Basal Forebrain Volume is Associated With Cortical Amyloid Burden in Cognitively Unimpaired Older Adults at Varying Genetic Risk for Alzheimer's Disease.

BACKGROUND: In mild cognitive impairment and dementia due to Alzheimer's disease (AD), postmortem and in vivo neuroimaging studies have demonstrated significant neuronal loss in the basal forebrain cholinergic system (BFCS), which provides the primary cholinergic input to the cerebral cortex. Within the BFCS, atrophy is most prominent in the nucleus basalis of Meynert (nbM), a posteriorly located cluster of magnocellular neurons. However, less is known surrounding the relationship between amyloid deposition, BFCS atrophy, and medial temporal lobe (MTL) volume loss in preclinical AD. This study

Therapeutic potential of Cola nitida against dexamethasone- and scopolamine-induced cognitive and cardiac dysfunction.

BACKGROUND: Cognitive dysfunction and cardiometabolic disturbances frequently coexist in neurological and metabolic disorders. Chronic systemic inflammation in cardiometabolic syndrome may promote neuroinflammation, damaging neurons and synapses and impairing memory pathways. Cola nitida is a medicinal plant widely recognized for its stimulant and therapeutic properties, yet its neuroprotective and cardiometabolic effects remain underexplored. Objective: This study investigated the effects of methanol extract of Cola nitida (CN) in a mouse model of memory and metabolic dysfunction induced by s

Modeling Gulf War Illness: anxiety, memory deficits, cholinergic and potassium channel imbalance in male and female mice.

Gulf War Illness (GWI) is a complex, multisystem condition that remains difficult to diagnose, with an etiology thought to involve interactions between stress and exposure to acetylcholinesterase (AChE) inhibitors of varying potency. Although rodent models using GWI-related neurotoxicants have been developed, many do not fully replicate war-theater conditions, particularly the inclusion of sarin exposure. We modeled a GW-like exposure in male and female C57BL/6J mice using pyridostigmine bromide (PB), permethrin (PER), N,N-diethyl-meta-toluamide (DEET), moderate stress, and Diisopropylfluoroph

Multimodal beneficial effects of BNN27, a nerve growth factor synthetic mimetic, in the 5xFAD mouse model of Alzheimer's disease.

Alzheimer's Disease (AD) is an incurable and debilitating progressive, neurodegenerative disorder which is the leading cause of dementia worldwide. Neuropathologically, AD is characterized by the accumulation of Aβ amyloid plaques in the microenvironment of brain cells and neurovascular walls, chronic neuroinflammation, resulting in neuronal and synaptic loss, myelin and axonal failure, as well as significant reduction in adult hippocampal neurogenesis. The hippocampal formation is particularly vulnerable to this degenerative process, due to early dysfunction of the cholinergic circuit. Neurot

Functional atropine sensitive purinergic responses in the healthy rat bladder.

While acetylcholine is regarded to be the main directly contractile transmitter substance in the urinary bladder, interactions with other transmitters likely occur. Presently, the interplay between purinergic and cholinergic signalling was investigated to unravel the involvement of the urothelium and efferent neurons in the functionally important purinergically evoked release of acetylcholine in vitro. Functional characterization of receptor subtypes involved in this interplay was also performed. In vitro organ bath experiments with electrical field stimulation (EFS) or administration of agoni

Taenia larvae possess distinct acetylcholinesterase profiles with implications for host cholinergic signalling.

Larvae of the cestodes Taenia solium and Taenia crassiceps infect the central nervous system of humans. Taenia solium larvae in the brain cause neurocysticercosis, the leading cause of adult-acquired epilepsy worldwide. Relatively little is understood about how cestode-derived products modulate host neural and immune signalling. Acetylcholinesterases, a class of enzyme that breaks down acetylcholine, are produced by a host of parasitic worms to aid their survival in the host. Acetylcholine is an important signalling molecule in both the human nervous and immune systems, with powerful modulator

Inhibition of 12/15 LOX ameliorates cognitive and cholinergic dysfunction in mouse model of hypobaric hypoxia via. attenuation of oxidative/nitrosative stress.

12/15 Lipoxygenase has recently been described as potent propagator of oxidative stress and is closely associated with cognitive decline in neurodegenerative diseases. The mechanism/s behind 12/15 LOX involvement in cognitive deficits remain obscure. The current study has been designed to investigate the underlying role of 12/15LOX and effect of 12/15 LOX inhibition on hypobaric hypoxia-induced memory impairment and cholinergic deficits. Male Balb/c mice subjected to simulated hypobaric hypoxia/reoxygenation condition for 3days showed marked working memory impairment concomitant with hippocamp

Neocortical and hippocampal glucose hypometabolism following neurotoxic lesions of the entorhinal and perirhinal cortices in the non-human primate as shown by PET. Implications for Alzheimer's disease.

Temporoparietal glucose hypometabolism, neuronal loss in the basal forebrain cholinergic structures and preferential accumulation of neurofibrillary tangles in the rhinal cortex (i.e. in the entorhinal and perirhinal cortices) are three early characteristics of Alzheimer's disease. Based on studies of the effects of neurotoxic lesions in baboons, we previously concluded that damage to the cholinergic structures plays, at best, a marginal role in the association neocortex hypometabolism of Alzheimer's disease. In the present study, we have assessed the remote metabolic effects of bilateral neur

Neurally released pituitary adenylate cyclase-activating polypeptide enhances guinea pig intrinsic cardiac neurone excitability.

