Comment on "Hyperglycemia-triggered STAT3 activation drives thromboinflammation via NETs formation to promote hemorrhagic transformation after stroke".

This correspondence provides a critical perspective on the recent study by Zhao et al. defining the hyperglycemia-STAT3-neutrophil extracellular trap-thromboinflammation axis in post-ischemic stroke hemorrhagic transformation. Beyond three core unresolved scientific gaps concerning cell-type specificity, tissue plasminogen activator-associated pathological context, and glycemic model heterogeneity, we extend the discussion to supplementary translational constraints, the upstream regulatory role of mitochondrial oxidative stress, and implications for clinical trial design. These viewpoints aim

Neutrophils and monocytes in the pathogenesis of antiphospholipid syndrome.

Antiphospholipid syndrome (APS) is an autoimmune thromboinflammatory disorder in which antiphospholipid antibodies cause vascular thrombosis and obstetric morbidity. Growing evidence indicates that neutrophils and monocytes are key effector cells bridging inflammation and coagulation in APS. Neutrophils display heightened activation, enhanced glycolysis, and exaggerated NETosis, with downstream neutrophil extracellular traps promoting endothelial injury, complement activation, and trophoblast dysfunction. Dysregulated purinergic signaling in APS neutrophils further amplifies thromboinflammator…

MorphoMapping identifies redox-dependent control of NETotic states in primed neutrophils.

Neutrophils rapidly deploy phagocytosis, degranulation, and neutrophil extracellular trap (NET) formation to control infections, yet exaggerated NET formation contributes to tissue injury in inflammatory disease. Because NETosis is tightly linked to the cellular redox environment, we developed MorphoMapping, an imaging flow cytometry-based pipeline that resolves neutrophil morphotypes at single-cell resolution using purely morphology-derived features. Applying MorphoMapping to cytokine-primed primary human neutrophils exposed to the heme-targeted redox modulator WF10 (tetrachlorodecaoxide) rev

The heterogeneous roles of neutrophils in gastric cancer: scaffold or target?

Gastric cancer (GC) is a significant challenge for global health. Neutrophils, the predominant white blood cells in the innate immune system, are increasingly becoming known as potential contributors to either tumor-promoting or tumor-suppressive activities within different tumor biology settings. This review highlights such dual roles of neutrophils in GC, where complex interactions occur within the tumor microenvironment. Specifically, we focus on the formation and function of neutrophil extracellular traps (NETs), which have emerged as critical players in GC progression. NETs influence key …

Antioxidants as Modulators of NETosis: Mechanisms, Evidence, and Therapeutic Potential.

Neutrophil extracellular trap (NET) formation is a process known as NETosis and is a critical innate immune response mechanism that can become pathologically dysregulated in various inflammatory, autoimmune, infectious, and neoplastic diseases. Reactive oxygen species (ROS) play a central role in NETosis induction, making antioxidants a promising therapeutic approach. This review outlines the molecular mechanisms underlying NET formation and highlights three principal antioxidant-based inhibitory strategies: NADPH oxidase (NOX) inhibition, ROS scavenging, and myeloperoxidase (MPO) inhibition.

Animal toxins and NETosis: A new frontier in innate immune modulation.

Neutrophil extracellular traps are molecular structures that play a dual role in host defense and pathology. While NETs effectively capture and neutralize pathogens, excessive or dysregulated NET formation contributes to autoimmune and inflammatory disorders. Animal venoms contain bioactive molecules that can modulate neutrophil functions, including NETosis. They are capable of inducing, inhibiting, or degrading NETs through different mechanisms, such as signaling pathway modulation, or direct DNA hydrolysis. Such effects not only shape the course of envenomation but also reveal novel pathways…

The small molecule inhibitor 3PO is a modulator of neutrophil metabolism, ROS production, and NET release.

Neutrophils are key effector leukocytes of the innate immune system and play a pivotal role in defending the host against microbial infections. Recent studies have identified a crucial link between glycolysis and neutrophil cellular functions. Using human neutrophils, we have investigated the intricate relationship between glycolysis, extracellular glucose availability, and the enzyme 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3 (PFKFB3), in the regulation of reactive oxygen species (ROS) and neutrophil extracellular trap (NET) production. We have identified that PFKFB3 is elevated i

Neutrophils display antibacterial defense via non-canonical LC3 decoration of extracellular bacteria.

