Clinical and molecular evidence for the role of androgens and WT1 in testis descent.

Testicular maldescent is a common congenital disorder associated with testicular cancer and infertility. In this study, testis position was assessed in subjects with genital abnormalities due to AR mutations, Denys-Drash and WAGR syndromes or an unknown aetiology. Subjects with completely female genitalia and an AR mutation or an unknown aetiology had a greater proportion of maldescended testes (intra-abdominal and inguinal) than those with less severe abnormalities (P=0.00027 and P<0.000001, respectively). Whereas subjects with severe, moderate or mild abnormalities and an unknown aetiology,

Recurrent RTTN mutation leading to severe microcephaly, polymicrogyria and growth restriction.

Autosomal recessive missense Rotatin (RTTN) mutations are responsible for syndromic forms of malformation of cortical development, ranging from isolated polymicrogyria to microcephaly associated with primordial dwarfism and other major malformations. We identified, by trio based whole exome sequencing, a homozygous missense mutation in the RTTN gene (c.2953A > G; p.(Arg985Gly)) in one Moroccan patient from a consanguineous family. The patient showed early onset primary microcephaly, detected in the fetal period, postnatal growth restriction, encephalopathy with hyperkinetic movement disorders

Expression and prognosis analyses of the Tob/BTG antiproliferative (APRO) protein family in human cancers.

BACKGROUND: Despite advances in early diagnosis and treatment, cancer remains the major cause of mortality in the world. The Tob/BTG antiproliferative (APRO) protein family is reported to participate in diverse human diseases. However, there's little known about their expression and prognostic values in most human cancers. METHODS: We performed a detailed cancer vs. normal analysis. The mRNA expression levels of APRO family in various cancers were analyzed via the Oncomine database. Moreover, the Kaplan-Meier Plotter and PrognScan databases were used to evaluate the prognostic values. RESULT

Analysis of gene expression in Ca2+-dependent activator protein for secretion 2 (Cadps2) knockout cerebellum using GeneChip and KEGG pathways.

In the mouse cerebellum, Ca2+-dependent activator protein for secretion 2 (CADPS2, CAPS2) is involved in regulated secretion from dense-core vesicles (DCVs), which contain neuropeptides including brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3). Capds2 knockout (KO) mice show impaired cerebellar development in addition to autistic-like behavioral phenotypes. To understand the molecular impact caused by loss of Capds2, we analyzed gene expression profiles in the Capds2 KO cerebellum using a GeneChip microarray and the KEGG Pathway database. Significant differential expression

An evolutionary and structure-based docking model for glucocerebrosidase-saposin C and glucocerebrosidase-substrate interactions - relevance for Gaucher disease.

Gaucher disease, the most prevalent lysosomal storage disorder, is principally caused by malfunction of the lysosomal enzyme glucocerebrosidase (GBA), a 497-amino acid membrane glycoprotein that catalyzes the hydrolysis of glucosylceramide to ceramide and glucose in the presence of an essential 84-residue activator peptide named saposin C (SapC). Knowledge of the GBA structure, a typical (beta/alpha)(8) TIM barrel, explains the effect of few mutations, directly affecting or located near the catalytic site. To identify new regions crucial for proper GBA functionality, we analyzed the interactio

A database to support the interpretation of human mismatch repair gene variants.

Germline mutations in the mismatch repair (MMR) genes MLH1, MSH2, MSH6, or PMS2 can cause Lynch syndrome. This syndrome, also known as hereditary nonpolyposis colorectal cancer (HNPCC), is an autosomal dominantly-inherited disorder predominantly characterized by colorectal and endometrial cancer. Truncating MMR gene mutations generally offer a clear handle for genetic counseling and allow for presymptomatic testing. In contrast, the clinical implications of most missense mutations and small in-frame deletions detected in patients suspected of having Lynch syndrome are unclear. We have construc

An integrative analysis reveals cancer risk associated with artificial sweeteners.

BACKGROUND: Artificial sweeteners (AS) have been widely utilized in the food, beverage, and pharmaceutical industries for decades. While numerous publications have suggested a potential link between AS and diseases, particularly cancer, controversy still surrounds this issue. This study aims to investigate the association between AS consumption and cancer risk. METHODS: Targets associated with commonly used AS were screened and validated using databases such as CTD, STITCH, Super-PRED, Swiss Target Prediction, SEA, PharmMapper, and GalaxySagittarius. Cancer-related targets were sourced from G

Identification of cathepsin L and urokinase-type plasminogen activator in plasma-derived small extracellular vesicles of breast cancer patients.

