cancer-specific expression, functional dispensability for normal development, and an unleveraged pro-oncogenic function in mRNA homeostasis, IGF2BP3 represents an excellent target. With no small molecule inhibitors of IGF2BP3 in clinical use, we aimed to identify new IGF2BP3 inhibitors using biochemical methods. A biochemical screen, followed by a cell-based counter screen led to the identification of compounds with protei

Aggregates associated with amyotrophic lateral sclerosis sequester the actin-binding protein profilin 2.

Amyotrophic Lateral Sclerosis (ALS) is a devastating neurodegenerative disease characterized by the degeneration of upper and lower motoneurons. The four most frequently mutated genes causing familial ALS (fALS) are C9orf72, FUS, SOD1, and TARDBP. Some of the related wild-type proteins comprise intrinsically disordered regions (IDRs) which favor their assembly in liquid droplets-the biophysical mechanism behind the formation of physiological granules such as stress granules (SGs). SGs assemble and dissolve dependent on the cellular condition. However, it has been suggested that transition from

Structural and Biophysical Basis for PFAS Binding by Human Sterol Carrier Protein-2.

Per- and polyfluoroalkyl substances (PFAS) are harmful environmental contaminants that bioaccumulate in human tissues and are linked to adverse health outcomes. While PFAS are known to bind to a variety of lipid binding proteins (LBPs), such as human serum albumin and fatty acid-binding proteins (FABPs), the broader molecular basis for their biological distribution and breadth of protein binding in humans remains unanswered. We hypothesize that some distribution and persistence of PFAS in humans arises from a distributed network of lipid transfer proteins that collectively solubilize and trans…

The RNA-Binding Protein Tristetraprolin Contributes to CFTR mRNA Stability in Cystic Fibrosis.

Cystic fibrosis (CF) is the most common inherited disorder and is characterized by an inflammatory phenotype. We found that in bronchial epithelium reconstituted form lung tissue biopsies from patients with CF, the RNA-binding protein tristetraprolin (TTP), a key regulator of inflammation, is dysregulated in cells that strongly express cytokines and ILs. TTP activity is regulated by extensive posttranslational modifications, particularly phosphorylation. We found that, in addition to mRNA downregulation, phosphorylated TTP (which cannot bind to mRNA) accumulated in CF cultures, suggesting that

Accurate de novo design of high-affinity protein-binding macrocycles using deep learning.

Developing macrocyclic binders to therapeutic proteins typically relies on large-scale screening methods that are resource intensive and provide little control over binding mode. Despite progress in protein design, there are currently no robust approaches for de novo design of protein-binding macrocycles. Here we introduce RFpeptides, a denoising diffusion-based pipeline for designing macrocyclic binders against protein targets of interest. We tested 20 or fewer designed macrocycles against each of four diverse proteins and obtained binders with medium to high affinity against all targets. For
