Naloxone-reversible monocyte dysfunction in patients with chronic fatigue syndrome.

We studied monocyte function in 35 consecutive patients with chronic fatigue syndrome (CFS) and 25 healthy controls. Eighty-five per cent of the patients showed monocyte dysfunction characterized by marked reduction in the number of monocytes displaying immunoreactive cytoskeletal vimentin filaments, a low phagocytosis index, and a reduced expression of HLA-DR antigens. These values increased dramatically after incubation of the patients' monocytes with the opioid antagonist naloxone. Other immunological abnormalities also noted in the patients were low lymphocyte blastogenesis and diminished

Opiate antagonist nalmefene inhibits ethanol-induced flushing in Asians: a preliminary study.

Ethanol-induced flushing (EIF) occurs in up to 80% of Asians and is characterized by facial flushing, tachycardia, and increased cardiac output. Since endogenous opiates and prostaglandins may be mediators of flushing syndromes, we attempted to block EIF in four Asian flushers with single doses of either the opiate antagonist nalmefene, or the prostaglandin synthesis inhibitor indomethacin. Nonflushers (2 Caucasian, 2 Asian) and four Asian flushers were given on separate days water, ethanol (0.4 g/kg p.o.), ethanol plus nalmefene (2 mg i.v.), or ethanol plus indomethacin (50 mg p.o.). Ethanol

The interaction of endogenous opiates with autonomic circulatory control in the dog.

The intravenous injection of either methionine or leucine enkephalin sharply reduces blood pressure, peak left ventricular pressure, and peak LV dP/dt in anesthetized dogs. The magnitude of the hypotensive response increases in proportion to the severity of the preceding surgical stress. The peptides are relatively ineffective after only simple surgical procedures but become highly effective when the autonomic balance is shifted toward sympathetic dependence after more complicated procedures or following bilateral carotid occlusion. The greater the animal's dependence upon sympathetic outflow

Local endogenous opiate activity in dog myocardium: receptor blockade with naloxone.

The participation of endogenous opiates in myocardial performance and coronary blood flow was investigated. Heart rate, left ventricular contractile force (LVCF), left coronary blood flow (LCBF), and left ventricular oxygen extraction were monitored in anesthetized dogs before and after intracoronary opiate receptor blockade with naloxone. LVCF consistently increased in a dose-dependent fashion following intracoronary naloxone. The increasing LVCF was accompanied by significant increases in LCBF and myocardial oxygen consumption, without changes in heart rate. Rapid onset of responses suggeste

Chlorpromazine vs. meperidine in the treatment of phencyclidine psychosis.

Patients with phencyclidine psychosis were treated with two 50 mg injections of either chlorpromazine (N = 10) or meperidine (N = 10). The chlorpromazine-treated group responded more rapidly, but the meperidine-treated group had greater overall improvement. It is suggested that meperidine may have a role in the emergency treatment of phencyclidine psychosis. These findings may provide support for the hypothesis that dopaminergic psychosis is mediated by opioids.

Regulation of body temperature and nociception induced by non-noxious stress in rat.

The effects of 3 different non-noxious stressors on body temperature (Tb) were investigated in the rat: (1) loose restraint in cylinders, (2) removal of the rats from cylinders, exposure to a novel environment and replacement in cylinders, a stressor called here 'novelty', and (3) gentle holding of the rats by the nape of the neck. Loose restraint and 'novelty' produced hyperthermia. On the contrary, holding induced hypothermia. Hypophysectomy (HX) reduced basal Tb, abolished restraint hyperthermia and reduced both 'novelty' hyperthermia and holding hypothermia. Dexamethasone ( DEXA ) had no e

Effects of naloxone on the cardiovascular responses to muscular contraction in decerebrate cats.

The cardiovascular responses to muscular contraction induced by ventral root (L7 and S1) stimulation were studied in unanesthetized decerebrate cats before and after the administration of the opiate antagonist naloxone. Intravenous naloxone (1.0-2.0 mg/kg) did not alter the heart rate or arterial pressure responses to either tetanic or repeated twitch contractions. However, naloxone did increase resting arterial pressure.

Morphiceptin: biphasic inhibition of mu-opiate receptor ligands.

The interaction between morphiceptin and the morphine (mu) opiate receptor present in rat brain membranes has been examined. Detailed competitive displacement curves of morphiceptin against the mu receptor ligands [3H]fentanyl and [3H]naloxone were biphasic, with Hill coefficients of 0.78 and 0.60 respectively. Hoftsee plots of these displacement curves suggested that 30-35% of the morphiceptin binding was to a high affinity site and the residual binding was to a site with lower affinity. These results indicate that morphiceptin binding to the mu opiate receptor does not obey the law of mass a

Effects of [D-Ala2]-methionine-enkephalin on blood pressure, heart rate, and baroreceptor reflex sensitivity in conscious cats.

Effects of intracerebroventricular (i.c.v) injection of [D-Ala2]-methionine-enkephalinamide (DAME) on blood pressure (BP), heart rate, and baroreceptor reflex sensitivity were studied in conscious cats. DAME was administered at doses between 5 and 100 nmoles. Blood pressure and heart rate increased dose dependently. The sensitivity of the baroreceptor reflex was attenuated for 15 to 60 minutes after DAME administration; this was independent of the BP changes. The effects of enkephalin on BP and baroreceptor reflex were abolished by i.c.v. naloxone. DAME caused pathological changes in the elect

Screening for anti-dysmenorrhea components of Wenjing decoction: spectrum-effect relationship analysis, and efficacy equivalence validation.