Small extracellular vesicles (sEVs) are nanosized vesicles released by both normal and cancer cells, carrying biomolecular cargo that may serve as disease biomarkers, including cancer. We investigated protease signatures of plasma-derived sEVs in chemotherapy-naïve stage I-III breast cancer (BC) patients. sEVs were enriched from plasma of BC patients (n = 49), healthy controls (n = 30), and benign cases (fibroadenoma; n = 15) using a total exosome isolation kit and their enrichment was verified by different approaches. Protease profiling using a Proteome Profiler array identified cathepsin L (

Biosystem Analysis of the Hypoxia Inducible Domain Family Member 2A: Implications in Cancer Biology.

The expression of HIGD2A is dependent on oxygen levels, glucose concentration, and cell cycle progression. This gene encodes for protein HIG2A, found in mitochondria and the nucleus, promoting cell survival in hypoxic conditions. The genomic location of HIGD2A is in chromosome 5q35.2, where several chromosomal abnormalities are related to numerous cancers. The analysis of high definition expression profiles of HIGD2A suggests a role for HIG2A in cancer biology. Accordingly, the research objective was to perform a molecular biosystem analysis of HIGD2A aiming to discover HIG2A implications in c

Proteomic profiling of Rhipicephalus (Boophilus) microplus midgut responses to infection with Babesia bovis.

Differences in protein expression in midgut tissue of uninfected and Babesia bovis-infected southern cattle ticks, Rhipicephalus (Boophilus) microplus, were investigated in an effort to establish a proteome database containing proteins involved in successful pathogen transmission. The electrophoretic separation of midgut membrane proteins was greatly improved by using liquid-phase isoelectric focusing combined with one-dimensional or two-dimensional (2-D) gel electrophoresis. A selection of differentially expressed proteins were subjected to analysis by capillary-HPLC-electrospray tandem mass

AFG2B gene variants and elevated protein expression in lupus nephritis: new insights into childhood-onset systemic lupus erythematosus.

BACKGROUND: Previous genome-wide association studies (GWAS) have associated chronic kidney disease with AFG2B variants. The AFG2B protein is linked to key immune-regulating processes through its interaction with proteasome proteins implicated in systemic inflammation. Dysregulation of these processes is a key factor in the onset of childhood-onset systemic lupus erythematosus (cSLE). This study aimed to investigate the frequency of AFG2B gene variants and the renal expression of the AFG2B protein in cSLE. METHODS: This two-phase, cross-sectional observational study included 45 patients with cS

Mining the secreted and membrane transcriptome of Hyalomma dromedarii ticks for identification of potential protective antigens.

BACKGROUND: Members belonging to the tick genus Hyalomma function as a multi-host reservoir for several pathogens and important parasites infesting large animals, such as camels, goats, cattle and sheep. In Egypt, there is a high risk of pathogen transmission as camels and cattle are imported from Sudan and Ethiopia and shipped to slaughterhouses and animal markets located in populated areas. Hyalomma dromedarii ticks are semi-desert vectors and, similar to other members of the genus Hyalomma, characterized by long-term feeding. During this process, different physiological, biochemical and imm

An Overview of Databases and Bioinformatics Tools for Plant Antimicrobial Peptides.

Antimicrobial Peptides (AMPs) are small, ribosomally synthesized proteins found in nearly all forms of life. In plants, AMPs play a central role in plant defense due to their distinct physicochemical properties. Due to their broad-spectrum antimicrobial activity and rapid killing action, plant AMPs have become important candidates for the development of new drugs to control plant and animal pathogens that are resistant to multiple drugs. Further research is required to explore the potential uses of these natural compounds. Computational strategies have been increasingly used to understand key

A six-gene prognostic signature for aflatoxin B1-associated hepatocellular carcinoma identified through integrated bioinformatics and network toxicology.

BACKGROUND: Aflatoxin B1 (AFB1) is a potent group 1 carcinogen closely associated with hepatocellular carcinoma (HCC), particularly in regions with high dietary exposure and concurrent hepatitis B virus infection. However, the molecular mechanisms by which AFB1 promotes HCC progression remain incompletely understood, and there is a lack of prognostic models specifically tailored to AFB1-associated HCC. Integrating bioinformatics and network toxicology approaches may help identify key genes and construct reliable predictive tools for this unique subtype of liver cancer. This study aims to ident

Identification of Cdc45-interacting partners uncovers two Leishmania donovani proteins involved in DNA replication.

BACKGROUND: Among fatalities due to parasitic diseases, deaths due to Leishmaniases rank second only to those caused by malaria. Endemic to 90 countries, more than a billion people are at risk of Leishmania infection. While there are no vaccines against Leishmaniases, treatments are available. However, the complexities of the treatment regimens, their high costs and toxic side effects, and most importantly the emerging problem of drug resistance, have continued to thrust forward research into this parasite's cellular processes. The parasite is digenetic, reproducing by binary fission in both h