Neutrophils play a pivotal role in the innate immune response to bacterial infection, being one of the first immune cells to reach infectious sites. Bacterial infection may induce neutrophil degranulation, production of neutrophil extracellular traps (NETs), or pathogen phagocytosis. While LC3 is typically linked to autophagy, here we observed a non-canonical role of LC3 when peripheral neutrophils interact with bacteria both in vivo and in vitro, using Shigella spp. as a model. Upon incubation with neutrophils, extracellular bacteria became labelled by LC3 (LC3+) along with granules-localised…

[Research progress in dual immune regulation of neutrophils in periodontal inflammation].

Periodontitis is a chronic inflammatory condition of the periodontal tissues triggered by bacterial biofilm, leading to manifestations such as gingival bleeding, tooth mobility, and eventual exfoliation. Neutrophils exhibit a dual role throughout the course of periodontitis, both in defense against pathogens and in potentially detrimental effects on periodontal tissues. This article elucidates the intricate mechanisms underlying the dual functions of neutrophils in periodontitis, including respiratory burst, neutrophil extracellular traps (NETs) formation, degranulation, and phagocytosis. Methyl Syringate: A Primary Driving Factor in Manuka Honeys Ability to Ameliorate Neutrophil Intracellular ROS Activity and NETosis.

BACKGROUND: Neutrophils use both the production of reactive oxygen species (ROS) and a specialized process called NETosis to defend the body from material deemed foreign. While these neutrophil behaviors are critical in preventing infection, a dysregulated response can lead to tissue damage and fibrosis at host-biomaterial interfaces. It was hypothesized that applying the flavonoids found in Manuka honey: chrysin, pinocembrin, and pinobanksin, and the phenolic compound methyl syringate to neutrophils exhibiting pro-inflammatory behavior will reduce ROS activity and prevent NETosis in primary h

A Narrative Review: The Role of NETs in Acute Respiratory Distress Syndrome/Acute Lung Injury.

Nowadays, acute respiratory distress syndrome (ARDS) still has a high mortality rate, and the alleviation and treatment of ARDS remains a major research focus. There are various causes of ARDS, among which pneumonia and non-pulmonary sepsis are the most common. Trauma and blood transfusion can also cause ARDS. In ARDS, the aggregation and infiltration of neutrophils in the lungs have a great influence on the development of the disease. Neutrophils regulate inflammatory responses through various pathways, and the release of neutrophils through neutrophil extracellular traps (NETs) is considered…

The effect of local anaesthetics on apoptosis and NETosis of human neutrophils in vitro: comparison between lidocaine and ropivacaine.

There are immunological consequences to the method by which neutrophils undergo cell death. Neutrophil apoptosis, called silent death, leads to the resolution of inflammation, while NETosis deepens and prolongs the inflammatory response and is associated with a worse prognosis of severe infections, e.g., sepsis. Besides nociceptive inhibition, local anaesthetics modulate leukocyte functions, even at low, clinically relevant concentrations. There is currently no data on ropivacaine NETosis, and this study aimed to evaluate the impact of clinical concentrations of ropivacaine (0.0007, 0.007 and

Casting a wide NET: an update on uncontrolled NETosis in response to COVID-19 infection.

Dysregulation of neutrophil extracellular trap (NET) formation has been shown to mediate disease pathology in multiple viral infections, including SARS-CoV-2. At the beginning of COVID-19 pandemic, Thierry and Roch wrote a perspective on the mechanisms by which severe SARS-CoV-2 infection may lead to uncontrolled NET formation that leads to acute respiratory distress syndrome (ARDS), systemic vascular permeability, and end organ damage. In this commentary, the progress that has been made in regards to the ideas postulated by the perspective will be discussed, with a focus on the therapeutics t

Neutrophil Interactions Stimulate Evasive Hyphal Branching by Aspergillus fumigatus.

Invasive aspergillosis (IA), primarily caused by Aspergillus fumigatus, is an opportunistic fungal infection predominantly affecting immunocompromised and neutropenic patients that is difficult to treat and results in high mortality. Investigations of neutrophil-hypha interaction in vitro and in animal models of IA are limited by lack of temporal and spatial control over interactions. This study presents a new approach for studying neutrophil-hypha interaction at single cell resolution over time, which revealed an evasive fungal behavior triggered by interaction with neutrophils: Interacting h
