Alpha-toxin is the unique lethal virulent factor produced by Clostridium septicum, which causes traumatic or non-traumatic gas gangrene and necrotizing enterocolitis in humans. Here, we analyzed channel formation of the recombinant septicum alpha-toxin and characterized its activity on living cells. Recombinant septicum alpha-toxin induces the formation of ion-permeable channels with a single-channel conductance of about 175pS in 0.1M KCl in lipid bilayer membranes, which is typical for a large diffusion pore. Septicum alpha-toxin channels remained mostly in the open configuration, displayed n

BACKGROUND: Serotonin (5-HT) and its receptors are present in central, the brain stem, and peripheral, the carotid body, tissues controlling the ventilatory responses to hypoxia. The exact action of serotonin and its nature are, however, unsettled. We hypothesized that the discrepant results on the ventilatory action of serotonin could be caused by the inability of serotonin to penetrate into the brain or the plasma membrane lipid bilayers, the target site of signal transduction cascades, after its exogenous administration.
OBJECTIVE: To study the penetrability of novel lipid derivatives of s…

Biological membranes are considered as laterally heterogeneous, containing disordered and ordered regions ranging in size from a few nanometers to micrometers. Pulsed dipolar EPR spectroscopy (PDS) is specifically designed to determine distances between spin-labeled molecules in the range from 1.5 to typically 8 nm. A non-steroidal anti-inflammatory drug ibuprofen, due to its amphiphilic nature, is integrated into the cell membrane. In this study, we investigated spin-labeled ibuprofen (ibuprofen-SL) in a lipid raft-mimicking bilayer formed by a mixture of dioleoyl-glycero-phosphocholine (DOPC

Many transmembrane (TM) proteins contain an amphipathic helix. These amphipathic helices can sense lipid composition, stabilize membrane curvature, and mediate protein-protein interactions. Here, we combined solid-state NMR spectroscopy (ssNMR) and molecular dynamics simulations to characterize the structure and membrane interactions of a minimal TM and amphipathic construct (residues 101-146) from Mycobacterium tuberculosis (Mtb) FtsL. Amino-acid-specific 15N-labeled oriented-sample ssNMR spectra in POPC/POPG (4:1 molar ratio) membranes uniquely defined the orientation of the TM helix (residu

Dengue virus (DENV) and Zika virus (ZIKV) capsid proteins efficiently recruit and surround the viral RNA at the endoplasmic reticulum (ER) membrane to yield nascent viral particles. However, little is known either about the molecular mechanisms by which multiple copies of capsid proteins assemble into nucleocapsids (NCs) or how the NC is recruited and wrapped by the ER membrane during particle morphogenesis. Here, we measured relevant interactions concerning this viral process using purified DENV and ZIKV capsid proteins, membranes mimicking the ER lipid composition, and nucleic acids in in vi

In red blood cell (RBC) diseases, the spleen contributes to anemia by clearing the damaged RBCs, but its unique ability to mechanically challenge RBCs also poses the risk of inducing other pathogenic effects. We have analyzed RBCs in hereditary spherocytosis (HS) and hereditary elliptocytosis (HE), two typical examples of blood disorders that result in membrane protein defects in RBCs. We use a two-component protein-scale RBC model to simulate the traversal of the interendothelial slit (IES) in the human spleen, a stringent biomechanical challenge on healthy and diseased RBCs that cannot be di

Many nonenveloped viruses engage host receptors that initiate capsid conformational changes necessary for genome release. Structural studies on the mechanisms of picornavirus entry have relied on in vitro approaches of virus incubated at high temperatures or with excess receptor molecules to trigger the entry intermediate or A-particle. We have induced the coxsackievirus B3 entry intermediate by triggering the virus with full-length receptors embedded in lipid bilayer nanodiscs. These asymmetrically formed A-particles were reconstructed using cryo-electron microscopy and a direct electron dete

We engineered covalently circularized nanodiscs (cNDs) which, compared with standard nanodiscs, exhibit enhanced stability, defined diameter sizes and tunable shapes. Reconstitution into cNDs enhanced the quality of nuclear magnetic resonance spectra for both VDAC-1, a β-barrel membrane protein, and the G-protein-coupled receptor NTR1, an α-helical membrane protein. In addition, we used cNDs to visualize how simple, nonenveloped viruses translocate their genomes across membranes to initiate infection.