ETHNOPHARMACOLOGICAL RELEVANCE: Primary dysmenorrhea (PD) is common and has a major impact on women's daily activities and quality of life. Wenjing decoction (WD), a classic Chinese medicine formula, has been widely used for thousands of years in China to treat PD. However, the key pharmacodynamic substances in WD responsible for its anti-dysmenorrhea efficacy are still unclear. AIM OF THE STUDY: This study aimed to screen WD for anti-dysmenorrhea components using the spectrum-effect relationship analysis and validate their efficacy equivalence. METHODS: The chemical composition of WD was id

Met-enkephalin and morphiceptin modulate a GABA-induced inward current in the CNS of Lymnaea stagnalis L.

1. The interaction between GABA and opioid peptides (met-enkephalin and morphiceptin) was studied on the identified, isolated and internally perfused neurons of Lymnaea stagnalis L. (Gastropoda, Basommatophora). 2. GABA (10(-7)-10(-5)M) activated a Cl-dependent inward current with about -20 mV equilibrium potential. Slow and fast GABA-induced inward currents were recorded with different kinetic parameters in distinct identified neurons. 3. Both types of GABA-induced inward currents were reduced or blocked by met-enkephalin (10(-7)-10(-5)M) and morphiceptin (10(-7)-10(-5)M) in a dose-dependent

Evidence of a distinct derangement of opioid tone in hyperinsulinemic patients with polycystic ovarian syndrome: relationship with insulin and luteinizing hormone secretion.

Recent data indicate that an altered opioid tone could be involved in the LH hypersecretion and metabolic alterations seen in polycystic ovary syndrome (PCOS). The aim of the present study was to investigate the presence of a common mechanism of action of opioids on altered insulin and gonadotropin release in patients suffering from PCOS. Twenty-eight women affected by PCOS and 8 normal ovulatory women were studied; an oral glucose tolerance test (OGTT) and GnRH tests were performed during the follicular phase before and after 6 weeks of naltrexone treatment (50 mg/day, orally). Plasma levels

The role of opioids in newborn pig fluid percussion brain injury.

The present study was designed to characterize the relationship between cerebral opioid concentration, cerebral hemodynamics, and cerebral oxygenation following percussion brain injury in neonatal pigs. Previous research found that opioids represent a significant vasoactive component in the regulation of the neonatal piglet cerebral circulation. Anesthetized newborn (1-5 days old) pigs equipped with a closed cranial window were connected to a percussion device consisting of a saline-filled cylindrical reservoir with a metal pendulum. Brain injury of moderate severity (1.9-2.3 atm.) was produce

[Opiate hypothesis in infantile autism? Therapeutic trials with naltrexone].

The opioid hypothesis suggests that childhood autism may result from excessive brain opioid activity during neonatal period which may constitutionally inhibit social motivation, yielding autistic isolation and aloofness (Panksepp, 1979). This hypothesis has now received strong support and is currently based on three types of arguments: (1) similarity between autistic symptomatology and abnormal behaviors induced in young animals by injections of exogenous opioids, such as increasing social aloofness and decreasing social vocalization; (2) direct biochemical evidence of abnormalities of periphe

Metabolic disorders of cattle.

Goldberger discovered human pellagra was a non-infectious disease, affecting mostly the small and the timid in overcrowded institutions. Symptoms were diarrhoea, dermatitis and dementia. The staff and older children escaped the disease. They ate the meat and left the small and timid with the gravy. The 'Goldberger syndrome' is observed during competitive feeding of livestock, in ketotic animals and in the zinc depleted which are lethargic and pick all day at their feed. The pellagra preventative factor was later found to be nicotinic acid, derived from the amino acid tryptophan. Deficiencies o

Alpha adrenergic and mu-2 opioid receptors are involved in morphine-induced suppression of splenocyte natural killer activity.

The immunosuppressive effects following acute morphine administration have been mapped to opioid receptors in the central nervous system, specifically to the periaqueductal gray matter of the mesencephalon. The mesencephalon is associated with sympathetic neuronal processes, and the spleen is innervated with sympathetic neurons that are in direct apposition with lymphocytes in the periarteriolar lymphatic sheath. Accordingly, we investigated adrenergic involvement following morphine administration on natural killer (NK) activity by splenic lymphocytes. Acute morphine administration (25 mg/kg s

Bilateral injections of a selective mu-receptor agonist (morphiceptin) into the medial preoptic nucleus produces a marked delay in the initiation of sexual behavior in the male rat.

We examined the putative functional role of the medial preoptic nucleus mu-receptor population in the expression of male copulatory behavior in sexually vigorous Long-Evans rats. In the first experiment, three doses of morphiceptin (10, 500, and 1000 ng) a selective mu-receptor agonist injected bilaterally into the medial preoptic nucleus, produced a marked delay in the initiation of male copulatory behavior compared to saline injected controls. These injections significantly lengthened intromission and mount latencies while having no appreciable effect on any other parameter of male copulator