Intracellular recordings were made in vitro from guinea-pig cardiac ganglia to determine whether endogenous neuropeptides such as pituitary adenylate cyclase-activating polypeptide (PACAP) or substance P released during tetanic neural stimulation modulate cardiac neurone excitability and/or contribute to slow excitatory postsynaptic potentials (sEPSPs). When nicotinic and muscarinic receptors were blocked by hexamethonium and atropine, 20 Hz stimulation for 10 s initiated a sEPSP in all innervated neurones. In 40% of the cells, excitability was enhanced after termination of the sEPSP. This sug

Neural Gastric Electrical Stimulation: Future Prospects in Managing Gastric Emptying Disorders.

Neural gastric electrical stimulation (NGES) is a multi-channel high-energy gastrointestinal stimulation technique that directly activates cholinergic motor neurons to evoke contractions that improve gastric emptying. NGES stimulation superimposes on spontaneous electromechanical activity to generate coordinated propagating contractions. In contrast, the most commonly applied form of gastric electrical stimulation (Enterra), employs paired pulses that stimulate vagal afferent pathways to reduce symptoms of nausea and vomiting, but does not enhance emptying. This review examines the evolution a

Calcitonin gene-related peptide- and adrenomedullin-induced facilitation of calcium current in submandibular ganglion.

OBJECTIVE: The control of saliva secretion is mainly under parasympathetic control. The submandibular ganglion (SMG) is a parasympathetic ganglion which receives inputs from preganglionic cholinergic neurons, and innervates the submandibular salivary gland to control saliva secretion. The aim of this study was to investigate if adrenomedullin (ADM) and/or calcitonin gene-related peptide (CGRP) modulate voltage-dependent calcium channel (VDCCs) current (I(Ca)) in SMG. DESIGN: The profile of CGRP and ADM actions in SMG was studied using the whole-cell configuration of the patch-clamp technique.…

Alzheimer's disease: a challenging enigma.

The gray matter of patients dying of senile dementia of the Alzheimer type (SDAT) displays severe histopathologic lesions, including neurofibrillary tangles, neuritic (senile) plaques, granulovacuolar degeneration, Hirano bodies, and loss of nerve cells. The differences between the changes seen in SDAT and those seen in normal aging of the brain are quantitative. Results of neurochemical studies suggest that cholinergic neurons are especially afflicted, but clinical trials administering choline precursors in patients' food hve thus far been unsuccessful. Theories of its cause include genetic p…

Effects of aging on signal transmission and transduction systems in the gerbil brain: morphological and autoradiographic study.

The Mongolian gerbil was used as a model of aging because of its relatively short lifespan, genetic homogeneity and the fact that data had been collected previously. Furthermore, gerbils have been widely used in biomedical investigations of stroke and epilepsy. Age-related differences in signal transmission and transduction systems were investigated in brains of three-, 11- and 21-month-old gerbils by morphological and in vitro receptor autoradiographic studies. Morphometric analysis revealed a decreased number of neurons in layer III of the occipital cortex and also a decrease in cerebellar P

Cholinergic innervation of the primate hippocampal formation. I. Distribution of choline acetyltransferase immunoreactivity in the Macaca fascicularis and Macaca mulatta monkeys.

The cholinergic innervation of the hippocampal formation of Macaca fascicularis (cynomolgus) and Macaca mulatta (rhesus) monkeys was investigated by immunohistochemical procedures using a monoclonal antibody directed against choline acetyltransferase. The distribution of choline acetyltransferase in the monkey demonstrated both similarities and differences with the staining patterns observed in the rat or with acetylcholinesterase in the monkey. While both of these latter preparations demonstrated labeled cells, for example, no choline acetyltransferase labeled neurons were observed in the mon

Cognitive Correlates of Basal Forebrain Atrophy and Associated Cortical Hypometabolism in Mild Cognitive Impairment.

Degeneration of basal forebrain (BF) cholinergic nuclei is associated with cognitive decline, and this effect is believed to be mediated by neuronal dysfunction in the denervated cortical areas. MRI-based measurements of BF atrophy are increasingly being used as in vivo surrogate markers for cholinergic degeneration, but the functional implications of reductions in BF volume are not well understood. We used high-resolution MRI, fluorodeoxyglucose-positron emission tomography (PET), and neuropsychological test data of 132 subjects with mild cognitive impairment (MCI) and 177 cognitively normal

In vivo mapping of cholinergic terminals in normal aging, Alzheimer's disease, and Parkinson's disease.

To map presynaptic cholinergic terminal densities in normal aging (n = 36), Alzheimer's disease (AD) (n = 22), and Parkinson's disease (PD) (n = 15), we performed single-photon emission computed tomography using [123I]iodobenzovesamicol (IBVM), an in vivo marker of the vesicular acetylcholine transporter. We used coregistered positron emission tomography with [18F]fluorodeoxyglucose for metabolic assessment and coregistered magnetic resonance imaging for atrophy assessment. In controls (age, 22-91 years), cortical IBVM binding declined only 3.7% per decade. In AD, cortical binding correlated i
