Tumor-Infiltrating Mast Cells Enhance Neoadjuvant Therapy Efficacy in ESCC via Modulation of the Desmoplastic Microenvironment.

OBJECTIVE: Neoadjuvant therapy is essential for locally advanced esophageal squamous cell carcinoma (ESCC), but its efficacy requires improvement. This study investigated the role of tumor-infiltrating mast cells (MCs) in treatment response and prognosis. METHODS: An integrated approach was employed, combining single-cell RNA sequencing of paired specimens from one ESCC patient pre- and post-neoadjuvant therapy, immunofluorescence analysis of a retrospective cohort of 68 treatment-naïve ESCC surgical specimens, bioinformatics analysis of public datasets, and in vitro mast cell activation assa

Spatial profiling identifies a distinct and topographically-defined tumor microenvironment that emerges during multiple myeloma evolution.

The pathobiology of multiple myeloma is influenced by cells of the bone marrow, but whether tumor support is organized in a spatially-defined tumor microenvironment (TME) remains unclear. Using spatial transcriptomics and imaging mass cytometry, we show that myeloma cells initially exist as scattered cells throughout the marrow, yet with disease evolution condense into a spatially-defined TME. Dense tumor nodules contain a cellular ecosystem distinct from the surrounding marrow, enriched for macrophages, conventional dendritic cells, and CD8⁺ T cells, as well as endothelial cells and THY1⁺ mes…

Biomaterial-based mitochondria-targeted nanomedicine for remodeling the breast cancer tumor microenvironment and overcoming therapy resistance.

Breast cancer treatment remains limited by therapeutic resistance, recurrence, and incomplete responses to immunotherapy, largely because tumor cells continuously adapt to the dynamic tumor microenvironment (TME). Mitochondria are central regulators of energy metabolism, redox balance, cell death, inflammatory signaling, and immune modulation. Stressors within the breast cancer TME, including hypoxia, nutrient deprivation, acidosis, matrix stiffening, oxidative stress, and treatment pressure, can remodel mitochondrial metabolism and dynamics, thereby promoting tumor survival, invasion, immune …

Small-molecule photosensitizer for enhanced antiangiogenesis and immunosuppressive tumor microenvironment reprogramming in anti-metastatic photodynamic therapy.

Tumor angiogenesis, a well-recognized barrier to effective antitumor treatment, drives tumor growth, recurrence, and metastasis by facilitating tumor cell intravasation, impairing immune cell infiltration, and fostering an immunosuppressive tumor microenvironment. Herein, a small-molecule drug T-Pc was rationally designed and synthesized by covalent conjugation of the alkylating agent temozolomide (TMZ) and the photosensitizer zinc phthalocyanine (ZnPc). The dual antitumor and anti-angiogenic actions of low-dose TMZ and ZnPc augment the in vitro performance of T-Pc, conferring the conjugate wi…

Reprogramming the immune suppressive tumor microenvironment in glioma enhances the efficacy of immune-mediated gene therapy.

Gliomas account for ∼80% of primary malignant brain tumors. Many CNS WHO grade 2-3 and some grade 4 gliomas harbor mutant isocitrate dehydrogenase 1 (mIDH1), which causes a gain-of-function mutation (IDH1 R132H) leading to the production of 2-hydroxyglutarate (2HG). Mutant IDH1-induced 2HG, through epigenetic reprogramming elicits an immune-permissive tumor microenvironment (TME). An immunosuppressive mechanism in the glioma TME involves adenosine production via the ectoenzyme CD73. This study investigates mIDH1's influence on CD73 expression and adenosine levels. We demonstrate that mIDH1 gli…

Research progress of SPP1 in cancer regulation: Tumorigenesis, metastasis, tumor microenvironment, immunity and therapeutic targets (Review).

OBJECTIVES: Psoriatic arthritis (PsA) has been defined as a systemic, chronic, inflammatory arthritis, usually seronegative for rheumatoid factor (RF), associated with cutaneous psoriasis. The exact prevalence of PsA is unknown and its estimation has been difficult, partly due to the lack of a widely accepted classification criteria. Agreed and validated criteria will facilitate comparison between centres and different countries in the areas of epidemiology, outcome studies and therapeutic trials. A number of classification criteria have been published by Moll & Wright (M & W), Bennett's, Vase

Cross-Strand Chimeric RNA Signature Predicts Prognosis and Identifies Tumor Immune Microenvironment Associations in Gastric Cancer.

Cross-strand chimeric RNAs (cscRNAs) represent an emerging class of noncanonical fusion transcripts arising from bidirectional transcription, yet their landscape and functional significance in gastric cancer remain unexplored. Here, we systematically investigated cscRNA expression patterns, prognostic relevance, and tumor immune microenvironment associations in gastric cancer through integrated computational and experimental approaches. Analysis of 372 TCGA-STAD samples revealed tumor-specific cscRNA enrichment that was independent of clinicopathological variables including tumor stage, histol

The CAGE Framework (Calcium-Gated Excitability): From Single Neurotransmitters to Calcium States in the Tumor Microenvironment.