Because of the perpetual development of resistance to current therapies for malaria, the Medicines for Malaria Venture developed the Malaria Box to facilitate the drug development process. We tested the 80 most potent compounds from the box for bilayer-mediated effects on membrane protein conformational changes (a measure of likely toxicity) in a gramicidin-based stopped flow fluorescence assay. Among the Malaria Box compounds tested, four compounds altered membrane properties (p< 0.05); MMV007384 stood out as a potent bilayer-perturbing compound that is toxic in many cell-based assays, sugges

Interactions of the amyloid β-protein (Aβ) with neuronal cell membranes, leading to the disruption of membrane integrity, are considered to play a key role in the development of Alzheimer's disease. Natural mutations in Aβ42, such as the Arctic mutation (E22G) have been shown to increase Aβ42 aggregation and neurotoxicity, leading to the early-onset of Alzheimer's disease. A correlation between the propensity of Aβ42 to form protofibrils and its effect on neuronal dysfunction and degeneration has been established. Using rational mutagenesis of the Aβ42 peptide it was further revealed that the

Optical stimulation has enabled important advances in the study of brain function and other biological processes, and holds promise for medical applications ranging from hearing restoration to cardiac pace making. In particular, pulsed laser stimulation using infrared wavelengths >1.5 μm has therapeutic potential based on its ability to directly stimulate nerves and muscles without any genetic or chemical pre-treatment. However, the mechanism of infrared stimulation has been a mystery, hindering its path to the clinic. Here we show that infrared light excites cells through a novel, highly gene

'In dopaminergic neurons, alpha-synuclein (alphaS) partitions between a disordered cytosolic state and a lipid-bound state. Binding of alphaS to membrane phospholipids is implicated in its functional role in synaptic regulation, but also impacts fibril formation associated with Parkinson's disease. We describe here a solution NMR study in which alphaS is added to small unilamellar vesicles of a composition mimicking synaptic vesicles; the results provide evidence for multiple distinct phospholipid-binding modes of alphaS. Exchange between the free state and the lipid-bound alphaS state, and be

Econazole, one of imidazole antifungals, has been reported to exhibit an inhibitory action on Mycobacterium tuberculosis and its multidrug-resistant strains under in vitro and ex vivo conditions. There is a chemotherapeutic potential of econazole against tuberculosis. We have revealed that Zn(2+) at micromolar concentrations potentiates the cytotoxicity of imidazole antifungals by increasing membrane Zn(2+) permeability. It is reminiscent of a possibility that econazole exhibits harmful action on human in the presence of Zn(2+) at a physiological range when the agent is systemically administer

Aurein 2.5 (GLFDIVKKVVGAFGSL-NH(2)) is an amphibian antimicrobial peptide. Here, characterisation studies showed the peptide to exhibit molecular areas at an air / water interface (1.77 - 3.1 nm(2)), which are in agreement with the adoption of alpha-helical structures. Lipid monolayer studies showed aurein 2.5 to induce maximal surface pressure changes of circa 7 mN m(-1) in monolayers formed from phosphatidylglycerol (PG) and circa 6 mN m(-1) in those formed from phosphatidylethanolamine (PE). These data indicate that the membrane interactions of the peptide are amphiphilicity driven with no

The spectroscopic and transmission electron microscopy (TEM) studies of interaction between chlorpromazine (CPZ) and dimyristoyl phosphatidylglycerol (DMPG) bilayer by using gold nanoparticles (AuNPs) as probes are reported. The DMPG bilayer-protected AuNPs were prepared by a simple one-step method. The DMPG bilayer tethered on the AuNPs was considered as a biomembrane model. The addition of CPZ affected the surface plasmon resonance (SPR) and morphology of the prepared AuNPs, and this effect was monitored by UV-vis spectroscopy and TEM. The interaction between CPZ and DMPG bialyer was CPZ con

The aim of the present study is to examine the effect of phospholipids on the lipid destabilization induced by chlorpromazine (CPZ) and imipramine (IP) at different levels of pH. The large unilamellar vesicles (LUV) are formed in the presence of calcein (60 mM). The vesicles containing calcein have been incubated in the presence of CPZ and IP at pH 4.5 and pH 8. At pH 4.5 CPZ and IP induce a rapid release of the calcein encapsulated in the liposomes. Calcein release, at equal concentrations of pharmacological agent, is more important by CPZ than by IP. At pH 8, the calcein release was more imp

Antibiotics were used as tools for probing the electrical properties of tight epithelia (i.e., epithelia with high electrical resistance). The electrical effects of the polyene antibiotic nystatin were assessed in two mammalian epithelia: the rabbit descending colon and the rabbit urinary bladder. In both tissues, luminal application of the drug increased transepithelial potential and conductance and virtually eliminated apical membrane resistance by increasing permeabilities to Na+, K+, and Cl-. Application of another antibiotic, gramicidin D, produced similar effects on the urinary bladder e

Nitric oxide plays a critical role in regulating vascular tone, but excessive nitric oxide release during septic shock results in hypotension due to excessive vasodilation and the formation of toxic free radicals. VBI-S is a phospholipid nanoparticle based fluid composed of lipid bilayers formed primarily by phosphatidylcholine and micelles of soybean oil encapsulated by a monolayer of phosphatidylcholine. These nanoparticles offer a novel solution by absorbing and redistributing nitric oxide and nitrite, potentially mitigating the harmful effects of excessive nitric oxide in sepsis. This pape…
