Glucagon-Like Peptide-1 Receptor Agonist Use in Obese Patients Is Associated With Decreased Risk of Revision and Retears After Arthroscopic Rotator Cuff Repair.

PURPOSE: To evaluate the impact of preoperative glucagon-like peptide-1 receptor agonist (GLP-1 RA) use on medical complications, emergency department use, and orthopaedic complications in obese patients undergoing arthroscopic rotator cuff repair. METHODS: Obese patients (defined by relevant ICD-10 codes and confirmed by body mass index measurements) undergoing primary arthroscopic rotator cuff repair before June 2023 with at least 2-year follow-up were found in the TriNetX database and stratified according to GLP-1 RA use before surgery. Patients prescribed GLP-1 RAs were propensity-matched

GLP-1 Receptor Agonists as Pharmacologic Treatment for Obesity After Spinal Cord Injury: Clinical Considerations.

Individuals with spinal cord injury (SCI) face a disproportionate burden of obesity and cardiometabolic disease, however, evidence to guide pharmacologic treatment is minimal. Mainstay diet and exercise recommendations often yield modest, unsustainable results for those with SCI. In the general population, glucagon-like peptide-1 receptor agonists and dual agonists (GLP-1s) have emerged as potent therapies for obesity and related cardiometabolic disorders. Initially developed for type 2 diabetes mellitus, GLP-1s improve glycemic control, promote substantial weight loss, and reduce other cardio

Adherence and Persistence to Injectable versus Oral GLP-1 Receptor Agonists in Patients with Type 2 Diabetes Mellitus: A Comparative Systematic Review and Analysis.

BACKGROUND: Type 2 diabetes mellitus (T2DM) requires sustained glycemic control to prevent long-term complications. Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) are effective therapies, traditionally administered via injection. The introduction of oral formulations, such as oral semaglutide, may improve patient adherence and persistence. METHODS: A systematic review with descriptive pooled synthesis was conducted using studies identified from PubMed, ClinicalTrials.gov, and relevant real-world observational data sources. Adherence and persistence outcomes were summarized and compared…

Protecting Musculoskeletal Development and Physical Function in Adolescents on GLP-1 Therapy.

Glucagon-like peptide-1 receptor agonists (GLP-1s), including semaglutide and liraglutide, are effective pharmacologic options for pediatric obesity, producing significant weight loss, cardiometabolic improvements, and reductions in barriers to physical activity. By alleviating pain, mechanical load, and psychosocial burden, these agents can also create a window to initiate sustainable health behaviors in adolescents. However, use during critical developmental periods raises concerns regarding long-term effects on musculoskeletal maturation. Adolescence is marked by rapid accrual of muscle and…

Evolution of incretin-based therapies: From GLP-1 monotherapy to dual and triple agonists: A new era in metabolic therapy.

Incretin-based therapies have revolutionised the management of metabolic disorders, transitioning from DPP-4 inhibitors to advanced GLP-1 re … d evidence. While DPP-4 inhibitors offer modest glycaemic benefits, GLP1RAs such as liraglutide and semaglutide have demonstrated significant weight loss and cardiometabolic protection. Dual GIP/GLP-1 agonist tirzepatide and triple agonist retatrutide have shown unprecedented efficacy, with up to 24% body weight reduction and improvement in hepatic and inflam …

Trends in utilization of glucagon-like peptide‑1 receptor agonists, sodium-glucose cotransporter‑2 inhibitors, and metabolic bariatric surgery in U.S. adults with diabetes and obesity.

Median treatment duration was 240 days for GLP‑1RAs and 254 days for SGLT2is; ∼50% were adherent. Tirzepatide had the highest adherence and persiste …

The use of pharmacotherapies in non-cirrhotic metabolic dysfunction-associated steatohepatitis: a UK expert consensus.

Metabolic dysfunction-associated steatohepatitis (MASH), a potentially progressive form of metabolic dysfunction-associated steatotic liver disease (MASLD), increases risk of fibrosis progression, cirrhosis, and liver-related and cardiometabolic morbidity. The first licensed pharmacotherapies, resmetirom and semaglutide, mark a shift in management but practical guidance for real-world implementation is lacking. The British Association for the Study of the Liver and British Society of Gastroenterology MASLD special interest group developed consensus recommendations on patient selection, lifesty…

Glucagon-like peptide-1 (GLP-1) receptor agonists and the dual glucose-dependent insulinotropic polypeptide (GIP)/GLP-1 receptor agonist tirzepatide are increasingly used to treat obesity and type 2 diabetes in men of reproductive age, but practical guidance for counseling men planning fatherhood remains limited. Alopecia as an Emerging Adverse Effect Associated With Glucagon-Like Peptide-1 (GLP-1) Receptor Agonists for Weight Loss: A Scoping Review.

Alopecia has recently been reported as a potential emerging adverse effect associated with glucagon-like peptide-1 receptor agonists (GLP-1RAs), which are widely prescribed for weight loss. While gastrointestinal symptoms remain the most frequently documented adverse effects, an increasing number of reports describe hair loss events in patients treated with semaglutide, liraglutide, tirzepatide, and dulaglutide. This scoping review aimed to synthesize the available evidence regarding this potential association. A systematic literature search was conducted across PubMed, Scopus, Google Scholar,…

Roflumilast ameliorates GAN diet-induced non-alcoholic fatty liver disease by reducing hepatic steatosis and fibrosis in ob/ob mice.

Non-alcoholic fatty liver disease (NAFLD) is a highly prevalent progressive liver disease. Currently, there is only one drug for NAFLD treatment, and the options are limited. Phosphodiesterase-4 (PDE-4) inhibitors have potential in treating NAFLD. Therefore, this study aims to investigate the effect of roflumilast on NAFLD. Here, we fed ob/ob mice to induce the NAFLD model by GAN diet. Roflumilast (1 mg/kg) was administered orally once daily. Semaglutide (20 nmol/kg), used as a positive control, was injected subcutaneously once daily. Our findings showed that roflumilast has beneficial effects…