The tumor microenvironment (TME) is an innervated and neurochemically active setting. To date, its cholinergic and GABAergic signaling have largely been analyzed as separate systems, contributing to inconsistent findings across the literature. We propose a Calcium-Gated Excitability (CAGE) model, in which α7 nicotinic acetylcholine receptor (nAChR), α4β2 nAChR, GABA type A (GABAA) and type B (GABAB) receptors, and α2δ-1 converge on a single downstream variable: Ca2+ flux. α7 conducts Ca2+ directly; α4β2 may redirect cholinergic input toward GABA release; GABAA and GABAB set the direction of th…

Longitudinal spatial multi-omics delineates tumor microenvironment remodeling across sequential EGFR-TKIs in EGFR-mutant NSCLC.

BACKGROUND: Resistance to therapy is a frequent occurrence in patients with epidermal growth factor receptor (EGFR)-mutant non-small-cell lung cancer (NSCLC) who are treated with EGFR-tyrosine kinase inhibitors (EGFR-TKIs). However, spatial information on how the tumor microenvironment (TME) changes within the same patient from baseline to resistance to first- and then third-generation EGFR-TKIs is scarce. Here, we used rare consecutive re-biopsy samples to build an exploratory, longitudinal, and spatially resolved atlas, aiming to describe within-patient TME remodeling patterns as a resource …

Nutritional biomarkers regulating tumor immune microenvironment in osteosarcoma and as predictors of immune checkpoint inhibitor responses.

Osteosarcoma(OS) exhibits poor and heterogeneous responses to immune checkpoint inhibitors(ICIs), largely due to its highly tumor immunosuppressive microenvironment(TIME). This review explores how nutritional biomarkers regulate the OS tumor microenvironment(TME) and serve as predictors of ICIs efficacy. Key biomarkers, including prognostic nutritional index(PNI), vitamin D and IDO modulate tumor-infiltrating lymphocytes (TILs), CD8+ T-cell function, macrophage polarization, and cytokine-metabolite crosstalk through mechanisms such as nutrient depletion, lactate acidification, and inflammatory…

CD300e is a driver of the immunosuppressive tumor microenvironment and colorectal cancer progression via macrophage reprogramming.

BACKGROUND: Colorectal cancer (CRC) progression is shaped by the tumor microenvironment, particularly tumor-associated macrophages (TAMs), which often adopt immunosuppressive functions. CD300e, a myeloid receptor involved in immune regulation, has an uncharacterized role in CRC. METHODS: Functional studies were conducted in azoxymethane/dextran sodium sulfate and MC38 murine CRC models using CD300e systemic and myeloid-specific CD300e knockout mice, and adoptive transfer experiments assessed macrophage-intrinsic effects. Human studies included analysis of CD300e expression in matched tumor an…

Gastric Signet Ring Cell Carcinoma: Tumor Microenvironment Reprogramming and Novel Therapeutic Targets With Emphasis on GRIN2D.

Gastric Signet Ring Cell Carcinoma (GSRCC) is an increasingly recognized subtype of gastric cancer, particularly prevalent in South Asian populations and regions within India. This carcinoma is distinguished by its abundant cytoplasmic mucinous cells and aggressive clinical behavior, often affecting younger individuals and leading to a poor prognosis due to its advanced-stage presentation and resistance to standard treatments. A critical factor in its progression is the tumor's uniquely immunosuppressive and stromal-rich microenvironment, characterized by dysfunctional immune infiltrates, acti

Patient-Derived 3D-Bioprinted Models of Pancreatic Cancer: Toward Personalized Therapy and Overcoming Tumor Microenvironment Challenges.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive and treatment-resistant solid tumors, mainly due to its complex tumor microenvironment (TME). Characterized by dense desmoplastic stroma, immune suppression, and metabolic rewiring, the TME impairs drug delivery and eventually leads to therapeutic failure. Conventional models, such as two-dimensional (2D) cultures and mouse models, fail to recapitulate the cellular and mechanical intricacies of PDAC, limiting their translational relevance. Three-dimensional (3D) models have emerged as promising tools to better simulate tumor…

Neoadjuvant Immunotherapy Promotes the Formation of Mature Tertiary Lymphoid Structures in a Remodeled Pancreatic Tumor Microenvironment.

Pancreatic ductal adenocarcinoma (PDAC) is a rapidly progressing cancer that responds poorly to immunotherapies. Intratumoral tertiary lymphoid structures (TLS) have been associated with rare long-term PDAC survivors, but the role of TLS in PDAC and their spatial relationships within the context of the broader tumor microenvironment remain unknown. In this study, we report the generation of a spatial multiomic atlas of PDAC tumors and tumor-adjacent lymph nodes from patients treated with combination neoadjuvant immunotherapies. Using machine learning-enabled hematoxylin and eosin image classif…

Deciphering the secrets of tumor-initiating cells in pancreatic ductal adenocarcinoma microenvironment: mechanisms and therapeutic opportunities.

Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive malignancy characterized by poor prognosis, strong resistance to therapy, and a dense immunosuppressive tumor microenvironment (TME). A small subset of cells known as cancer stem cells (CSCs), or tumor-initiating cells (TICs), are increasingly recognized as key contributors to tumor initiation, metastasis, immune evasion, and treatment failure. These cells are defined by their self-renewal capacity, plasticity, and resistance to chemotherapeutic and targeted therapies. Pancreatic cancer stem cells (PaCSCs) are maintained by specifi…
